The effects of risperidone on the acquisition of schedule-induced polydipsia in rats were investigated in a chronic dose regime followed by seven days of withdrawal. Risperidone dose-dependently suppressed water intake and number of panel pushes. Drinking efficiency and free water intake in home cages were unchanged. By comparing the effects of risperidone in the present paper with the effects of different-neuroleptics in the same procedure from an earlier study, it appears that the effect of risperidone is intermediary to that of the 'typical' and 'atypical' neuroleptics. Risperidone is known to increase prolactin secretion in treating mental illness patients. This side-effect is thought to be mediated via central signaling pathway. A 48-year-old woman suffering from treatment-resistant psychogenic stiff neck and severe depression had been treated with a varied regimen of medications, including antidepressants, neuroleptics, and antiepileptics. These treatments did not result in improvement. Therefore, she was treated with a combination of sertraline and risperidone. Depressive symptoms decreased within a few weeks of treatment initiation. The atypical antipsychotic risperidone combines dopaminergic and serotonergic antagonism. This results in a drug that is both clinically effective, reducing positive and negative symptoms of schizophrenia, and has a low incidence of adverse effects.
