Illuminating chromaffin granules: compartmentalization of endocannabinoids in bovine adrenal glands supports role in hormone modulation.

The endocannabinoid system (ECS) plays a major role in the central stress response. However, its role in the sympathetic nervous system remains poorly understood. Knowledge about the spatial distribution and function of endocannabinoids (eCBs) in adrenals is lacking. Here, we analysed lipid eCBs and alpha-hemoglobin-derived peptide endocannabinoids (pepcans) across distinct zones of the bovine adrenal gland and investigated the effects of RVD-hemopressin (pepcan-12) and anandamide (AEA) on steroidogenesis. To challenge the hypothesis that pepcans are produced in the adrenal medulla, bovine chr

Piperine inhibits biofilm formation and efflux activity and dysregulates lipid metabolism in Mycobacterium abscessus.

The nontuberculous Mycobacterium abscessus is a human pathogen that causes chronic lung infections and soft tissue infections. The bacterium forms biofilms and efflux pumps contribute to its tolerance of antibiotics. Efflux pumps also transport lipids and other molecules to the bacterial outer cell surface for biofilm formation. The effects of piperine, an alkaloid derived from black pepper, on biofilm formation, efflux activity and lipid biosynthesis in M. abscessus have not been reported. We report that, at sub-minimum inhibitory concentration levels, piperine inhibits biofilm formation in M

Investigation of endocannabinoids in plasma and their correlation with physical fitness and resting state functional connectivity of the periaqueductal grey in women with fibromyalgia: An exploratory secondary study.

OBJECTIVES: The endocannabinoid system is a neuromodulatory system linked to pain, distress, and physical activity. The endocannabinoids (eCBs) arachidonoylethanolamide (AEA) and 2-arachidonoylglycerol are known to activate the cannabinoid receptors. The eCB system has been reported to be acutely affected by physical activity. Altered connectivity related to pain intensity in patients with fibromyalgia (FM) has been reported. However, how the peripheral eCBs, including eCB-like N-acylethanolamines (NAEs) concentrations, associate with pain-related regions of the brain and physical activity in

The impact of piperine on the metabolic conditions of patients with NAFLD and early cirrhosis: a randomized double-blind controlled trial.

Non-alcoholic fatty liver disease (NAFLD) is a metabolic dysfunction of the liver defined as an abnormal accumulation of fat within the liver without secondary triggers like alcohol consumption or viral hepatitis. Piperine, the bio-active ingredient of black pepper, can exert a significant function in treatment of individuals with NAFLDand early cirrhosis. We investigated the impact of piperine consumption with a duration of 12 weeks on patients with NAFLD and early cirrhosis compared toplacebo consumption. In a double-blind study, patients with NAFLD and early stage of cirrhosis were haphazar

Oral Administration of Piperine Ameliorates Experimental Autoimmune Uveitis.

This study aimed to evaluate the impact of piperine on experimental autoimmune uveitis (EAU). EAU was induced by immunization with interphotoreceptor retinoid-binding protein emulsified in complete Freund adjuvant. Starting from day 8 post-induction, Lewis rats were given piperine (0, 20, 40, and 80 mg/kg-P.O.) or prednisolone (10 mg/kg-P.O.) for 18 consecutive days. The 80 mg/kg dose of piperine demonstrated superior regression of clinical symptoms, increased nitric oxide levels, and enhanced IDO activity in eye homogenates compared to other doses. The 40 and 80 mg/kg doses of piperine were m

Antifungal Activity of Piperine-based Nanoemulsion Against Candida spp. via In Vitro Broth Microdilution Assay.

Candidemia leaves a trail of approximately 750,000 cases yearly, with a morbidity rate of up to 30%. While Candida albicans still ranks as the most predominantly isolated Candida species, C. glabrata comes in second, with a death rate of 40-50%. Although infections by Candida spp are commonly treated with azoles, the side effects and rise in resistance against it has significantly limited its clinical usage. The current study aims to address the insolubility of piperine and provide an alternative treatment to Candida infection by formulating a stable piperine-loaded O/W nanoemulsion, comprised

Synthesis and evaluation of Piperine analogs as thioredoxin reductase inhibitors to cause oxidative stress-induced cancer cell apoptosis.

Inhibiting thioredoxin reductase (TrxR) to disrupt the redox equilibrium and induce tumor cell apoptosis is a significant tumor therapeutic strategy. Piperine, a natural product from black pepper, has been demonstrated to suppress tumor cell proliferation by enhancing reactive oxygen species (ROS), subsequently leading to cell death. However, the development of Piperine as an active molecule is hampered by its weak cytotoxicity. To develop a compound with higher activity, we synthesized 22 Piperine analogs and evaluated their pharmacological properties. Ultimately, B5 was screened by the resul

Piper retrofractum extract and its component piperine promote lymphangiogenesis via an AKT- and ERK-dependent mechanism.

