Stimulatory effects of endogenous and exogenous nitric oxide on gastric acid secretion in anesthetized rats.

We previously reported the stimulatory effect of endogenous nitric oxide (NO) on gastric acid secretion in the isolated mouse whole stomach and histamine release from gastric histamine-containing cells. In the present study, we investigated the effects of endogenous and exogenous NO on gastric acid secretion in urethane-anesthetized rats. Acid secretion was studied in gastric-cannulated rats stimulated with several secretagogues under urethane anesthesia. The acid secretory response to the muscarinic receptor agonist bethanechol (2 mg/kg, s.c.), the cholecystokinin(2) receptor agonist pentagas

Fish oil supplementation and essential fatty acid deficiency reduce nitric oxide synthesis by rat macrophages.

Both fish oil-derived omega-3 polyunsaturated fatty acid (omega 3 PUFA) supplementation and essential fatty acid (EFA) deficiency have been shown to exert anti-inflammatory effects and, hence, to ameliorate immune-mediated glomerulonephritis. The mechanisms underlying these effects include alterations in the production of eicosanoids, cytokines (that is, tumor necrosis factor, TNF-alpha) and reactive oxygen species by blood borne cells. Because, in addition to these mediators nitric oxide (NO) is also implicated in glomerular injury, we have examined if both diets affected macrophage NO produc

Comparison of in vitro effects of triflusal and acetysalicylic acid on nitric oxide synthesis by human neutrophils.

Recent studies have suggested that the protective anti-ischemic effects of acetylsalicylic acid are stronger than the inhibition of platelet thromboxane A2 synthesis. Since ischemic events still occur in acetylsalicylic acid-treated patients, the development of new drugs with more powerful protective effects is needed. We compared the effects of a new platelet antiaggregating drug, 2-acetoxy-4-trifluoromethyl-benzoic acid (triflusal) and of acetylsalicylic acid on the interaction between human neutrophils and platelets, examining the capability of neutrophils to generate nitric oxide (NO). Computational Foretelling and Experimental Implementation of the Performance of Polyacrylic Acid and Polyacrylamide Polymers as Eco-Friendly Corrosion Inhibitors for Copper in Nitric Acid.

Copper is primarily used in many industrial processes, but like many other metals, it suffers from corrosion damage. Polymers are not only one of the effective corrosion inhibitors but also are environmentally friendly agents in doing so. Hence, in this paper, the efficacy of two polyelectrolyte polymers, namely poly(acrylic acid) (PAA) and polyacrylamide (PAM), as corrosion inhibitors for copper in molar nitric acid medium was explored. Chemical, electrochemical, and microscopic tools were employed in this investigation. The weight-loss study revealed that the computed inhibition efficiencies

Retinoic acid-mediated enhancement of the cholinergic/neuronal nitric oxide synthase phenotype of the medial septal SN56 clone: establishment of a nitric oxide-sensitive proapoptotic state.

It is unclear what mechanisms lead to the degeneration of basal forebrain cholinergic neurons in Alzheimer's or other human brain diseases. Some brain cholinergic neurons express neuronal nitric oxide (NO) synthase (nNOS), which produces a free radical that has been implicated in some forms of neurodegeneration. We investigated nNOS expression and NO toxicity in SN56 cells, a clonal cholinergic model derived from the medial septum of the mouse basal forebrain. We show here that, in addition to expressing choline acetyltransferase (ChAT), SN56 cells express nNOS. Treatment of SN56 cells with re

Carnosic acid ameliorates methotrexate-induced male infertility: targeting testicular redox imbalance and nitric oxide/CGMP signaling.

PURPOSE: Methotrexate (MTX), a folate antagonist used in the treatment of cancer and autoimmune disorders, unfortunately, induces testicular toxicity and impairs male fertility. Although the tissue-protective properties of Carnosic acid (CAR) are known, its effectiveness in mitigating MTX-induced reproductive toxicity has not been adequately studied. This study explores the protective role of CAR against the toxicity of MTX in rat testes. METHODS: Forty male Wistar rats were divided into four groups: control, MTX (20 mg/kg i.p., day 7), MTX + low-dose CAR (20 mg/kg p.o., 14 days), and MTX + h

Nitric oxide synthesis stimulated by arachidonic acid accumulation via PPARα in acetylcholine-stimulated gastric mucous cells.

NEW FINDINGS: What is the central question of this study? Arachidonic acid (AA) stimulates NO production in antral mucous cells without any increase in [Ca2+ ]i . Given that the intracellular AA concentration is too low to measure, the relationship between AA accumulation and NO production remains uncertain. Is AA accumulation a key step for NO production? What is the main finding and its importance? We demonstrated that AA accumulation is a key step for NO production. The amount of AA released could be measured using fluorescence-HPLC. The intracellular AA concentration was maintained at < 1

Cesium removal from a water system using a polysulfone carrier containing nitric acid-treated bamboo charcoal.

