Diverse Roles of the Salicylic Acid Receptors NPR1 and NPR3/NPR4 in Plant Immunity.

The plant defense hormone salicylic acid (SA) is perceived by two classes of receptors, NPR1 and NPR3/NPR4. They function in two parallel pathways to regulate SA-induced defense gene expression. To better understand the roles of the SA receptors in plant defense, we systematically analyzed their contributions to different aspects of Arabidopsis (Arabidopsis thaliana) plant immunity using the SA-insensitive npr1-1 npr4-4D double mutant. We found that perception of SA by NPR1 and NPR4 is required for activation of N-hydroxypipecolic acid biosynthesis, which is essential for inducing systemic acq

PRR2, a pseudo-response regulator, promotes salicylic acid and camalexin accumulation during plant immunity.

Calcium signalling mediated by Calmodulin (CaM) and calmodulin-like (CML) proteins is critical to plant immunity. CaM and CML regulate a wide range of target proteins and cellular responses. While many CaM-binding proteins have been identified, few have been characterized for their specific role in plant immunity. Here, we report new data on the biological function of a CML-interacting partner, PRR2 (PSEUDO-RESPONSE REGULATOR 2), a plant specific transcription factor. Until now, the physiological relevance of PRR2 remained largely unknown. Using a reverse genetic strategy in A. thaliana, we id

Plant immunity triggered by engineered in vivo release of oligogalacturonides, damage-associated molecular patterns.

Oligogalacturonides (OGs) are fragments of pectin that activate plant innate immunity by functioning as damage-associated molecular patterns (DAMPs). We set out to test the hypothesis that OGs are generated in planta by partial inhibition of pathogen-encoded polygalacturonases (PGs). A gene encoding a fungal PG was fused with a gene encoding a plant polygalacturonase-inhibiting protein (PGIP) and expressed in transgenic Arabidopsis plants. We show that expression of the PGIP-PG chimera results in the in vivo production of OGs that can be detected by mass spectrometric analysis. Transgenic plan

African Burkitt's lymphoma: a plant, Euphorbia tirucalli, reduces Epstein-Barr virus-specific cellular immunity.

Endemicity of Burkitt's lymphoma (BL) coincides with profusion of a plant Euphorbia tirucalli in tropical Africa. E. tirucalli contains 4-deoxyphorbol ester that enhances Epstein-Barr virus (EBV) infection of B lymphocytes. In this study, we found that 4-deoxyphorbol ester reduced EBV-specific cytotoxic T-cell function. Furthermore, the B lymphocytes dually exposed to EBV and 4-deoxyphorbol ester were resistant to EBV-specific T cell cytotoxicity, through down-regulation of latent membrane protein 1 (LMP1), the major target to EBV-specific cytotoxic T-cells. These immunologic findings strength

Loss of Arabidopsis ferroxidases LPR1 and LPR2 enhances plant immunity by promoting leaf iron deposition.

Iron (Fe) is a critical nutrient for both plants and their pathogens, and its distribution and homeostasis critically influence the outcome of plant-pathogen interactions. Low Phosphate Root 1 (LPR1) and LPR2 are ferroxidases that regulate Fe translocation in the xylem, but their role in plant immunity remains unknown. Here, we report that loss of LPR1 and LPR2 in Arabidopsis thaliana confers enhanced resistance to both virulent (Pseudomonas syringae pv. Tomato DC3000, Pst) and avirulent (Pst-avrRpt2) bacterial strains. This enhanced resistance is associated with excessive Fe deposition in the

Effector FsSCR6 contributes to the virulence of Fusarium sacchari on sugarcane and suppresses plant immunity.

Pokkah Boeng disease caused by Fusarium sacchari seriously threatens the yield and quality of sugarcane worldwide. Effectors play a crucial role in the infection and colonization of pathogens. However, there were few reports on the virulence functions of F. sacchari effectors. To characterize effector functions and unravel the pathogenic mechanisms of F. sacchari, we identified an effector FsSCR6, which was vital for the virulence of F. sacchari. Gene knockout mutants showed no difference in growth rates and colony morphology from wild-type. However, the virulence of knockout mutants was sever

LURP1, a SARD1-regulated protein, coordinates with the helper NLR ADR1 to promote plant immunity.

