Strain-specified characteristics of mouse synthetic prions.

Synthetic prions were produced in our laboratory by using recombinant mouse prion protein (MoPrP) composed of residues 89-230. The first mouse synthetic prion strain (MoSP1) was inoculated into transgenic (Tg) 9949 mice expressing N-terminally truncated MoPrP(Delta23-88) and WT FVB mice expressing full-length MoPrP. On first and second passage in Tg9949 mice, MoSP1 prions caused disease in 516 +/- 27 and 258 +/- 25 days, respectively; numerous, large vacuoles were found in the brainstem and gray matter of the cerebellum. MoSP1 prions passaged in Tg9949 mice were inoculated into FVB mice; on fi…

Prions and the blood and immune systems.

Prion diseases take a number of forms in animals and humans. They are caused by conformational change in widely expressed prion protein leading to the formation of intracellular aggregates. Although the main focus of disease is the central nervous system, it is known that involvement of the immune system occurs in peripherally transmitted disease in particular. Animal experiments suggest that in some prion diseases follicular dendritic cells in the germinal centers are a major site of initial accumulation, and that abnormal prion protein and infectivity are detectable in peripheral lymphoid ti

A virus behind the mask of prions?

The prion notion, that a host-encoded protein reacts with itself to become an infectious entity, has become highly favoured as the causal agent in transmissible encephalopathies (TSEs) such as endemic scrapie of sheep, epidemic bovine encephalopathy (BSE), and rare forms of human Creutzfeldt-Jakob Disease (CJD). Much of the claimed "wealth of data" supporting the notion of prions, however, has been irreproducible as well as contradicted by many experimental observations. By 1989, for example, it was already shown that the "infectious prion conformation" could be eliminated without altering eit…

Prions and the human transmissible spongiform encephalopathies.

The last 5 years have seen the emergence of a new disease in humans (vCJD), mainly in the United Kingdom. This emergence has been accompanied by an explosion of scientific data on a novel group of the responsible infectious agents called prions and has profound implications for infection control and transfusion policies. Also of concern is the finding of prions in neural, gingival, pulpal, and salivary tissue in animal models and significant titers of infectivity from extraneural organs (particularly, in cases of vCJD, in lymphoreticular tissues). There is limited information on the presence o…

Creutzfeldt-Jakob disease--identifying prions and carriers.

Two major obstacles to be overcome in minimizing the possibility of transmitting Creutzfeldt-Jakob disease or variant Creutzfeldt-Jakob disease are associated with identifying the prion on disinfected surgical instruments and identifying carriers of the protease-resistant form of the prion. New developments indicate that the means for doing both soon may be available.

[Human spongiform encephalopathies. Diseases caused by prions].

Spongiform encephalopathies or prion diseases are common denominators for a group of diseases, all fatal, which show characteristic neuropat … ficance. PrP is highly conserved among mammals and is expressed in most tissues. Diseases caused by prions are exceptional as they can occur sporadically and are transmissible both genetically and infectiously. This review attempts to elucidate the present knowledge of the natural history of these diseases in man.

Prions--infectious pathogens causing the spongiform encephalopathies.

The novel properties of the scrapie and Creutzfeldt-Jakob disease (CJD) transmissible agents readily distinguish them from viruses and viroids; thus, they have been labeled "prions". The scrapie prion contains a protein(s) which is required for infectivity; recently a 27,000 to 30,000 MW protein which purifies with the prion has been identified. The similarities between the scrapie and CJD agents suggest that CJD is also caused by a prion. Recent studies show that the time courses of both scrapie and CJD are determined by an autosominal dominant gene denoted PID (prion incubation determinant).…

Biochemical fingerprints of prion diseases: scrapie prion protein in human prion diseases that share prion genotype and type.

The phenotype of human prion diseases is influenced by the prion protein (PrP) genotype as determined by the methionine (M)/valine (V) polymorphism at codon 129, the scrapie PrP (PrPSc) type and the etiology. To gain further insight into the mechanisms of phenotype determination, we compared two-dimensional immunoblot profiles of detergent insoluble and proteinase K-resistant PrP species in a type of sporadic Creutzfeldt-Jakob disease (sCJDMM2), variant CJD (vCJD) and sporadic fatal insomnia (sFI). Full-length and truncated PrP forms present in the insoluble fractions were also separately anal

Sodium hypochlorite inactivation of human CJD prions.

Prion diseases are transmissible, fatal neurologic diseases of mammals caused by the accumulation of mis-folded, disease associated prion pr … earch has shown that concentrated sodium hypochlorite (bleach) was effective at inactivation of CJD prions derived from brains of mice or guinea pigs. Unfortunately, human prions adapted to rodents may mis-fold differently than in humans, and the rodent adapted prions may not have the same resistance or susceptibility to inactivation present in bona fide …

Prions hijack tunnelling nanotubes for intercellular spread.

In variant Creutzfeldt-Jakob disease, prions (PrP(Sc)) enter the body with contaminated foodstuffs and can spread from the intestinal entry site to the central nervous system (CNS) by intercellular transfer from the lymphoid system to the peripheral nervous system (PNS). Although several means and different cell types have been proposed to have a role, the mechanism of cell-to-cell spreading remains elusive. Tunnelling nanotubes (TNTs) have been identified between cells, both in vitro and in vivo, and may represent a conserved means of cell-to-cell communication. Here we show that TNTs allow t…

The cellular prion protein beyond prion diseases.

The cellular prion protein (PrPC), a cell surface glycoprotein originally identified for its central role in prion diseases (also called transmissible spongiform encephalopathies), has recently been implicated in the pathogenesis of other neurodegenerative disorders, such as Alzheimer’s and Parkinson’s diseases, by acting as a toxicity-transducing receptor for different misfolded protein isoforms, or in some case by exerting neuroprotective effects. Interestingly, PrPC has also been reported to play unexpected functions outside the nervous system, for example by contributing to mye

Human prion diseases with variant prion protein.

Recent molecular genetic studies revealed that the human prion protein (PrP) gene has a large repertoire of polymorphisms and mutations. Each variant PrP seems to correspond to a distinct type of prion diseases. We report herein that it is useful to classify prion diseases into plaque type or non-plaque type, based on the distribution of PrP in the central nervous system. The variant PrP including codon 102, codon 105, codon 129, codon 145 and insertional polymorphisms belong to the plaque type prion diseases, whereas the wild-type PrP and the variants including codon 180, codon 200, and codon
