Phytomedicines (medicines derived from plants) for sickle cell disease.

BACKGROUND: Sickle cell disease (SCD), a common recessively inherited haemoglobin disorder, affects people from sub-Saharan Africa, the Middle East, Mediterranean basin, Indian subcontinent, Caribbean and South America. It is associated with complications and a reduced life expectancy. Phytomedicines (medicine derived from plants in their original state) encompass many of the plant remedies from traditional healers which the populations most affected would encounter. There has been little systematic appraisal of their benefits. OBJECTIVES: To assess the benefits and risks of phytomedicines in

RDB--a new product of plant origin for cell culture dispersion.

RDB is a crude preparation extracted from a plant source. It is a very potent agent for dispersion of monolayer cell cultures as well as for the preparation of primary cell suspensions. It was found to be most effective in dispersing a wide range of animal and mosquito cells and dissociating neuronal and muscular tissue for primary cell cultures. RDB was also found to be useful for preparation of primary culture for chick embryo fibroblasts. After treatment with RDB cell viability was found to be 98% as compared to 85% in trypsin treated monolayer cultures. Routine dispersion of monolayer cult

Stress-Resistant Plant Cell-Wall Inspired Cell Armor Enables Red Blood Cell Lyophilization.

Lyophilization offers an attractive route for ambient-temperature storage of biological systems by eliminating cold-chain dependence and suppressing solution-phase degradation. However, lyophilizing mammalian cells remains challenging due to their vulnerability to coupled stresses, including ice-induced mechanical injury and membrane destabilization during freezing and dehydration. In nature, stress-resistant plant cells survive extreme conditions through multifunctional cell walls. Inspired by their natural survival strategies, we engineer an artificial membrane armor (AMA) that enables effic…

Adaptogens in chemobrain (Part II): Effect of plant extracts on chemotherapy-induced cytotoxicity in neuroglia cells.

BACKGROUND: Cancer chemotherapy-induced cognitive impairments are apparently associated with harmful effects on physiological functions of brain cells. Adaptogens, are known to exhibit neuroprotective effects and to increase cognitive functions in clinical studies. In our previous study (Seo et al., 2018), we demonstrated that selected adaptogenic extracts significantly attenuate cytostatic-induced regulation of more than 100 genes involved in the activation of neuronal death and inhibiting neurogenesis. Neuroprotective and cytoprotective activities of adaptogens rise the question about their

Quantitative HPLC-MS analysis of nucleotide sugars in plant cells following off-line SPE sample preparation.

An analytical workflow was developed for the absolute quantification of uridine diphosphate (UDP)-sugars in plant material in order to compare their metabolism both in wild-type Arabidopsis thaliana and mutated plants (ugd2,3) possessing genetic alterations within the UDP-glucose dehydrogenase genes involved in UDP-sugar metabolism. UDP-sugars were extracted from fresh plant material by chloroform-methanol-water extraction and further purified by solid-phase extraction with a porous graphitic carbon adsorbent with extraction efficiencies between 80 ± 5 % and 90 ± 5 %. Effects of phytoestrogens and other plant-derived compounds on mesenchymal stem cells, bone maintenance and regeneration.

Phytoestrogens and other plant-derived compounds and extracts have been developed for the treatment of menopause-related complaints and disorders, e.g. hot flushes and osteoporosis. Since estrogens have been discussed to enhance the risk for hormone-sensitive cancers, research activities try to find alternatives. Phytoestrogens like genistein and resveratrol as well as other plant-derived compounds are capable of substituting for estrogens to some extent. Their effects on mesenchymal stem cells and the tissues derived therefrom have been investigated in vitro and in preclinical settings. Besid

In vitro activities of plant extracts on human Loa loa isolates and cytotoxicity for eukaryotic cells.

Loa loa, a filarial worm, can cause fatal encephalitis in humans. In an attempt to find alternatives to the standard treatments (ivermectin and diethylcarbamazine citrate), we tested 12 methanolic extracts of nine traditional plant remedies. The extracts (100-0.09 microg/ml) were incubated with 20 Loa loa microfilariae isolated from patients at 37 degrees C with 5% CO(2) in modified Eagle's medium supplemented with 10% fetal serum and antibiotics. Activity was evaluated 120 h later by counting live microfilariae under a microscope. Cytotoxicity for eukaryotic cells was estimated by measuring 3

Plant polyphenols effectively protect HaCaT cells from ultraviolet C-triggered necrosis and suppress inflammatory chemokine expression.