In Arabidopsis, loss of transcription factors Calmodulin-Binding Transcription Activators 1/2/3 (CAMTA1/2/3) results in enhanced defense responses with increased salicylic acid (SA) and N-hydroxy-pipecolic acid (NHP) biosynthesis. This is achieved through modulating the expression of master immune transcription factors SAR Deficient 1 (SARD1) and Calmodulin-Binding Protein 60-like g (CBP60g). SARD4 is an NHP biosynthesis enzyme, the camta2/3 sard4 triple mutant remains dwarfed and autoimmune. To identify positive immune regulators of the SA and NHP pathways, a forward genetic screen was perfor

A conserved small RNA-generating gene cluster undergoes sequence diversification and contributes to plant immunity.

Small RNA-mediated gene silencing contributes to plant immunity. The secondary small interfering RNA (siRNA) pathway promotes defense by silencing target genes in invading fungal and oomycete pathogens. Many secondary siRNAs are derived from transcripts potentially encoding pentatricopeptide repeat (PPR) proteins. Here, we report that siRNA production is an ancient function of a conserved clade of PPR genes that undergo extensive within-species diversification. In Arabidopsis thaliana, siRNA-source PPRs are physically clustered on Chromosome 1. These sequences are diversified through gene dupl

An effector essential for virulence of necrotrophic fungi targets plant HIRs to inhibit host immunity.

Phytopathogens often secrete effectors to enhance their infection of plants. In the case of Sclerotinia sclerotiorum, a necrotrophic phytopathogen, a secreted protein named SsPEIE1 (Sclerotinia sclerotiorum Plant Early Immunosuppressive Effector 1) plays a crucial role in its virulence. During the early stages of infection, SsPEIE1 is significantly up-regulated. Additionally, transgenic plants expressing SsPEIE1 exhibit increased susceptibility to different phytopathogens. Further investigations revealed that SsPEIE1 interacts with a plasma membrane protein known as hypersensitive induced reac

Plant immunity induced by COS-OGA elicitor is a cumulative process that involves salicylic acid.

Plant innate immunity offers considerable opportunities for plant protection but beside flagellin and chitin, not many molecules and their receptors have been extensively characterized and very few have successfully reached the field. The Effect of Plant Quercetin on Immune Function of Microtus Fortis.

Quercetin, a widely occurring phytogenic flavonoid, is the primary secondary metabolite in the diet of Microtus fortis. This study investigated the effects of dietary quercetin on immune organ development and cytokine levels in Microtus fortis. Microtus fortis were fed a standard diet containing 0, 1.5%, 3%, 5%, or 8% quercetin for 20 consecutive days. The results show that, compared to the control group, treatment with 5% quercetin increased the spleen and thymus indices by 14.47% and 11.67%, respectively. The proliferation of T lymphocytes, B lymphocytes and macrophages in Microtus fortis va

Plant sterols and sterolins: a review of their immune-modulating properties.

Beta-sitosterol (BSS) and its glycoside (BSSG) are sterol molecules which are synthesized by plants. When humans eat plant foods phytosterols are ingested, and are found in the serum and tissues of healthy individuals, but at concentrations orders of magnitude lower than endogenous cholesterol. Epidemiological studies have correlated a reduced risk of numerous diseases with a diet high in fruits and vegetables, and have concluded that specific molecules, including b-carotene, tocopherols, vitamin C, and flavonoids, confer some of this protective benefit. However, these epidemiologic studies ha

Glycan recognition by a plant damage-sensing immune receptor.

Pathogens target and degrade the extracellular matrix surrounding plant cells. A central question is how cell wall-derived damage-associated molecular patterns (DAMPs) are recognized and integrated to trigger immune responses. We address this question by determining the structure of the extracellular multidomain of the IGP1 receptor in both apo form and bound to the cellulose-derived DAMP cellotriose. Structural analyses reveal that constitutive interactions of Leucine Rich Repeat (LRR)-malectin domains preconfigure IGP1 for ligand recognition and that the receptor features a highly specific s

Hopeaphenol targets histidine kinase signaling across kingdoms to suppress bacterial virulence and potentiate plant immunity.

