Endocytic Activity by Apicomplexa Parasites.

Macromolecules are incorporated by eukaryotic cells through a process known as endocytosis, intensely analyzed in mammalian and yeast cells, but still lacking deep studies in pathogenic protists. Here we present what is presently known on endocytic activity carried out in some members of the Apicomplexa group (mainly concentrated in Plasmodium and Toxoplasma, but with references to Eimeria, Babesia, and Theileria). In most cells, endocytic activity takes place throughout the cell's surface. In apicomplexans, it is restricted to a special surface structure known as a micropore (also called cyto

Expansion microscopy of apicomplexan parasites.

Apicomplexan parasites comprise significant pathogens of humans, livestock and wildlife, but also represent a diverse group of eukaryotes with interesting and unique cell biology. The study of cell biology in apicomplexan parasites is complicated by their small size, and historically this has required the application of cutting-edge microscopy techniques to investigate fundamental processes like mitosis or cell division in these organisms. Recently, a technique called expansion microscopy has been developed, which rather than increasing instrument resolution like most imaging modalities, physi…

Organelle Dynamics in Apicomplexan Parasites.

Apicomplexan parasites, such as Toxoplasma gondii and Plasmodium falciparum, are the cause of many important human and animal diseases. While T. gondii tachyzoites replicate through endodyogeny, during which two daughter cells are formed within the parental cell, P. falciparum replicates through schizogony, where up to 32 parasites are formed in a single infected red blood cell and even thousands of daughter cells during mosquito- or liver-stage development. These processes require a tightly orchestrated division and distribution over the daughter parasites of one-per-cell organelles such as t…

Potent Tetrahydroquinolone Eliminates Apicomplexan Parasites.

Apicomplexan infections cause substantial morbidity and mortality, worldwide. New, improved therapies are needed. Herein, we create a next generation anti-apicomplexan lead compound, JAG21, a tetrahydroquinolone, with increased sp3-character to improve parasite selectivity. Relative to other cytochrome b inhibitors, JAG21 has improved solubility and ADMET properties, without need for pro-drug. JAG21 significantly reduces Toxoplasma gondii tachyzoites and encysted bradyzoites in vitro, and in primary and established chronic murine infections. Moreover, JAG21 treatment leads to 100% survival. Fu

Cell division in apicomplexan parasites.

Toxoplasma gondii and Plasmodium falciparum are important human pathogens. These parasites and many of their apicomplexan relatives undergo a complex developmental process in the cells of their hosts, which includes genome replication, cell division and the assembly of new invasive stages. Apicomplexan cell cycle progression is both globally and locally regulated. Global regulation is carried out throughout the cytoplasm by diffusible factors that include cell cycle-specific kinases, cyclins and transcription factors. Local regulation acts on individual nuclei and daughter cells that are devel…

Parasites and chronic renal failure.

Suppression of the human immune system results in an increase in susceptibility to infection by various infectious agents. Conditions such as AIDS, organ transplantation and chronic renal insufficiency (CRI) are the most important cause of insufficient immune response against infections. Long term renal disorders result in uremia, which can suppress human immune system. Parasitic infections are one of the most important factors indicating the public health problems of the societies. These infections can be more hostile and life threatening in susceptible individuals than in the normal people.

[Blastocystis hominis- parasites or commensals? ].

Blastocystis hominis (B. hominis) is a cosmopolitan pro- tozoa which parasitizes the human large intestine. This parasite had been considered to be commensal of the large intestine for a long time, because even an intense invasion may be asymptomatic. However, this species is now being regarded as a parasitic organism. In this paper the latest data concerning the epidemiology, diagnostics and treatment of B. hominis invasion have been cited and discussed.

Hosts and parasites as aliens.

Over the past decades, various free-living animals (hosts) and their parasites have invaded recipient areas in which they had not previously occurred, thus gaining the status of aliens or exotics. In general this happened to a low extent for hundreds of years. With variable frequency, invasions have been followed by the dispersal and establishment of non-indigenous species, whether host or parasite. In the literature thus far, colonizations by both hosts and parasites have not been treated and reviewed together, although both are usually interwoven in various ways. As to those factors permitti…

Gastrointestinal parasites in the immunocompromised.

