Zbtb18 Deficiency Impairs the Hippocampal Development and Neurite Growth.

Previous studies of the transcription factor Zbtb18 have primarily focused on its role in cortical development, whereas its function during hippocampal development remains poorly understood. In this study, we investigated the role of Zbtb18 in mouse hippocampal development using histological, transcriptomic, and functional approaches. Immunostaining demonstrated that Zbtb18 was partially overlapped with the neural progenitor marker Pax6, intermediate progenitor marker Tbr2, postmitotic neuronal marker Tbr1, and neuronal marker NeuN in the hippocampal primordium from embryonic day 13.5. Loss of

Slit2 inactivates GSK3β to signal neurite outgrowth inhibition.

Slit molecules comprise one of the four canonical families of axon guidance cues that steer the growth cone in the developing nervous system. Apart from their role in axon pathfinding, emerging lines of evidence suggest that a wide range of cellular processes are regulated by Slit, ranging from branch formation and fasciculation during neurite outgrowth to tumor progression and to angiogenesis. However, the molecular and cellular mechanisms downstream of Slit remain largely unknown, in part, because of a lack of a readily manipulatable system that produces easily identifiable traits in respons

LIN7-IRSp53: A novel pathway for filopodia and neurite formation?

Filopodia are dynamic, actin-rich finger-like structures that protrude from the cell membrane and play important roles in cell migration and neurite initiation and outgrowth. The insulin receptor substrate protein of 53 kDa (IRSp53) and the mammalian Diaphanous members of the formin family of proteins (mDia) are two key players in the formation of filopodia and neurites. IRSp53 is an adaptor protein that acts at the membrane:actin interface, coupling membrane deformation with F-actin polymerization. mDia formin proteins, instead, can nucleate and polymerize linear actin filaments. Emerging gen…

Oligomerization of ZFYVE27 (Protrudin) is necessary to promote neurite extension.

ZFYVE27 (Protrudin) was originally identified as an interacting partner of spastin, which is most frequently mutated in hereditary spastic paraplegia. ZFYVE27 is a novel member of FYVE family, which is implicated in the formation of neurite extensions by promoting directional membrane trafficking in neurons. Now, through a yeast two-hybrid screen, we have identified that ZFYVE27 interacts with itself and the core interaction region resides within the third hydrophobic region (HR3) of the protein. We confirmed the ZFYVE27's self-interaction in the mammalian cells by co-immunoprecipitation and c

Beta-Bungarotoxin induction of neurite outgrowth in NB41A3 cells.

In this study, different concentrations of beta-Bgt were used to treat cultured NB41A3 cells. Inverted phase contrast microscopy was then used 24h after treatment to observe the outgrowth of neurite. We found a clear outgrowth of neurite at beta-Bgt concentrations of 357 nM. However, using a cytotoxicity assay to study apoptosis, we found no significant difference in the rate of cell death in cell cultures treated with either 357 nM or 714 nM. Western blotting showed that after treatment with beta-Bgt, there was a notable decrease in small G protein Cdc42 and a marked increase in RhoA protein.

Presbycusic neuritic degeneration within the osseous spiral lamina.

OBJECTIVE: To describe a neglected anatomic variant occurring with presbycusis. STUDY DESIGN: Retrospective temporal bone histopathology study. METHODS: Quantitative analysis of peripheral hair cells, neurites, neurons, and the stria vascularis in temporal bones from individuals who had presbycusis. Fifty-three patients aged 65 years or older and with a down-sloping audiogram and clinical diagnosis of presbycusis were reviewed. Nine cases had normal hair and ganglion cell populations but reduced peripheral processes (neuritic presbycusis). These were compared with five normal-hearing control…

Gelsolin overexpression enhances neurite outgrowth in PC12 cells.

The rational design of therapies for treating nerve injuries requires an understanding of the mechanisms underlying neurite extension. Neurite motility is driven by actin polymerization; however, the mechanisms are not clearly understood. One actin accessory protein, gelsolin, is involved with remodeling the cytoskeleton, although its role in cell motility is unclear. We report a two-fold upregulation of gelsolin upon differentiation with nerve growth factor. Cells that were genetically modified to overexpress gelsolin have longer neurites and a greater neurite motility rate compared to contro…

Quantitative reconstruction of neuronal mitochondrial network in neurites and somata in rat hippocampus and prefrontal cortex.