Oxidative stress is a common response of epidermal cells to a variety of noxious stimuli such as ultraviolet (UV) radiation from solar light and proinflammatory cytokines from skin-infiltrating leukocytes. Here, we report that two types of plant-derived antioxidants, the phenylpropanoid glycoside verbascoside as well as the flavonoids rutin and quercetin possess protective effects against UVC-induced cell damage and proinflammatory activation. The molecules under investigation were effective against the loss of cell integrity associated with necrosis in doses consistent with their antioxidant

Escherichia coli common pilus (ECP) targets arabinosyl residues in plant cell walls to mediate adhesion to fresh produce plants.

Outbreaks of verotoxigenic Escherichia coli are often associated with fresh produce. However, the molecular basis to adherence is unknown beyond ionic lipid-flagellum interactions in plant cell membranes. We demonstrate that arabinans present in different constituents of plant cell walls are targeted for adherence by E. coli common pilus (ECP; or meningitis-associated and temperature-regulated (Mat) fimbriae) for E. coli serotypes O157:H7 and O18:K1:H7. l-Arabinose is a common constituent of plant cell wall that is rarely found in other organisms, whereas ECP is widespread in E. coli and other

The Plant Compound Sulforaphane Attenuates Induction of Cytokines and Other Early Activation Events in Jurkat Cells.

Sulforaphane (SFN), a bioactive isothiocyanate found in cruciferous vegetables, has gained considerable interest for its anti-inflammatory and immunomodulatory properties. Previous work in our lab demonstrated that tert-butylhydroquinone (tBHQ), a common food preservative and synthetic antioxidant, suppresses early T cell activation events, including production of interleukin-2 (IL-2) and other cytokines, cell proliferation and induction of interleukin-2 receptor alpha (CD25) and other inducible cell surface receptors. Based on these findings, the current study explores the effects of sulforap

Plant cells at the organ surface use mechanical cues to activate a specific growth-control program.

During morphogenesis of multicellular organs, cells in distinct positions need to meet specific functional requirements to form the appropriate tissue type. For example, epidermal tissues need to form at the organ surface. In plants, the epidermis is widely considered essential for morphogenesis due to its role in both restricting and promoting growth. In the root, epidermal cells are partially covered by a protective root cap and partially positioned at the organ surface. Here, we propose that epidermal cells at the organ surface have unique requirements for growth control due to high mechani

A hydroxyl coumarin-chalcone-based fluorescent probe for sensing copper ions in plant and living cells.

Copper ions are essential trace elements for living organisms. However, its excessive levels in the liver and kidneys may lead to severe health issues. Therefore, developing effective fluorescent probes for detecting copper ions is crucial. A hydroxyl coumarin chalcone derivative featuring a hydroxyl substituent has been developed as a fluorescent probe for monitoring Cu2+. The probe can selectively detect Cu2+ in aqueous solutions, exhibiting a visible color change from red to light yellow to the naked eye. This is accompanied by a quenching of the red fluorescence, with a limit of detection

Plant and animal-derived fusion nanovesicles rescue inflammation-compromised osteogenic potential of periodontal ligament stem cells.

Periodontitis is a chronic inflammatory disease affecting the supporting tissues of the teeth and has emerged as a global public health issue. Current therapies primarily address pathogenic factors and alleviate symptoms, with limited options available for complete restoration and reconstruction of already absorbed periodontal bone tissue. In this study, we developed a nanotherapeutic strategy utilizing fusion nanovesicles (FVs) to modulate the inflammatory microenvironment and create a regenerative niche for periodontal ligament stem cells (PDLSCs), which play a crucial role in periodontal ti

Genome editing by introduction of Cas9/sgRNA into plant cells using temperature-controlled atmospheric pressure plasma.

Previously, we developed a technique to introduce a superfolder green fluorescent protein (sGFP) fusion protein directly into plant cells using atmospheric-pressure plasma. In this study, we attempted genome editing using CRISPR/Cas9 (clustered regularly interspaced short palindromic repeats/CRISPR associated protein 9) system using this protein introduction technique. As an experimental system to evaluate genome editing, we utilized transgenic reporter plants carrying the reporter genes L-(I-SceI)-UC and sGFP-waxy-HPT. The L-(I-SceI)-UC system allowed the detection of successful genome editin…