PURPOSE OF REVIEW: Parasites and other infections have many effects on the gastrointestinal tract of individuals who are immunocompromised. Few reviews focus on parasitic infections, which are covered here. RECENT FINDINGS: The review first examines recent advances in our understanding of the taxonomy, diagnosis and treatment of pathogens such as cryptosporidia, cyclospora, isospora and microsporidia, which are recognized causes of diarrhoea in the immunocompromised, and discusses possible links between amoebiasis and HIV. The complex interactions of both intact and abnormal immune systems wi…

Bartonellae as elegant hemotropic parasites.

Bartonella species are hemotropic bacterial parasites of a wide range of mammals that occasionally cause disease in humans. The low prevalence of clinical manifestations compared to the high prevalence of infection underlines the elegance of these parasites that carefully exploit their hosts in a manner that optimizes their transmission. Recent research efforts have begun to determine the strategies involved in this exploitation, and significant progress has been made in unraveling an unusually complex natural cycle. Studies aimed at determining bacterial attributes involved in parasitism char…

Gene targeting in malaria parasites.

Gene targeting, which permits alteration of a chosen gene in a predetermined way by homologous recombination, is an emerging technology in malaria research. Soon after the development of techniques for stable transformation of red blood cell stages of Plasmodium falciparum and Plasmodium berghei, genes of interest were disrupted in the two species. The main limitations of gene targeting in malaria parasites result from the intracellular growth and slow replication of these parasites. On the other hand, the technology is facilitated by the very high rate of homologous recombination following tr…

Respiratory distress caused by parasites.

Parasitic etiologies of pulmonary symptoms generally represent some degree of immunocompromise or occur as uncommon presentations of common parasitic infections. In regions where HIV infection is more prominent, PCP probably will be the most common parasitic respiratory disease seen. In geographic regions where helminthic infections are more common, patients with respiratory symptoms and eosinophilia probably will be seen more often and require a search for a specific etiology. In more rare circumstances, travel to an endemic area may suggest a more exotic parasitic etiology for respiratory sy

The molecular epidemiology of parasites.

The explosion of new techniques, made available by the rapid advance in molecular biology, has provided a battery of novel approaches and technology which can be applied to more practical issues such as the epidemiology of parasites. In this review, we discuss the ways in which this new field of molecular epidemiology has contributed to and corroborated our existing knowledge of parasite epidemiology. Similar epidemiological questions can be asked about many different types of parasites and, using detailed examples such as the African trypanosomes and the Leishmania parasites, we discuss the t…

Immune responses to filarial parasites.

Filarial nematode parasites are long-lived organisms which are responsible for one of the major tropical diseases. Their ability to survive may be related to the antigen-specific anergic state observed in carriers of the parasite. Severe disease symptoms are associated with the breakdown of anergy. Different stages of the parasite life cycle are anti-genetically distinct, and a state of concomitant immunity is therefore possible which prevents superinfection. The evidence for these conclusions is discussed, in the context of the need for an effective vaccine against filarial parasites which wi…

[Intestinal parasites and lactose malabsorption].

Breath-test was performed for diagnosis of lactose malabsorption in 50 Gabonese children of normal nutritional status, aged 5 to 15 years, with parasites in stools, but without diarrhoea or digestive symptoms. Control group was unparasitized and consisted of 17 children and 18 young adults living in the same area. Parasites discovered by stool examination were Ascaris lumbricoides in 76% of parasitized children, Trichuris trichiura in 58%, Giardia in 24%, Entamoeba histolytica in 20%, Schistosoma intercalatum in 16% and Necator Americanus in 14%. Children were given a 10 g lactose load and adu…

Parasites of the Christmas turkey.

In the temperate West, the turkey remains popular fare for festive feasts. It is a large bird, amenable to intensive rearing, and now represents one of the cheapest forms of poultry meat available (Box 1). In the USA alone, nearly 100 million birds are raised annually - mainly in Minnesota and North Carolina. But intensive rearing can incur risks of epizootic parasitic diseases, often responsible for severe economic losses. Improved management and medication have reduced the impact of some, such as 'gapezvorm disease', histomoniasis and intestinal coccidiosis; leucocytozoonosis now presents le

External parasites of dairy cattle.

Insects such as stable flies, house flies, horn flies, face flies, mosquitoes, horse flies, deer flies, cattle grubs, and lice as well as ticks and mites are the major external parasites of dairy animals in the United States. These pests cause obvious discomfort to livestock and economic effects of heavy populations usually are discerned easily. However, research efforts to date have failed to establish reproducible losses caused by specific ectoparasites. Research efforts involving a multidisciplinary systems approach are necessary to provide the data needed to develop total dairy management …