INTRODUCTION: Mitochondrial networks exhibit striking heterogeneity in their morphology and distribution across different neuronal compartments, reflecting the diverse metabolic demands of these structures. METHODS: In this study, we used automated tape-collecting ultramicrotome scanning electron microscopy (ATUM-SEM) to reconstruct and quantify mitochondrial networks in the somata and neurites of neurons in the rat prefrontal cortex (PFC) and hippocampus (HPC; CA1 stratum radiatum). We developed an automated segmentation pipeline based on an attention-enhanced 3D U-Net to extract all mitocho…

Cyclosporin A as an Alternative Neuroimmune Strategy to Control Neurites and Recover Neuronal Tissues in Leprosy.

Leprosy, also known as Hansen's disease, continues to have a substantial impact on infectious diseases throughout the world. Leprosy is a chronic granulomatous infection caused by Mycobacterium leprae and shows a wide clinical and immunopathological spectrum related to the immune response of the host. This disease affects the skin and other internal organs with a predilection to infect Schwann cells, which play an active role during axonal degeneration, affecting peripheral nerves and promoting neurological damage. This chronic inflammation influences immune function, leading to neuroimmune di

Neuronal myosin-X is upregulated after peripheral nerve injury and mediates laminin-induced growth of neurites.

The successful outcome of peripheral neuronal regeneration is attributed both to the growth permissive milieu and the intrinsic ability of t … cones and that silencing of myosin-X using RNA interference impairs the integrin-mediated growth of neurites on laminin, but has no effect on non-integrin mediated growth on N-cadherin.

The small GTPase Rac is involved in clustering of hippocampal neurons and fasciculation of their neurites.

In hippocampal neurons cultured from brains of newborn rats, the glutamate receptor agonist N-methyl-D-aspartate induced the clustering of neuronal perikarya and the fasciculation of neurites. In addition, N-methyl-D-aspartate activated the small GTPase Rac1. Other stimuli of Rac activity, such as the Rho kinase inhibitors Y-27632, H-1152, and H89, as well as the cytotoxic necrotizing factor-1 from Escherichia coli, also caused neuronal clustering and neurite bundling. In neurons transiently transfected with dominant negative Rac1N17 neither N-methyl-D-aspartate nor Y-27632 induced clustering …

Purified extracts from short-time-predegenerated rats' sciatic nerves promote the regrowth of injured hippocampal neurites.

Our previous studies revealed that purified extracts (submicrosomal fractions) obtained from peripheral nerves predegenerated for 7-, 28-, and 35-days facilitated neurite outgrowth from the injured hippocampus. It is recently known that totally transected peripheral nerve exhibits biphasic neurite-promoting activity. The early phase lasts 7 days. The aim of the present study was to find whether extracts obtained from short-time predegenerated (1-6 days) peripheral nerves exert any neurotrophic effect and when this influence is maximal. Experiments were carried out on adult male Wistar rats. Sc

Parvalbumin-immunoreactive dystrophic neurites and aberrant sprouts in the cerebral cortex of patients with Alzheimer's disease.

Parvalbumin-immunoreactive dystrophic neurites and aberrant terminal sprouts associated with senile plaques, together with preserved density of parvalbumin-immunoreactive neurons, were found in the cerebral cortex of two patients with Alzheimer's disease, one of them familiar, in which a biopsy of the left frontal lobe was carried out for diagnostic and counselling purposes. These findings suggest that, although parvalbumin-immunoreactive cells are relatively resistant to degeneration in Alzheimer's disease, parvalbumin-immunoreactive neuronal processes can degenerate in some cases of Alzheime…

Initiation and growth of ectopic neurites and meganeurites during postnatal cortical development in ganglioside storage disease.

The incidence of cortical pyramidal neurons displaying meganeurites or enlarged axon hillocks with ectopic spines and neurites was evaluated developmentally using feline models of GM1 and GM2 gangliosidosis. Results of these studies demonstrated that the onset of ectopic neurite growth occurred after the elaboration of dendrites on cortical pyramidal neurons, and that the time of onset of this renewed dendritogenesis was similar in the two diseases. Initiation and growth of ectopic neurites also correlated in a general way with onset and progression of clinical deterioration in both diseases. …

Neuroaxonal dystrophy in experimental Creutzfeldt-Jakob disease: electron microscopical and immunohistochemical demonstration of neurofilament accumulations within affected neurites.

Neuroaxonal dystrophy is a feature of neuronal degeneration encountered in all subacute spongiform "virus" encephalopathies, including scrapie and Creutzfeldt-Jakob disease (CJD). By immunohistochemical techniques, the accumulation of 200 kDa neurofilament protein was demonstrated in affected neurites in murine CJD. These neurites exhibited the ultrastructural features of dystrophic neurites encountered in other neurodegenerative disorders, particularly Alzheimer's disease. These findings support the hypothesis that impairment of slow axoplasmic transport is a common pathogenetic mechanism for…