Small molecules can control plant disease either by disarming the pathogen or by priming host immunity, but single compounds that do both are rare and mechanistically unexplained. Here we show that hopeaphenol (HP), a resveratrol tetramer from stilbene-producing plants, acts on histidine kinase (HK) signaling on both sides of the Arabidopsis- Pseudomonas interaction. In Pseudomonas syringae pv. tomato DC3000, HP represses the type III secretion regulon and motility genes, restricts surface motility independently of effector delivery, and binds a defined subset of virulence-associated sensor ki

Decoding the rhizosphere immune signaling network: Microbiome-driven modulation of plant immunity and disease resistance.

Classical plant immunity models rely on pattern-triggered immunity (PTI) and effector-triggered immunity (ETI), yet these frameworks largely overlook the rhizosphere microbiome as a functional determinant of disease resistance. Plants act as holobionts, their root exudates comprising flavonoids, coumarins, benzoxazinoids, strigolactones, and primary metabolites selectively recruit microbial communities whose composition is shaped by host genotype and dynamically reprogrammed under biotic and abiotic stress. Recruited microorganisms return a diverse signal range including flagellin, siderophore

The dual role of Xanthomonas effector XccXynB in plant cell wall deconstruction and immunity activation.

The GH10 endoxylanase XccXynB from Xanthomonas campestris pv. campestris (Xcc) functions as a PAMP, triggering PTI responses and enhancing disease resistance in plants, in addition to its polysaccharide degradation capability. Plant pathogens of the Xanthomonas genus are capable of infecting a wide array of economically important plant species. Among their virulence factors, the GH10 family xylanase, XynB, is typically responsible for the predominant extracellular xylanase activity. However, the detailed biochemical characteristics of this enzyme in most Xanthomonas pathogens remain insufficie

Late Blight Effector Pi05910 Suppresses Plant Immunity by Promoting Destabilisation of Host Scaffold Protein RACK1.

Late blight, caused by oomycete pathogen Phytophthora infestans, is one of the most devastating diseases of potato and tomato. Plant resistance to pathogens relies on a sophisticated innate immune system, which is often targeted by pathogen-derived effectors. P. infestans secretes numerous RXLR effectors to suppress host immunity. In this study, we show that the P. infestans core effector Pi05910 functions as a virulence factor and targets the host scaffold protein Receptor for Activated C Kinase 1 (RACK1), which is identified to positively regulate potato resistance to late blight. Functional

Pseudomonas syringae lytic transglycosylase HrpH interacts with host ubiquitin ligase ATL2 to modulate plant immunity.

Pseudomonas syringae deploys a type III secretion system (T3SS) to deliver effector proteins to facilitate infection of plant cells; however, little is known about the direct interactions between T3SS components and plants. Here, we show that the specialized lytic transglycosylase (SLT) domain of P. syringae pv. tomato (Pst) DC3000 T3SS component HrpH is necessary for effector translocation. HrpH and its SLT domain induce host cell death and suppress pattern-triggered immunity (PTI). Transgenic hrpH-Arabidopsis plants exhibit decreased PTI responses and enhanced susceptibility to Pst DC3000Δhr

A nanodiamond-formulated plant protein induces robust immunity against porcine epidemic diarrhea virus in piglets.

Porcine epidemic diarrhea virus (PEDV) continues to be a major infectious threat in swine, especially endangering piglets. The COE and S1D domains have been identified as crucial antigens suitable for designing subunit vaccines. Nanodiamonds (NDs), owing to their biocompatibility, large surface area, and modifiable surfaces, have gained interest as novel carriers to improve recombinant protein vaccines. In this study, we transiently expressed a COE-S1D fusion protein containing the GCN4pII motif (COE-S1D-pII) in Nicotiana benthamiana. The recombinant protein was subsequently mixed with nanodia
