Isolation of Shewanella algae from rectal swabs of patients with bloody diarrhoea.

Shewanella algae is an emerging bacteria rarely implicated as a human pathogen. It was infrequently recovered from clinical specimens probably because of inadequate processing of non-fermenting oxidase-positive gram-negative bacilli. We report here isolation of S. algae in pure culture and mixed with E. coli from two cases of acute gastroenteritis with bloody mucous containing diarrhea occurring at the same time. As this organism is not a normal flora of the gut, the possible source of infection may be fish contaminated with the organism. Whether this bacterium can be considered an enteric pat…

Epidemiology and clinical features of Shewanella infection over an eight-year period.

Shewanella is a rare human pathogen that can lead to fatal infections. However, clinical information about this bacterium remains scarce. In this study, we retrospectively reviewed all patients with laboratory isolates of Shewanella over an 8-y period to assess risk factors, clinical manifestations and outcome. Twenty-nine patients were identified. Shewanella was most commonly isolated from intra-abdominal specimens (48.2%), followed by skin and soft tissue specimens (27.6%), blood (13.8%) and sputum (10.3%). Malignancy, hepatobiliary disease and diabetes mellitus were common underlying diseas…

Cellulitis due to Shewanella algae: Crucial diagnostic clues from basic microbiological tests.

Shewanella spp. are infrequently implicated in human infections but they are emerging pathogens with particular significance in regions with warm climates, such as Southeast Asia. This is a case of a middle-aged diabetic and hypertensive man who presented with worsening congestive heart failure symptoms associated with fever and a painful right leg. His right leg had numerous scabs and was tender, warm and erythematous. He was provisionally diagnosed with decompensated heart failure precipitated by cellulitis and uncontrolled hypertension. His blood grew non-fermentative, oxidase-positive and …

Expression of blaA underlies unexpected ampicillin-induced cell lysis of Shewanella oneidensis.

Shewanella oneidensis is a facultative anaerobic γ-proteobacterium possessing remarkably diverse respiratory capacities for reducing various organic and inorganic substrates. As a veteran research model for investigating redox transformations of environmental contaminants the bacterium is well known to be a naturally ampicillin-resistant microorganism. However, in this study we discovered that ampicillin has a significant impact on growth of S. oneidensis. Particularly, cell lysis occurred only with ampicillin at levels ranging from 0.49 to 6.25 µg/ml but not at 50 µg/ml. This phenotype is att…

Molecular Underpinnings of Fe(III) Oxide Reduction by Shewanella Oneidensis MR-1.

In the absence of O(2) and other electron acceptors, the Gram-negative bacterium Shewanella oneidensis MR-1 can use ferric [Fe(III)] (oxy)(hydr)oxide minerals as the terminal electron acceptors for anaerobic respiration. At circumneutral pH and in the absence of strong complexing ligands, Fe(III) oxides are relatively insoluble and thus are external to the bacterial cells. S. oneidensis MR-1 and related strains of metal-reducing Shewanella have evolved machinery (i.e., metal-reducing or Mtr pathway) for transferring electrons from the inner-membrane, through the periplasm and across the outer-…

c-Type cytochrome-dependent formation of U(IV) nanoparticles by Shewanella oneidensis.

Modern approaches for bioremediation of radionuclide contaminated environments are based on the ability of microorganisms to effectively catalyze changes in the oxidation states of metals that in turn influence their solubility. Although microbial metal reduction has been identified as an effective means for immobilizing highly-soluble uranium(VI) complexes in situ, the biomolecular mechanisms of U(VI) reduction are not well understood. Here, we show that c-type cytochromes of a dissimilatory metal-reducing bacterium, Shewanella oneidensis MR-1, are essential for the reduction of U(VI) and for…

First case report of Shewanella indica isolated from a hospitalized patient in Serbia.

