BACKGROUND & AIM: 25-hydroxyvitamin D (25OHD) level, the vitamin D status biomarker, is influenced by genetic and environmental factors, most notably, sun exposure. Additionally, skin pigmentation modifies the amount of ultraviolet radiation that penetrates the skin to initiate vitamin D synthesis. The aim of this study was to identify genetic loci associated with 25OHD among African and Asian ancestry individuals, adjusting for its key environmental determinant, ambient ultraviolet radiation B (UVB).
METHODS: A cumulative and weighted UVB dose was estimated for each participant based on thei

The rate of genome damage elimination after therapeutic exposure to ionising radiation was estimated in stage I testicular seminoma patients monitored over a seven-year follow-up. DNA damage elimination in peripheral lymphocytes of ten subjects was analysed by the chromosome aberration assay. Seven years after the end of radiotherapy, significantly increased frequency of ring and dicentric chromosomes was still detected in comparison with baseline values. These results indicate the induction of genome instability. Long-term follow-up studies of cancer patients after radiotherapy could give us

The induction of genomic instability in TK6 human lymphoblasts by exposure to (137)Cs gamma radiation was investigated by measuring the frequency and characteristics of unstable clones isolated approximately 36 generations after exposure. Clones surviving irradiation and control clones were analyzed for 17 characteristics including chromosomal aberrations, growth defects, alterations in response to a second irradiation, and mutant frequencies at the thymidine kinase and Na(+)/K(+) ATPase loci. Putative unstable clones were defined as those that exhibited a significant alteration in one or more

Ionizing radiation directly and indirectly affects gene expression within the plant genome. To access the antioxidant response of rice to different types of ionizing radiation, rice seeds were exposed to gamma-ray, cosmic-ray and ion beam radiation. Exposure to ionizing radiation dramatically decreased the shoot length in all plants but not the root length compared with a non-irradiated plant. Electron spin resonance, confirmed that the number of free radicals in cell was greatly increased by different types of ionizing radiation. The measurement of the MDA, chlorophyll, carotenoids contents a

Adrenocortical carcinoma (ACC) is a rare malignancy with limited therapeutic options following the failure of standard adjuvant therapies. We present a 54-year-old female with stage IV cortisol-secreting ACC that progressed despite radical adrenalectomy, adjuvant radiation, and mitotane therapy. Comprehensive genomic profiling revealed a complex molecular landscape: a "double hit" driving hypercortisolism (GNAS activation and HSD11B2 alteration) and a high tumor mutational burden (11.6 mutations/megabase) driven by converging instability mechanisms (MSH6, TP53, DAXX, DNMT3A, and KMT2A somatic

Epidemiological studies have revealed a radiation-related increase in the risk of developing acute lymphoblastic leukemia (ALL). Our recent study revealed early induction and increased risk of precursor B-cell (pB) lymphomas in mice after radiation exposure. However, the genomic landscape of radiation-induced B-cell lymphomas remains unclear. To identify the relevant genetic alterations in mice, whole-exome sequencing was performed on both early-onset and late-onset B-cell lymphomas that developed spontaneously or after gamma-irradiation. In addition to multiple driver mutations, the data reve

This study was aimed to evaluate the protection against radiation of human peripheral blood lymphocytic DNA by a formulation of three isolated active principles of Podophyllum hexandrum (G-002M). G-002M in various concentrations was administered 1h prior to irradiation in culture media containing blood. Radioprotective efficacy of G-002M to lymphocytic DNA was estimated using various parameters such as dicentrics, micronuclei (MN), nucleoplasmic bridges (NPB) and nuclear buds (NuB) in binucleated cells. Certain experiments to ascertain the G2/M arrest potential of G-002M were also conducted. I

The impact of environmental mutagens and carcinogens on the mammary gland has recently received a lot of attention. Among the most generally accepted carcinogenic agents identified as factors that may increase breast cancer incidence are ionizing radiation and elevated estrogen levels. However, the molecular mechanisms of mammary gland aberrations associated with radiation and estrogen exposure still need to be further elucidated, especially the interplay between elevated hormone levels and radiation. Therefore, in the present study, we investigated molecular changes induced in rat mammary gla

The results of series investigations of late effects after Chernobyl accident are discussed. Genomic instability induced by chronic irradiation of cultural cells in Chernobyl zone and in laboratory conditions have been studied. It was shown that low level prolonged irradiation result in increase of frequency of cells with micronuclei, giant cells, enhancement of radiosensitivity in descendents of early irradiated cells. Chronic low doses irradiation doesn't induce the adaptive response. Comparative investigation of adaptive response in blood lymphocytes of people (adults and children) living i

INTRODUCTION: Ionizing radiation (IR) is a well-established mutagen capable of inducing structural genomic alterations. The 1987 Goiânia radiological accident involving Caesium-137 represents one of the most significant cases of accidental human exposure to IR outside nuclear facilities.
OBJECTIVE: This study aimed to investigate the occurrence of somatic copy number variations (CNVs) as potential biomarkers of IR exposure in individuals accidentally exposed to Cesium-137.
METHODS: A case-control study was conducted including 20 exposed and 25 non-exposed individuals. Chromosomal microarray

