A self-filling micropipette for the isolation of single coccidial oocysts.

A disposable, self-filling micropipette is described for the isolation of single oocysts of chicken coccidia from laboratory stocks. A sterile 1 microliter capillary tube is mounted, using sealing wax, into the narrower end of a broken-off Pasteur pipette. This micropipette is pushed into one end of a 60 cm length of flexible plastic tubing and a 5 cm glass tube is pushed into the other end. The apparatus is used by holding the glass tube in one's mouth, maintaining a positive pressure in the buccal cavity, while the tip of the capillary tube in the micropipette is juxtaposed with a single ooc…

Cryptosporidium Oocyst Purification Using Discontinuous Gradient Centrifugation.

Many laboratory studies in cryptosporidial research require a source of purified oocysts. Sources can include experimentally infected laboratory animals or from samples collected from naturally infected animals and from clinical cases of human cryptosporidiosis. Purification of oocysts can be accomplished with readily available laboratory equipment including tabletop centrifuges and microcentrifuges. Following purification, oocysts can be stored in antibiotic-supplemented buffers or in 2.5% aqueous potassium dichromate for over 6 months. Ultimately, oocyst viability and infectivity decline to …

Artemia is capable of spreading oocysts of Cryptosporidium and the cysts of Giardia.

The capability for ingesting and spreading the fixed oocysts of Cryptosporidium and fixed cysts of Giardia, two waterborne protozoan parasites, by Artemia franciscana, a microcrustacean widely used as live diet in fish and shellfish larviculture, was demonstrated using differential interference contrast and immunofluorescence microscopy. Our findings suggest the possibility that this microcrustacean could serve as a disseminating vehicle of both parasites in aquatic environments.

Sources and species of cryptosporidium oocysts in the Wachusett Reservoir watershed.

Understanding the behavior of Cryptosporidium oocysts in the environment is critical to developing improved watershed management practices for protection of the public from waterborne cryptosporidiosis. Analytical methods of improved specificity and sensitivity are essential to this task. We developed a nested PCR-restriction fragment length polymorphism assay that allows detection of a single oocyst in environmental samples and differentiates the human pathogen Cryptosporidium parvum from other Cryptosporidium species. We tested our method on surface water and animal fecal samples from the Wa…

Cryptosporidium oocysts in a water supply associated with a cryptosporidiosis outbreak.

An outbreak of cryptosporidiosis occurred in and around Clitheroe, Lancashire, in northwest England, during March 2000. Fifty-eight cases of diarrhea with Cryptosporidium identified in stool specimens were reported. Cryptosporidium oocysts were identified in samples from the water treatment works as well as domestic taps. Descriptive epidemiology suggested that drinking unboiled tap water in a single water zone was the common factor linking cases. Environmental investigation suggested that contamination with animal feces was the likely source of the outbreak. This outbreak was unusual in that …

Evaluation for detection of Cryptosporidium oocysts in diarrheal feces of calves.

For the detection of Cryptosporidium oocysts, fecal samples were collected from 201 calves which showed diarrhea. Among the 201 samples, 29 samples (14.4%) were positive for Cryptosporidium spp. by the DMSO-modified acid-fast stain (MAFS), 23 samples (11.4%) were positive by commercial kit (Meridian Diagnostics, Cincinnati, Ohio) and 23 by the indirect immunofluorescence antibody (IFA) assay employing the monoclonal antibody (mAb C6). When tested by both IFA and MAFS, 20 fecal samples were positive for Cryptosporidium oocysts whereas 169 fecal samples were negative. If the MAFS is considered a…

Attempts to immunize chickens with Cryptosporidium baileyi oocyst extract.

In order to study the possibility of immunization against Cryptosporidium baileyi with extracted crude antigen, Arbor Acres chickens were injected intramuscularly with 80 micrograms of C. baileyi oocyst-derived proteins (uninfected immunized, UI) or inoculated orally with 8 x 10(5) viable C. baileyi oocysts (infected control, IC) at 1 wk of age. The immunization was repeated in the UI group at 2 wk of age. Uninfected (UC) birds served as controls. All animals in UI, IC, and UC groups were challenged orally with 8 x 10(5) C. baileyi oocysts at the age of 4 wk. Blood samples were collected when …

Mediation of immunity to Toxoplasma gondii oocyst shedding in cats.