Over the past decade, Shewanella spp. have been increasingly recognized as opportunistic pathogens, particularly in patients with malignancies, neutropenia, severe heart failure, renal insufficiency, and hepatobiliary diseases. Shewanella indica is a rarely reported species within the Shewanella genus, and its role in human infection remains poorly documented. In this study, we report the first case of S. indica isolated from the stool of a critically ill 72-year-old man in Serbia with multiple pre-existing comorbidities and co-infections. Initial identification using the VITEK 2 system miscla…

Humin accelerates interspecies electron transfer between Shewanella oneidensis MR-1 and Methanosarcina barkeri.

Existing researches involving accelerated interspecies electron transfer (IET) by solid redox mediators focus mainly on the conductive nature of these materials. Although non-conductive solid redox mediator-humin has been reported to promote methanogenic performance of anaerobic granular sludge, likely through accelerating IET of microorganisms, this phenomenon has not been validly proven. In this study, a wetland sediment sourced HM (HMWS) was added into a co-culture of a syntrophic bacteria Shewanella oneidensis MR-1 and an archaeal Methanosarcina barkeri with ethanol as sole electron donor …

Differential degradation of petroleum hydrocarbons by Shewanella putrefaciens under aerobic and anaerobic conditions.

The complexity of crude oil composition, combined with the fluctuating oxygen level in contaminated environments, poses challenges for the bioremediation of oil pollutants, because of compound-specific microbial degradation of petroleum hydrocarbons under certain conditions. As a result, facultative bacteria capable of breaking down petroleum hydrocarbons under both aerobic and anaerobic conditions are presumably effective, however, this hypothesis has not been directly tested. In the current investigation, Shewanella putrefaciens CN32, a facultative anaerobic bacterium, was used to degrade pe…

Upon receiving electrons from Shewanella putrefaciens CN32, BC-AQDS was reduced to BC-AH2DS, which subsequently facilitated the reduction of Cr(VI) to Cr(III). Goethite and riboflavin synergistically enhance Cr(VI) reduction by Shewanella oneidensis MR-1.

Bioreduction of Cr(VI) is cost-effective and environmentally friendly, however, the slow bioreduction rate limits its application. In this study, the potential synergistic enhancement of Cr(VI) bioreduction by shewanella oneidensis MR-1 (S. oneidensis) with goethite and riboflavin (RF) was investigated. The results showed that the S. oneidensis reaction system reduce 29.2% of 20 mg/L Cr(VI) after 42 h reaction, while the S. oneidensis/goethite/RF reaction system increased the Cr(VI) reduction rate to 87.74%. RF as an efficient electron shuttle and Fe(II) from goethite bioreduction were identif…

EDDS enhanced Shewanella putrefaciens CN32 and α-FeOOH reductive dechlorination of carbon tetrachloride.

S,S-ethylenediamine-N,N-disuccinic acid (EDDS) enhanced reductive dissolution of α-FeOOH by Shewanella putrefaciens CN32 (CN32), resulting in formation of surface-bound Fe(II) species (FeIIEDDS) to improve reductive dechlorination of carbon tetrachloride (CT). The pseudo-first-order rate constants for bio-reduction extents of α-FeOOH by CN32 in the presence of 1.36 mM EDDS was 0.023 ± 0.0003 d-1 which was higher than without EDDS. The enhancement mechanism of bio-reduction was attributed to the strong complexation ability of EDDS to formed FeIIIEDDS, which could be better utilized by CN32. The…

Capsular polysaccharide-mediated protein loading onto extracellular membrane vesicles of Shewanella vesiculosa HM13.

Bacterial extracellular membrane vesicles (EMVs) play crucial physiological roles, including intercellular communication and nutrient acquisition, which depend on the functions of their cargo molecules. However, the molecular mechanisms governing selective cargo loading onto EMVs remain poorly understood. Here, we investigated the role of capsular polysaccharide (CPS) in the association of P49, the major cargo protein of EMVs from Shewanella vesiculosa HM13, with EMVs. Deleting genes neighboring the P49 gene led to a loss of EMV-associated CPS and a defect in P49 loading. In vitro binding assa…

Exploiting interspecies interactions in a Corynebacterium-Shewanella co-culture improves succinic acid production.

