Oleanolic Acid and Ursolic Acid Improve Bone Properties and Calcium Balance and Modulate Vitamin D Metabolism in Aged Female Rats.

Oleanolic acid (OA) and ursolic acid (UA) are the major chemical constituents in Fructus Ligustri Lucidi (FLL), a kidney-tonifying Chinese herb that is previously shown to improve bone properties and enhance calcium balance in aged female rats. The present study was designed to study if OA and UA act as the active ingredients in FLL to exert the positive effects on bone and mineral metabolism in aged rats. Aged (13-month-old) Sprague-Dawley female rats were randomly assigned to four groups with oral administration of drug or vehicle treatment for 12 weeks: medium calcium diet (MCD, 0.6% calciu…

A Combination of Ursolic Acid and Oleanolic Acid Disrupts Bioenergetic Homeostasis in Prostate Cancer and Inhibits Cancer Growth by Cytotoxic Autophagy.

Oleanolic acid (OA) and ursolic acid (UA) are bioactive pentacyclic triterpenoids (PTTs) and potential therapeutics for cancers. If tested individually, their therapeutic efficacy is limited. The antitumor potential of OA, UA, and their combination was tested against castration-resistant prostate cancer cells (CRPCs) in vitro and in vivo. UA, OA, and UA + OA (1:1) inhibited the cell proliferation and clonogenic growth of CRPC cells. Compared to the potency of individual OA and UA on these cells, their potency was synergistically enhanced when tested as an OA + UA mix. Neither of these compound…

Oleanolic acid prevents ferroptosis and enhances oocyte competence during in vitro maturation in a porcine model.

The in vitro production of porcine embryos is a valuable model for livestock biotechnology and human reproductive research. However, oocyte quality is often compromised during culture by ferroptosis, which is an iron-dependent form of cell death driven by lipid peroxidation and redox imbalance. This study induced ferroptosis using RSL3 during porcine oocyte maturation to evaluate the protective potential of oleanolic acid (OA), an antioxidant triterpenoid compound. Exposure to RSL3 increased Fe2+ accumulation (as detected by FerroOrange staining), lipid peroxidation (as determined by the MDA a…

Oleanolic acid exerts bone anabolic effects via activation of osteoblastic 25-hydroxyvitamin D 1-alpha hydroxylase.

Oleanolic acid (OA) is previously shown to exert bone protective effects in aged animals. However, its role in regulating osteoblastic vitamin D bioactivation, which is one of major causes of age-related bone loss, remains unclear. Our results revealed that treatment of OA significantly increased skeletal CYP27B1 expression and circulating 1,25(OH)2D3 in ovariectomized mice (p <0.01). Moreover, OA upregulated CYP27B1 protein expression and activity, as well as the vitamin D-responsive bone markers alkaline phosphatase (ALP) activity and osteopontin (OPN) protein expression, in human osteoblast…

Oleanolic acid improved intestinal immune function by activating and potentiating bile acids receptor signaling in E. coli-challenged piglets.

BACKGROUND: Infection with pathogenic bacteria during nonantibiotic breeding is one of the main causes of animal intestinal diseases. Oleanolic acid (OA) is a pentacyclic triterpene that is ubiquitous in plants. Our previous work demonstrated the protective effect of OA on intestinal health, but the underlying molecular mechanisms remain unclear. This study investigated whether dietary supplementation with OA can prevent diarrhea and intestinal immune dysregulation caused by enterotoxigenic Escherichia coli (ETEC) in piglets. The key molecular role of bile acid receptor signaling in this proce…