Immunity to Toxoplasma gondii, as measured by oocyst shedding, was studied in cats. In 3 trials, 12 3-mo-old T. gondii-free cats were fed tissue cysts of the ME-49 strain of T. gondii. All cats shed T. gondii oocysts for approximately 1 wk starting 3-5 days after ingesting tissue cysts. One cat became ill because of toxoplasmic pneumonia and was killed 17 days after inoculation. The remaining cats remained clinically normal. Approximately 3 mo after primary infection, these 11 cats (immune) and 11 age-matched or littermate uninfected cats (nonimmune) were challenged orally with tissue cysts of…

Feeder insects differ in passage of coccidian oocysts in captive reptiles.

Coccidia (Apicomplexa) may have a devastating effect on captive reptiles, particularly chameleons and bearded dragons (Pogona vitticeps). This study investigated the potential of three common feeder insects - the Argentine cockroach (Blaptica dubia), the banana cricket (Gryllus assimilis) and the superworm larva (Zophobas morio) - to act as passive vectors for coccidian oocysts, specifically Isospora amphiboluri and Choleoeimeria pogonae, common enteric parasites of captive bearded dragons. Faecal samples from experimentally infected bearded dragons were fed to the insects to assess the passag…

Cryptosporidium oocysts: prevalence in dogs in Abuja, Federal Capital Territory, Nigeria.

Cryptosporidium is known to be a zoonotic protozoan parasite, located mainly intracellularly causing the disease called cryptosporidiosis, a diarrheic disease of human and animals. Risk factors for dogs’ illness includes exposure to contaminated drinking water from well, borehole and tap water. Hunting as an activity of some dogs serve as a major exposure. The objectives of this study is to determine the prevalence and risk factors associated with fecal shedding of Cryptosporidium oocysts in dogs in Abuja, Federal Capital Territory (FCT). This study was carried out from January to May 2018 in …

Comparison of current methods used to detect Cryptosporidium oocysts in stools.

In this review all of the methods that are currently in use for the investigation of Cryptosporidium in stool material are highlighted and critically discussed. It appears that more qualifications and background knowledge in this field regarding the diagnosis of the Cryptosporidium parasite is required. Furthermore, there is no standardization for the protocols that are commonly used to either detect oocysts in faeces or to diagnose the Cryptosporidium infection. It is therefore necessary to initiate further education and research that will assist in improving the accuracy of the diagnosis of …

Global iTRAQ-based proteomic profiling of Toxoplasma gondii oocysts during sporulation.

UNLABELLED: Toxoplasma gondii is a medically and economically important protozoan parasite. However, the molecular mechanisms of its sporulation remain largely unknown. Here, we applied iTRAQ coupled with 2D LC-MS/MS proteomic analysis to investigate the proteomic expression profile of T. gondii oocysts during sporulation. Of the 2095 non-redundant proteins identified, 587 were identified as differentially expressed proteins (DEPs). Based on Gene Ontology enrichment and KEGG pathway analyses the majority of these DEPs were found related to the metabolism of amino acids, carbon and energy. Prot…

Preferential transport of Cryptosporidium parvum oocysts in variably saturated subsurface environments.

When oocysts of the protozoan Cryptosporidium parvum contaminate drinking water supplies, they can cause outbreaks of Cryptosporidiosis, a common waterborne disease. Of the different pathways by which oocysts can wind up in drinking water, one has received little attention to date; that is, because soils are often considered to be perfect filters, the transport of oocysts through the subsoil to groundwater is generally ignored. To evaluate the significance of this pathway, a series of laboratory experiments investigated subsurface transport of oocysts. Experiment 1 was carried out in a vertica…

Asymptomatic shedding of Cryptosporidium oocysts by red deer hinds and calves.