Laboratory scale sorption and desorption experiments were performed to investigate the cesium (Cs) removal efficiency of a bead-shaped polysulfone carrier containing HNO3-treated bamboo charcoal (BC). The average Cs removal efficiency of BC only and of polysulfone carrier without BC after 1 h sorption reaction was 53 and 18%, respectively. However, the Cs removal efficiency for the polysulfone carrier with 5% HNO3-treated BC (P-5N-BC) after 1 h and 24 h reaction was 66 and 98%, respectively. The Cs removal efficiency after 24 h reaction remained >85% over a wide range of pH and temperature con

Smoke-Isolated Karrikins Stimulated Tanshinones Biosynthesis in Salvia miltiorrhiza through Endogenous Nitric Oxide and Jasmonic Acid.

Although smoke-isolated karrikins (KAR₁) could regulate secondary metabolism in medicinal plants, the signal transduction mechanism has not been reported. This study highlights the influence of KAR₁ on tanshinone I (T-I) production in Salvia miltiorrhiza and the involved signal molecules. Results showed KAR₁-induced generation of nitric oxide (NO), jasmonic acid (JA) and T-I in S. miltiorrhiza hairy root. KAR₁-induced increase of T-I was suppressed by NO-specific scavenger (cPTIO) and NOS inhibitors (PBITU); JA synthesis inhibitor (SHAM) and JA synthesis inhibitor (PrGall), which indicated tha

Endometritis impairs luteal development, function, and nitric oxide and ascorbic acid concentrations in buffalo (Bubalus bubalis).

A vast majority of the world buffalo resource is concentrated in tropical and subtropical countries. Apart from heat stress and poor nutritional availability, endometritis is one of the most commonly encountered reproductive problems limiting fertility and consequently productive potential of the species. As demonstrated recently, endometritis impairs growth and follicular fluid composition of the largest follicle in buffalo. In the present study, the effect of endometritis on luteal development, function, nitric oxide (NO), and ascorbic acid was investigated. Reproductive tracts were collecte

Role of uncoupled endothelial nitric oxide synthase in abdominal aortic aneurysm formation: treatment with folic acid.

It has been shown that endothelial NO synthase (eNOS) uncoupling occurs in hypertension and atherosclerosis. However, its causal role in vascular pathogenesis has not been characterized previously. Here, we challenged eNOS preuncoupled hyperphenylalaninemia (hph)-1 mice (deficient in eNOS cofactor tetrahydrobiopterin biosynthetic enzyme GTPCHI) with angiotensin II (Ang II; 0.7 mg/kg per day, 14 days). Both wild-type and hph-1 groups developed hypertension similarly up to day 6 to 7. Thereafter, ≈14% of Ang II-infused (0.7 mg/kg per day) hph-1 mice (n=72) started to die suddenly of ruptured abd

Cyclosporine A attenuates 3-nitropropionic acid-induced Huntington-like symptoms in rats: possible nitric oxide mechanism.

Cyclosporine A is a well-known immunosuppressant drug that is currently used for prevention of allograft rejection. The current study was conducted to explore the therapeutic potential of cyclosporine A against 3-nitropropionic acid (3-NP)-induced neurotoxicity, an animal model of Huntington disease (HD). Systemic administration of 3-NP (10 mg/kg) for 14 days significantly impaired body weight, motor activity, biochemical parameters (raised lipid peroxidation, nitrite concentration, depletion of superoxide dismutase [SOD] and catalase), and mitochondrial enzymes. Cyclosporine A (2.5, 5, and 10

Abscisic acid (ABA) inhibits light-induced stomatal opening through calcium- and nitric oxide-mediated signaling pathways.

Nitric oxide (NO) is an important signaling component of ABA-induced stomatal closure. However, only fragmentary data are available about NO effect on the inhibition of stomatal opening. Here, we present results supporting that, in Vicia faba guard cells, there is a critical Ca2+-dependent NO increase required for the ABA-mediated inhibition of stomatal opening. Light-induced stomatal opening was inhibited by exogenous NO in V. faba epidermal strips. Furthermore, ABA-mediated inhibition of stomatal opening was blocked by the specific NO scavenger cPTIO, supporting the involvement of endogenous

Water, sulfur dioxide and nitric acid adsorption on calcium carbonate: a transmission and ATR-FTIR study.

