[Cerebrovascular complications and hereditary protein S deficiency: 2 cases].

Hereditary protein S deficiency (HSPD) is a predisposing factor to recurrent venous thrombosis but is not currently associated with stroke. We report two cases of HSPD revealed by stroke in young adults. The first one was a 36-year-old patient whith a pure motor hemiplegia, who gradually recovered without sequelae. Total and free protein S was decreased (55 and 10%). One of his brothers died from pulmonary embolism at 20 years and a sister had low protein S level without clinical signs. The second case was a 26-year-old patient who had a right hemiplegia with aphasia due to an infarction in mi…

Protein S deficiency in middle-aged women with stroke.

We examined the relationship between free protein S deficiency and cerebrovascular disease by reviewing the records of all patients with the diagnoses of cerebral thrombosis, cerebral embolism, and cerebral vascular occlusion who were referred for coagulation studies over a 12-month period. We assayed for free protein S antigen, protein C antigen, and antithrombin III and tested for lupus-like anticoagulant and anticardiolipin antibody. Twenty-two of 267 patients (8.2%) admitted with thrombotic strokes were referred for coagulation studies. Free protein S antigen was significantly lower in wom…

S-100-protein-positive dendritic cells in nodular vasculitis.

We investigated the distribution of S-100-protein-positive dendritic cells in 20 cases of nodular vasculitis (erythema induratum Bazin). The cells were identified by immunohistochemical demonstration in paraffin sections. S-100-protein-positive dendritic cells were present in all cases and comprised up to 11% of the infiltrating cells in lymphohistiocytic areas (mean 3.7%). Because of the dendritic shape and the association with lymphocytes the cells are considered to represent T-zone histiocytes. Our findings suggest a possible role of delayed-type hypersensitivity in nodular vasculitis.

Severe arterial cerebral thrombosis in a patient with protein S deficiency (moderately reduced total and markedly reduced free protein S): a family study.

Deficiency of protein S has been associated with an increased risk of thrombotic disease as already shown for protein C deficiency. Deficiencies of any of these two proteins predispose to venous thrombosis but have been only rarely associated with arterial thrombosis. In this study we describe a case of severe cerebral arterial thrombosis in a 44-year old woman with protein S deficiency. The defect was characterized by moderately reduced levels of total and markedly reduced levels of free protein S. C4b-bp level was normal. Protein C, AT III and routine coagulation tests were within the normal…

Nitric oxide production and protein S-nitrosation in algae.

Key roles for nitric oxide in signalling processes and plant physiological processes are now well established. In particular, the identification and functional characterisation of proteins regulated by S-nitrosation, a NO-dependent post-translational modification, provided remarkable insights into the subtle mechanisms by which NO mediates its effects. Nevertheless, and despite the considerable progress in understanding NO signalling, the question of how plant cells produce NO is not yet fully resolved. Interestingly, there is now compelling evidence that algae constitute promising biological

Population-Scale Studies of Protein S Abnormalities and Thrombosis.

IMPORTANCE: Clinical decision-making in thrombotic disorders is impeded by long-standing uncertainty regarding the magnitude of venous and arterial thrombosis risk associated with low protein S. Population-scale multiomic datasets offer an unprecedented opportunity to answer questions regarding the epidemiology and clinical impacts of protein S deficiency. OBJECTIVE: To evaluate the risk associated with protein S deficiency across multiple thrombosis phenotypes. DESIGN, SETTING, AND PARTICIPANTS: Cross-sectional study using longitudinal population cohorts derived from the UK Biobank (n = 426…

Protein S-Palmitoylation as Potential Therapeutic Target for Dermatoses.

Protein S-palmitoylation is a pivotal yet poorly integrated research field in dermatology. This reversible post-translational lipid modification primarily occurs on cysteine residues and is principally catalyzed by zinc finger and Asp-His-His-Cys DHHC-domain containing proteins (zDHHCs). The S-palmitoylation/depalmitoylation cycle directly affects protein localization, trafficking, stability, and protein-protein interaction, thereby regulating a variety of signaling pathways, including those mediating inflammation and immune reaction. Accumulating evidence has indicated that S-palmitoylation r…

We established quantitative chemical proteomics for global and site-specific profiling of protein S-2-carboxypropylation, which successfully identified 403 S-2-carboxypropylated proteins and 120 cysteine modification sites from HEK293T cells. Through bioinformatic analysis, we found that C2cp-modified proteins were involved in a variety of critica …

Skin necrosis: a rare complication of protein S deficiency.

Hereditary protein S deficiency is an autosomal dominant disorder leading to recurrent venous thrombosis and, less commonly, to arterial thrombosis. Cases of skin necrosis have been documented in patients with protein C or S deficiency while being treated with warfarin. We describe herein a patient with protein S deficiency who developed significant skin necrosis without being exposed to warfarin. She had a protracted clinical course resulting in gangrene and transmetatarsal amputation. Recognition of this rare complication and an earlyhematology referral may prevent dismal outcomes in patient…

Micro-ribonucleic Acid 494 regulation of protein S expression.

BACKGROUND: Acquired protein S (PS) deficiency is highly associated with elevated circulating estrogen levels resulting from pregnancy, oral contraceptives, and estrogen replacement therapy; however, the mechanism of estrogen-mediated acquired PS deficiency remains poorly understood. Increasing evidence indicates that estrogen receptor signaling can indirectly modulate the expression of target genes at the post-transcriptional level by modulating the expression of microRNAs (miRNAs), and miRNAs have also been demonstrated to be involved in the regulation of hemostasis. OBJECTIVES: To investig

Anticoagulant treatment with rivaroxaban in severe protein S deficiency.

We report a case of a 6-year-old girl with severe protein S deficiency due to a homozygous mutation and recurrent episodes of skin necrosis. She developed purpura fulminans at birth and a catheter-related venous thrombosis complicated by massive pulmonary embolism at the sixth day of life. Long-term oral anticoagulant therapy with a vitamin K-antagonist was started with a therapeutic range of the international normalized ratio of prothrombin time between 2.0 and 3.0. Unfortunately, this common range was not sufficient because recurrent episodes of warfarin-induced skin necrosis developed if th…

17beta-Estradiol induces protein S-nitrosylation in the endothelium.

AIMS: Estrogen induces nitric oxide (NO) in the endothelium and appears to protect against inflammation and atherosclerosis. NO can induce post-translational protein modifications such as cysteine S-nitrosylation in the cellular proteins which may exert anti-inflammatory effects. However, whether estrogen can induce protein S-nitrosylation in the endothelium is not known. Given this background, we investigated the role of 17beta-estradiol (E2beta), the major form of estrogen in the body, on endothelial protein S-nitrosylation. METHODS AND RESULTS: Experiments were performed in human umbilical…

Moyamoya disease and protein S deficiency: a case report.

A 2-year-old child who acutely developed hemiplegia and seizure was found to have moyamoya disease and heterozygous protein S deficiency. This case report should alert physicians to the possible coexistence of moyamoya disease and protein S deficiency, even in the case of typical moyamoya disease. The intimate relationship between the two require further study.

[Ischemic infarct, mitral valve prolapse and protein S deficiency].

Ischaemic stroke in young people, that is in those under 45 years of age, forms a distinct entity. Whereas in the case of older patients the main cause is arteriosclerosis, in the younger group a broad spectrum of etiologies can be found. It is for this reason that in these latter cases we must carry out a diagnostic study that is not only more exhaustive but also additionally is aimed at seeking out specific pathologies as for instance a possible association with vasculitis, infectious diseases, hematological abnormalities or unclear cardiopathology. Given the therapeutic transcendency that f