Levels of Cryptosporidium infection in a group of red deer were monitored over a period of 1 year. Faecal samples were examined on an approximate monthly basis from adult hinds and calves for the presence of Cryptosporidium oocysts. The water-ether sedimentation method followed by sucrose flotation and a monoclonal antibody identification procedure were used. It was found that apparently healthy adult deer were shedding low numbers of oocysts in their faeces throughout the year and that there appeared to be a periparturient increase in the numbers of oocysts shed. Samples taken from 6-month-ol…

Immunoassay for viable Cryptosporidium parvum oocysts in turbid environmental water samples.

Cryptosporidium parvum oocysts in drinking water have been implicated in outbreaks of diarrheal disease. Current methods for monitoring environmental exposures to C. parvum only account for total number of oocysts without regard for the viability of the parasite. Measurement of oocyst viability, as indicated by an oocyst's ability to excyst, is useful because over time oocysts lose the ability to excyst and become noninfective. Thus, correlating the number of viable oocysts in drinking water with incidence and risk for disease should be more reliable than using the total number of oocysts. We …

Cryptosporidium serpentis oocysts and microsporidian spores in feces of captive snakes.

Fecal smears of 90 snakes, 29 lizards, and 8 turtles and tortoises were tested for Cryptosporidium spp. oocysts and microsporidian spores. Microsporidian spores measured mean = 3.7 microm in length and mean = 2.3 microm in width and were present in feces of 19 snakes and 1 lizard (16%); 13 of these snakes also shed Cryptosporidium serpentis oocysts. The oocysts were numerous in all positive samples, whereas microsporidian spores were always sparse, irrespective if whether fecal samples contained the oocysts. Retrospective examination of reptile clinical records revealed that all animals sheddi…

Hydrogen peroxide gas plasma sterilization is effective against Cryptosporidium parvum oocysts.

The aim of this work was to evaluate in an immunosuppressed rat cryptosporidiosis model a new method that combines vacuum and low-temperature hydrogen peroxide gas plasma for sterilization of endoscopic material contaminated by Cryptosporidium parvum. Rats were challenged with oocysts either air-dried or air-dried and treated with vacuum alone or associated with gas plasma. No rat was found infected after gas plasma exposure of oocysts, whereas vacuum or air-drying alone resulted only in a decreased infectivity.

Survey of Cryptosporidium oocysts from adult cattle in a slaughter house.

Cryptosporidian oocysts were surveyed in rectal stools of adult cattle which were carried into slaughterhouse from April 1995 to July 1996. We morphologically and histologically investigated oocysts, and experimentally infected the isolated oocysts to mice and rats. Cryptosporidian oocysts were detected from 24 of 512 cattle (4.7%). They were spherical or ovoid, and the size was 7.0-7.9 x 5.3-6.1 microm. Mice and rats inoculated orally with 10(5)-10(7) oocysts became infected and discharged oocysts in the feces for a period of more than two months. Developing parasites were detected from the s…

Murine infection model for maintenance and amplification of Cryptosporidium parvum oocysts.

Propagation of Cryptosporidium parvum is problematic because in vitro development of the parasite is poor and animals are only briefly susceptible as neonates. At present oocysts of the parasite are usually procured by passage in neonatal sheep or cattle. In the present study, large numbers of oocysts of C. parvum could be isolated following infection of dexamethasone-treated adult C57BL/6 mice. The amount of immunosuppressive drug and the regimen of administration were critical for successful maintenance of the parasite, however. Routinely, 10 mice (age, 8 to 12 weeks) were injected four time…

Efficacy of activated sludge in removing Cryptosporidium parvum oocysts from sewage.

Primary clarifier effluent (procedure B) and final effluent (procedure A) from a wastewater treatment plant were enriched with Cryptosporidium parvum oocysts obtained from the feces of naturally infected calves. Procedure B samples alone were subjected to a laboratory simulation of activated-sludge treatment. Coccidium-free neonatal CD-1 mice were then inoculated intragastrically with procedure A or procedure B samples. Seven days after inoculation, the intensity of oocyst infection in procedure B mice was 91% less than in procedure A mice (controls).