BACKGROUND: The DNA damage response (DDR) is a mechanism that protects cells against radiation-induced oxidative DNA damage by causing cell cycle arrest and apoptosis. TP63 is a member of the tumour suppressor TP53 gene family, and ΔNp63α, a TP63 splicing variant, is constitutively expressed in the stem cell-containing basal layer of stratified epithelial tissues, including the mammary gland, where it plays a critical role in stemness and tissue development. ΔNp63α has been reported to transcriptionally inhibit the tumour suppression protein p53. This p53-repressive activity may cause genomic

Adenosine (A) to inosine (I) RNA editing is the most prevalent RNA editing mechanism in humans and plays critical roles in tumorigenesis. However, the effects of radiation on RNA editing were poorly understood, and a deeper understanding of the radiation-induced cancer is imperative. Here, we analyzed BEP2D (a human bronchial epithelial cell line) and radiation-induced malignantly transformed cell lines with next generation sequencing. By performing an integrated analysis of A-to-I RNA editing, we found that single-nucleotide variants (SNVs) might induce the downregulation of ADAR2 enzymes, an

BACKGROUND: Soft tissue sarcomas (STS), have significant inter- and intra-tumoral heterogeneity, with poor response to standard neoadjuvant radiotherapy (RT). Achieving a favorable pathologic response (FPR ≥ 95%) from RT is associated with improved patient outcome. Genomic adjusted radiation dose (GARD), a radiation-specific metric that quantifies the expected RT treatment effect as a function of tumor dose and genomics, proposed that STS is significantly underdosed. STS have significant radiomic heterogeneity, where radiomic habitats can delineate regions of intra-tumoral hypoxia and radiores

Epidemiological studies have revealed that ionizing radiation is a risk factor for acute lymphoblastic leukemia. Humans can be exposed to radiation via clinical radiotherapies or spaceflight, yet our knowledge of the potential carcinogenic effects of various types of radiation remains incomplete. To address this shortcoming, we analyzed the development of precursor B-cell lymphoma (pBL) in B6C3F1 mice after irradiation with gamma rays or heavy ions (carbon, silicon, argon, or iron ions) followed by array comparative genomic hybridization, whole-exome sequencing, and RNA-sequencing analyses. In this paper I track the history of post-1945 human genetics and genomics emphasizing the importance of ideas about risk to the scientific study and medical management of human heredity. Drawing on my own scholarship as it is refracted through important new work by other scholars both junior and senior, I explore how radiation risk and then later disease risk mattered to the development of genetics and genomics, particularly in the United States. In this context I excavate one of the central ironies of post-war human genetics: while studies of DNA as the origin and cause of diseases have been

There is a concern over the potential use of radioactive isotopes as a weapon of terror. The detonation of a radiation dispersal device, the so-called "dirty bomb" can lead to public panic. In order to estimate risks associated with radiation exposure, it is important to understand the biological effects of radiation exposure. Based on this knowledge, biomarkers to monitor potentially exposed populations after a radiological accident can be developed and would be extremely valuable for emergency response. While the traditional radiation exposure biomarkers based on cytogenetic assays serve as

Genomic architecture is a key factor shaping the landscape of linkage disequilibrium between divergently selected traits. By facilitating the detection of intrachromosomal variants, long-read data fueled recent interest in the role of structural variants in adaptive radiation. The Neotropical cichlid genus Crenicichla provides a compelling system for replicated adaptive radiation in riverine environments in which at least two lineages show repeated ecomorphological evolution. The lack of South American cichlid reference genomes has thus far limited investigation of chromosomal evolution across

Exposure to ionizing radiation is a well-known risk factor for a number of human cancers, including leukemia and thyroid cancer. It has been known for a long time that exposure of cells to radiation results in extensive DNA damage; however, a small number of studies have tried to explain the mechanisms of radiation-induced carcinogenesis. The high prevalence of RET/PTC rearrangements in patients who have received external radiation, and the evidence of in vitro induction of RET rearrangements in human cells, suggest an enhanced sensitivity of the RET genomic region to damage by ionizing radiat

OBJECTIVE: Cancer survivorship has thrown the spotlight on the incidence of nonmalignant chronic diseases in cancer patients. Endothelial injury is increasingly recognized as a consequence of cancer treatment, particularly after radiation therapy (RT). This review is to provide a current understanding on the pathophysiological mechanisms and predictive biomarkers of radiation-induced vascular injury.
RECENT FINDINGS: Radiation directly impacts vasculature by causing endothelial apoptosis and senescence, and alterations in normal homeostasis. This altered milieu at the endothelial surface may

Histopathology and clinical staging have historically formed the backbone for allocation of treatment decisions in oncology. Although this has provided an extremely practical and fruitful approach for decades, it has long been evident that these data alone do not adequately capture the heterogeneity and breadth of disease trajectories experienced by patients. As efficient and affordable DNA and RNA sequencing have become available, the ability to provide precision therapy has become within grasp. This has been realized with systemic oncologic therapy, as targeted therapies have demonstrated im
