The Emerging Role of Sialic Acids in Obesity and Diabetes: Molecular Mechanisms and Therapeutic Perspectives.

Sialic acids are terminal monosaccharides that cap glycans on glycoconjugates. Accumulating clinical and experimental evidence shows that obesity, insulin resistance, and diabetes are accompanied by changes in sialic-acid levels. In these conditions, the sialic-acid axis is also broadly remodeled: writers (sialyltransferases), erasers (neuraminidases), and readers (Siglecs) are dysregulated across adipose tissue, liver, pancreas, endothelium, and blood, shifting insulin signaling and inflammatory tone. This review summarizes relevant studies from the perspectives of disease clinical indicators…

Expediting chemoenzymatic access to bacterial 'sialic acid-like' sugars via mapping of wild type sialic acid aldolase enzyme substrate tolerance and promiscuity.

Bacterial 'sialic-acid-like' sugars, including legionaminic acid and pseudaminic acid, are critical for the virulence and immune evasion of numerous human pathogens, including pan-drug resistant Acinetobacter baumannii and global gut pathogens Campylobacter jejuni and Helicobacter pylori. Despite this importance, a detailed understanding of the role these sugars play in pathogenesis is hampered by the challenging synthesis of their 9-deoxy nonulosonic acid backbones. Herein, we expedite access to these complex glyco-architectures through exploration of the biocatalytic potential of sialic acid

Sialic Acids and Tetraspanins: An Insight into their Potential as Indicators of Human Sperm Quality.

Since routine sperm analysis cannot detect molecular-level defects, efforts continue to find additional markers for selecting the "highest quality" sperm. This study analyzes ejaculates from normozoospermic and non-normozoospermic men, including teratozoospermics (T), asthenoteratozoospermics (AT), and oligoasthenoteratozoospermics (OAT), by two approaches. The first is distribution of sialic acid residues, molecules critical for sperm migration and survival in female reproductive tract, using lectins with distinct binding specificities (SNA I, MAL I, WGA). The second is the localization of me

Interventional Effects of Edible Bird's Nest or Sialic Acids on Sepsis-Induced Immunosuppression in Mice.

Edible bird's nest (EBN) and its key component, sialic acid (SA), were investigated for their potential to counteract sepsis-induced immunosuppression. In a murine "two-hit" sepsis model, EBN and SA significantly enhanced survival, ameliorated multi-organ damage, and reduced bacterial dissemination. Crucially, they reversed immunosuppression by restoring the TNF-α-secreting capacity of immune cells. Proteomic and KEGG analyses identified T cell proliferation as a central mechanism, which was validated by CFSE assay. Furthermore, we demonstrated that this enhanced T cell proliferation is mediat

Diagnostic and prognostic potential of antibodies against O-acetylated sialic acids in canine visceral leishmaniasis.

Employing bovine submaxillary mucin (BSM) as the coating agent, an enzyme-linked immunosorbent assay (BSM-ELISA) was developed to detect antibodies directed against O-acetylated sialic acids (O-AcSA) in canine visceral leishmaniasis (CVL). Serum samples were collected from 50 dogs previously screened by a parasite-ELISA to detect anti-leishmanial antibodies and designated as seropositive (n = 30) and seronegative (n = 20). The BSM-ELISA detected anti-O-AcSA antibodies in 29 out of 30 seropositive dogs and was negative in 15 out of 20 seronegative dogs; the sensitivity and specificity of the as…

Distribution of mono-, di, and tri-O-acetylated sialic acids in normal and neoplastic colon.

The purpose of this study was to compare the expression of O-acetylated sialic acids on normal colonic epithelial cells to that on primary and metastatic human adenocarcinoma of the colon and rectum. In 24 cases, the relative percentages of biosynthetically labeled non-, mono-, di-, and tri-O-acetylated sialic acids were measured after hydrolytic release, separation, and identification by paper chromatography. In one case, the presence of di- and tri-O-acetylated sialic acids was confirmed by fast atom bombardment-mass spectral analysis. Differences were observed in the expression of sialic ac…

The effect of enzymatic removal of sialic acids on the functional properties of protein C.

The effect of enzymatic removal of sialic acid residues on the functional properties of human protein C was examined. The rates of activation of native and asialo protein C were identical using either the thrombin thrombomodulin complex or the snake venom activator, protac. Desiaylated activated protein C (APC) was, two-three-fold more active than native protein C, both as an anticoagulant and in its ability to hydrolyze chromogenic substrates. The potentiating effect of the desialylation was confirmed by initially fully activating protein C followed by the desialylation process. In these stud

Sialoglycoproteins and sialic acids of Plasmodium knowlesi schizont-infected erythrocytes and normal rhesus monkey erythrocytes.

The effects of malaria infection on RBC sialic acids and sialoglycoproteins were studied with asexual blood-stage infections of Plasmodium knowlesi in rhesus monkeys. Glycoprotein radio-isotope labelling methods were used to compare the sialoglycoproteins of normal RBC and P. knowlesi schizont-infected RBC (SI-RBC). Tritiation of glycoproteins from SI-RBC with the standard sialidase + galactose oxidase/NaB3H4 method or standard periodate/NaB3H4 method was significantly decreased when compared to normal RBC. However, tritium uptake into glycoproteins was normal when SI-RBC were treated with 5-f…

Sialic acid-guided spatiotemporal hydrogel therapy for liver cancer.