Calcium carbonate (CaCO3) is a reactive component of mineral dust aerosol as well as buildings, statues and monuments. In this study, attenuated total reflection (ATR) and transmission Fourier transform infrared spectroscopy (FTIR) have been used to study the uptake of water, sulfur dioxide and nitric acid on CaCO3 particles at 296 K. Under atmospheric conditions, CaCO3 particles are terminated by a Ca(OH)(CO3H) surface layer. In the presence of water vapor between 5 and 95% relative humidity (RH), water molecularly adsorbs on the Ca(OH)(CO3H) surface resulting in the formation of an adsorbed

Chemically functionalized diglycolamide resin for rapid and efficient recovery of Am(III) from nitric acid medium.

A diglycolamide (DGA)-functionalized silica-based extraction chromatography resin was synthesized and evaluated for the separation of trivalent actinides from nitric acid medium. Batch sorption studies showed strong affinity of the resin toward Am3+, with distribution coefficients (Kd) exceeding 5000 mL/g above 3 M HNO3. Sorption equilibrium was achieved within 20 min, indicating rapid uptake kinetics suitable for separation applications. Equilibrium sorption studies were performed using Eu3+ as a non-radioactive surrogate for Am3+. The resin exhibited a maximum Eu3+ uptake capacity of 9.5±0.5

Nitric oxide donor-modified extracellular matrix hydrogel loaded with salvianolic acid B for diabetic wound healing.

Nitric oxide (NO) donor-based gas therapy has been widely explored to promote angiogenesis in diabetic wound treatment, particularly to compensate for insufficient endogenous NO levels in damaged tissues. Here, we present a skin-derived extracellular matrix (ECM)-based platform that leverages its inherent biocompatibility and cell affinity for efficient NO delivery. An NO donor was chemically grafted onto the ECM scaffold, which was subsequently processed into a hydrogel (H@ECM-SNO/Sal-B) capable of encapsulating salvianolic acid B (Sal B), a bioactive phytochemical with pro-healing potential.

Carnosic Acid Activates the STING/IRF3 Pathway to Induce Nitric Oxide-Mediated Apoptosis in Osteosarcoma Cells.

Osteosarcoma (OS) is a highly aggressive bone cancer with limited therapeutic options. Carnosic acid (CA), a phenolic diterpene with well-established antioxidant properties, has shown anticancer activity, yet its mechanisms in OS remain unclear. In this study, we found that CA suppressed proliferation and induced apoptosis in human osteosarcoma cells in a dose-dependent manner. Mechanistically, CA activated the STING/IRF3 signaling pathway and enhanced nitric oxide (NO) production, factors closely linked to redox modulation and mitochondrial apoptotic signaling. Pharmacological inhibition or s

Jasmonic acid improves cadmium tolerance in rice (Oryza sativa) by reducing the production of nitric oxide.

The involvement of jasmonic acid (JA) in the rice's response to cadmium (Cd) stress is well recognized, though the underlying mechanisms remain unclear. In this study, exposure to Cd stress rapidly elevated endogenous JA concentrations in rice roots, meanwhile, a mutant coleoptile photomorphogenesis 2 (cpm2) which produces less JA, was more sensitive to Cd stress than its wild type (WT). JA mitigated Cd toxicity by decreasing Cd absorption in root cell wall and shoot, which was achieved by up-regulating the expression of the Cd-chelation and efflux-related genes such as OsHMA3, OsABCG36 and Os

Lanthanum supplementation with abscisic acid modulates rhizosphere dynamics through changes in nitric oxide synthesis in wheat.

Lanthanum (La), a rare earth element (REE) found its wide applications in the agricultural land practices. Utilization of La as a fertilizer is based on its concentration for staple crops, including wheat (Triticum aestivum). We have investigated the role of a beneficial dose of La (60 μM) along with ABA (10 μM) in improving wheat root dynamics and defense systems (ascorbate-glutathione pathway, secondary metabolites, and nitric oxide (NO) biosynthesis). Co-application of La60 and ABA significantly enhanced all the root dynamic attributes along with defense systems and reduced production of re

Jasmonic acid and nitric oxide orchestrate a hierarchical melatonin cascade for Botrytis cinerea resistance in tomato.

Tomato gray mold, caused by Botrytis cinerea (B. cinerea), poses a major challenge to tomato production. In plants, B. cinerea resistance is positively regulated by melatonin; however, the regulatory mechanism of melatonin biosynthesis during B. cinerea infection is not known. Here, we established the working model of jasmonic acid (JA) and nitric oxide (NO) on melatonin biosynthesis in the state of B. cinerea infection. NO responded to B. cinerea infection earlier than JA. In the early stage of infection, tomato caffeic acid O-methyltransferase 2 (SlCOMT2) was S-nitrosylated by NO at Cys344,