Anaerobic production of succinic acid in Corynebacterium glutamicum is often constrained by metabolic imbalances and by-product formation. In this study, an engineered co-culture system was developed by pairing C. glutamicum with the electroactive bacterium Shewanella oneidensis to enhance succinic acid biosynthesis through interspecies interactions. The optimized co-culture achieved a succinic acid yield of 0.169 g/g, representing a 45.65% increase compared to monoculture, without markedly altering glucose consumption. Screening of five redox-active compounds identified riboflavin as the most…

Bridging clinical and environmental reservoirs: antimicrobial resistance in the emerging pathogen Shewanella algae.

The emergence of antimicrobial resistance in environmental bacteria threatens therapeutic efficacy in clinical settings. Shewanella algae, historically regarded as a marine saprophyte, is increasingly recognized as an emerging opportunistic pathogen. In this study, we analyzed 86 S. algae isolates from Spain (19 clinical and 67 environmental) and integrated them with 178 publicly available genomes to explore antimicrobial susceptibility patterns and genomic diversity. Penicillins and fosfomycin consistently showed poor activity, whereas piperacillin/tazobactam, third- and fourth-generation cep…

Electricity-powered cryptic CO2 fixation pathway in heterotrophic Shewanella oneidensis for acetate synthesis.

Microbial electrosynthesis of CO2 is a sustainable carbon neutral technology. Although known for its diverse and efficient extracellular electron transfer (EET) characteristics, the bacteria of Shewanella genus have never been reported for use in electrosynthesis of multi-carbon chemicals. Herein, the electricity-powered conversion of CO2 to acetate was achieved under ammonium regulation for the first time in the model strain (Shewanella oneidensis MR-1), due to the activation of its intrinsic reductive glycine pathway. A high electron flux from cathode into MR-1 was achieved through the uniqu…

Shewanella algae Bacteremia and Septic Shock Secondary to Necrotizing Fasciitis: A Case Report.

Shewanella algae (S. algae), a marine Gram-negative bacillus, is an emerging opportunistic pathogen capable of causing infections in humans. While mostly associated with mild skin and soft tissue infections, severe invasive infections, such as bacteremia and septic shock, are rare but increasingly reported. We present a case of fatal S. algae bacteremia and septic shock secondary to necrotizing fasciitis in a patient with significant comorbidities, including recent bariatric surgery, rapid weight loss, and possible malnutrition, highlighting the role of host factors in fulminant disease progre…

Chelating Effect of Siderophore Desferrioxamine-B on Uranyl Biomineralization Mediated by Shewanella putrefaciens.

In contaminated water and soil, little is known about the role and mechanism of the biometabolic molecule siderophore desferrioxamine-B (DFO) in the biogeochemical cycle of uranium due to complicated coordination and reaction networks. Here, a joint experimental and quantum chemical investigation is carried out to probe the biomineralization of uranyl (UO22+, referred to as U(VI) hereafter) induced by Shewanella putrefaciens (abbreviated as S. putrefaciens) in the presence of DFO and Fe3+ ion. The results show that the production of mineralized solids {hydrogen-uranium mica [H2(UO2)2(PO4)2·8H2…

Detection of S83V GyrA mutation in quinolone-resistant Shewanella algae using comparative genomics.

BACKGROUND: Shewanella algae is a zoonotic pathogen that poses a serious health threat to immunocompromised hosts. Treatment of S. algae infections is challenging due to the pathogen's intrinsic resistance to a variety of β-lactam antibiotics. Therapeutic options have become further limited by the emergence of quinolone-resistant strains. Currently, there are few studies concerning the genetic and molecular mechanisms underlying acquired quinolones resistance in S. algae. qnrA was once proposed as the candidate gene related to quinolones resistance in S. algae. However, recent studies demonstr…