Efficient delivery of plant-derived polyphenolic drugs to tumor sites in hepatocellular carcinoma (HCC) is challenging due to their rapid metabolism and the limited tumor-targeting capacity of current therapeutic strategies. To overcome these limitations, we developed a pH-responsive hydrogel-based drug delivery system (PA-CB) composed of a chitosan backbone functionalized with boronobenzoic acid (CB) and crosslinked with protocatechualdehyde (PA). Within this scaffold, protocatechuic acid (PCA) was incorporated as a model therapeutic agent to demonstrate the platform's ability to achieve cont

Linker-free Functionalization of Phosphorene Nanosheets by Sialic Acid Biomolecules.

The importance of sialic acid on cell functions has been recently unveiled, and consequently, great attention has been paid to its interaction with tumor cells. In this line of research, we have realized phosphorene nanosheets functionalized with sialic acid molecules for biological applications with no need for another linker molecule. The formation of phosphorene sheets is feasible by using hydrogen plasma treatment and conversion of amorphous phosphorus on silicon substrates into highly crystalline nanosheets. Through immersion of these freshly prepared nanosheets into an aqueous solution c

Advances in the development of entry inhibitors for sialic-acid-targeting viruses.

Over the past decades, several antiviral drugs have been developed to treat a range of infections. Yet the number of treatable viral infections is still limited, and resistance to current drug regimens is an ever-growing problem. Therefore, additional strategies are needed to provide a rapid cure for infected individuals. An interesting target for antiviral drugs is the process of viral attachment and entry into the cell. Although most viruses use distinct host receptors for attachment to the target cell, some viruses share receptors, of which sialic acids are a common example. This review aim…

New Therapeutic Strategies for Osteoarthritis by Targeting Sialic Acid Receptors.

Osteoarthritis (OA) is the most common degenerative joint disease characterized by articular cartilage degradation and joint degeneration. The articular cartilage is mainly formed by chondrocytes and a collagen-proteoglycan extracellular matrix that contains high levels of glycosylated proteins. It was reported that the shift from glycoproteins containing α-2,6-linked sialic acids to those that contain α-2,3 was associated with the onset of common types of arthritis. However, the pathophysiology of α-2,3-sialylation in cartilage has not been yet elucidated. We show that cartilage from osteoart…

Investigation of the Potential Use of Sialic Acid as a Biomarker for Rheumatoid Arthritis.

BACKGROUND: Sialic acids are a diverse family of sugar units attached to the outermost ends of sugar chains, which were demonstrated to be related to many diseases. We hypothesized sialic acids could be used as biomarkers for diagnosis of rheumatoid arthritis (RA). METHODS: The serum level of the main sialic acids, N-acetyl-D-neuraminic acid (Neu5Ac) in 163 RA patients and 50 healthy individuals were detected by pre-column derivatization-high performance liquid chromatography method. And three biomarkers to diagnose RA in clinic, C-reactive protein (CRP), rheumatoid factor (RF), and cyclic ci…

The concentration of total sialic acid in chronic hepatitis B and C.

BACKGROUND: The synthesis and glycosylation of glycoproteins and glycolipids take place in the liver. Thus, liver diseases may affect serum concentrations of some carbohydrate derivatives, especially the concentration of sialic acid which is attached to the end of oligosaccharide chains. The aim of this study was to measure and compare the serum concentration of total sialic acid in chronic hepatitis B and C. The hypothesis is that both viruses responsible for the development of inflammation work differently at the cellular level. METHODS: Serum samples were obtained from 90 patients sufferin

Autophagy regulates the stability of sialin, a lysosomal sialic acid transporter.

Macroautophagy plays a critical role in catabolizing cytosolic components via lysosomal degradation. Recent findings from our studies indicate that basal autophagy is required for the efficient lysosomal catabolism of sialyloligosaccharides, and that the downregulation of sialin, a lysosomal transporter of sialic acids can cause a significant delay in the cytosolic accumulation of such glycans. The findings reported herein show that the sialin protein level was increased when the autophagy process was inhibited. This effect appears to be specific to sialin, since the amount of LAMP1, another l…

Evaluation of sialic acid-analogs for the attenuation of amyloid-beta toxicity.

BACKGROUND: Amyloid-beta peptide (Aβ) is the main constituent of senile plaques and is implicated in the pathogenesis of Alzheimer's disease (AD). To that end, agents which either sequester Aβ or interfere with Aβ interaction/binding to cells have been investigated as a means to reduce the pathological effects of Aβ. METHODS: Different structural analogs of sialic acid (N-acetylneuramic acid) were used to decorate a chitosan backbone using EDC chemistry. FTIR and colorimetric assays were used to characterize the complexes. The ability of these complexes to attenuate Aβ toxicity was investigat

Sialic acid uptake is necessary for virulence of Pasteurella multocida in turkeys.

Many pathogenic bacteria employ systems to incorporate sialic acid into their membranes as a means of protection against host defense mechanisms. In Pasteurella multocida, an opportunistic pathogen which causes diseases of economic importance in a wide range of animal species, sialic acid uptake plays a role in a mouse model of systemic pasteurellosis. To further investigate the importance of sialic acid uptake in pathogenesis, sialic acid uptake mutants of an avian strain of P. multocida P-1059 (A:3) were constructed, characterized, and an in-frame sialic acid uptake deletion mutant was asses