Administration of Piper retrofractum extract (PRE) has been reported to alleviate edema, but the mechanism underlying this effect is unknown. Promotion of lymphangiogenesis is known to improve lymphedema, but the effect of PRE on lymphangiogenesis remains unclear. In the present study, we investigated whether PRE and specifically, piperine, the main component of PRE, can induce lymphangiogenesis. Treatments with PRE and piperine significantly promoted the proliferation, migration, and tube formation in human dermal lymphatic microvascular endothelial cells (HDLECs) but had no effect on the exp

Biofilm and hyphal inhibitory synergistic effects of phytoactives piperine and cinnamaldehyde against Candida albicans.

Oral candidiasis, the most common mycotic infection of the human oral cavity is non-life-threatening yet, if untreated, may advance as systemic infections. The ability of Candida albicans to adapt sessile lifestyle imparts resistance to drugs and host immunity. Consequently, due to the limited effectiveness of conventional antifungal treatment, novel therapeutic strategies are required. In the present study, synergistic interaction of phytochemicals, piperine, and cinnamaldehyde against the biofilm and hyphal of C. albicans was evaluated. Minimum inhibitory concentration (MIC) and biofilm inhi

Piperine, an active ingredient of white pepper, suppresses the growth of multidrug-resistant toxigenic Vibrio cholerae and other pathogenic bacteria.

Emergence and rapid spread of multidrug-resistant (MDR) bacteria including Vibrio cholerae are a global public health issue. Much attention has been paid to natural compounds, such as spices and herbs to find novel antimicrobial compounds as they are considered to be cheaper alternatives to develop as a drug. Here, we show that methanol extract of white pepper could inhibit the growth of V. cholerae O1 El Tor variant, responsible for the recent outbreaks/epidemics. Furthermore, we demonstrate for the first time that piperine, the major component of white pepper, showed a dose-dependent bacteri

Efficient antileishmanial activity of amphotericin B and piperine entrapped in enteric coated guar gum nanoparticles.

Amphotericin B (AmB) exhibits potential antileishmanial activity, with only a little rate of recurrence. However, low bioavailability and severe nephrotoxicity are among the major shortcomings of AmB-based therapy. Various AmB nanoformulations have been developed, which to an extent, have reduced its toxicity and increased the drug efficacy. To further reduce the nonspecific tissue distribution and the cost of the treatment, the current AmB-based formulations require additional improvements. Combination of natural bioenhancers with AmB is expected to further increase its bioavailability. There

Blood levels of the endocannabinoid anandamide are increased in anorexia nervosa and in binge-eating disorder, but not in bulimia nervosa.

The endocannabinoid system, consisting of two cannabinoid receptors (CB1 and CB2) and the endogenous ligands anandamide (arachidonoylethanolamide (AEA)) and 2-arachidonoylglycerol (2-AG), has been shown to control food intake in both animals and humans, modulating either rewarding or quantitative aspects of the eating behavior. Moreover, hypothalamic endocannabinoids seem to be part of neural circuitry involved in the modulating effects of leptin on energy homeostasis. Therefore, alterations of the endocannabinoid system could be involved in the pathophysiology of eating disorders, where a der

The effect of topical application of the platelet-activating factor-antagonist, Ro 24-0238, in psoriasis vulgaris--a clinical and immunohistochemical study.

Platelet-activating factor (PAF) is considered to be one of the most potent lipid mediators in allergic and inflammatory reactions. Suggestions that PAF is produced by cutaneous cells, and cells infiltrating the skin from the blood, have been reported. PAF has been identified in allergic cutaneous reactions and also in psoriatic lesions. The biological activity of PAF is thought to be mediated by cell membrane receptors. Studies revealed that PAF-antagonists can be active in animal models of cutaneous inflammation. In humans PAF-antagonists showed minimal therapeutic improvement in studies of

Low-intensity exercise reduces negative affect via circulating endocannabinoid dynamics in healthy young adults.

PURPOSE: Mental health disorders represent a critical challenge in modern society, necessitating the establishment of effective management strategies. Low-intensity exercise is thought to improve mood by activating endocannabinoids (eCBs), but the underlying mechanisms remain unclear. This study aimed to test the hypothesis that an increase in circulating eCB levels during low-intensity exercise contributes to a reduction in negative affect. METHODS: In a randomized crossover trial, 30 healthy participants completed two 60-min cycling exercise sessions at two intensities: low (30% heart rate

Alterations of the endocannabinoid system in adolescents with non-suicidal self-injury as a function of childhood maltreatment.

Non-suicidal self-injury (NSSI) is a highly prevalent phenomenon in adolescence, often associated with prior traumatic experiences. The development and maintenance of NSSI is associated with dysregulation of the stress response, and evidence suggests that the hypothalamic-pituitary-adrenal (HPA) axis plays an important role. The endocannabinoid system is a neuromodulatory system in close functional interaction with the HPA axis. Several studies have reported alterations of the endocannabinoid system in adult patients with post-traumatic stress disorder. However, the role of the endocannabinoid
