3D+t human sperm flagellum centerline dataset.

The study of human sperm motility has been a topic of interest for decades due to its crucial role in fertility and reproductive health. While most analyses rely on 2D+t imaging of head trajectories, sperm naturally swim in three dimensions (3D), driven by complex flagellar motion. However, the lack of comprehensive 3D+t datasets has limited progress in this field. To address this, we present 3D-SpermFlagella, the first large-scale 3D+t dataset of human sperm flagellum centerline annotations. This dataset contains 135 tracked and annotated sperm, derived from our previously published multifoca

The Genetic Defects of N-DRC in Male Infertility.

The nexin-dynein regulatory complex (N-DRC) is a large protein complex composed of at least 11 subunits (DRC1-DRC11) and plays a crucial role in ciliary and flagellar motility. It links adjacent doublets of microtubules (DMTs) between A and B microtubules, regulating dynein motor activity. Genetic defects in N-DRC subunits lead to primary ciliary dyskinesia (PCD) and abnormal flagellar motility. In recent years, an increasing number of genetic mutations in N-DRC subunits have been reported, associated with male infertility, characterized by multiple morphological abnormalities of the flagella

A novel epididymal quiescence factor inhibits sperm motility by modulating NOS activity and intracellular NO-cGMP pathway.

Mature and potentially motile spermatozoa stored in cauda epididymis in an inactive state for approximately 30 days; however, during ejaculation they regain motility. To understand the actual molecular mechanism of the sperm quiescence during caudal stay, a proteinaceous quiescence factor (QF) has been purified from caprine epididymal plasma to apparent homogeneity. In the present study complete purification, detailed characterization as well as mechanistic pathway of QF has been described. QF is purified to 215-fold with 45% activity recovery. It is a 59 kDa monomeric protein with isoelectric

Signaling mechanisms in mammalian sperm motility.

The goal of sperm is to fertilize the oocyte. To achieve that purpose, it must acquire motility in the epididymis and hyperactivated motility in the female reproductive tract. Motility is only achieved when the sperm presents a fully functional flagellum, is capable of producing energy to fuel the movement, and suffers epididymal maturation and capacitation. Since sperm is a transcriptionally silent cell, motility depends on the activation and/or inhibitions of key signaling pathways. This review describes and discusses the main signaling pathways involved in primary and hyperactivated motilit

Eggs regulate sperm flagellar motility initiation, chemotaxis and inhibition in the coral Acropora digitifera, A. gemmifera and A. tenuis.

Corals perform simultaneous mass spawning around the full moon. Most Acropora species release gamete bundles, which are a complex of eggs and sperm, into the seawater. Then, gamete bundles are separated into eggs and sperm. Eggs are fertilized when sperm and eggs come in contact with each other. However, it is still unclear how sperm meet the eggs of the same species in the presence of many eggs of different species and how eggs guard against the fertilization attempts by sperm of different species. In this study, we observed that A. digitifera, A. gemmifera and A. tenuis sperm showed motility

The origin and function of the posterior ring in mouse sperm head.

BACKGROUND: Despite extensive characterization of sperm structures, certain specialized subcellular structures are still not well understood. Among these, the posterior ring has been recognized for over half a century; yet, its molecular composition and biological role remain unknown. This research aims to define the molecular components and functional roles of the sperm posterior ring. METHODS: 3D-rendered confocal microscopy was used to determine the localization and developmental dynamics of SPEM3 and TEX50 in mature spermatozoa and spermatids at different steps of spermiogenesis. Aquaporin3 is a sperm water channel essential for postcopulatory sperm osmoadaptation and migration.

In the journey from the male to female reproductive tract, mammalian sperm experience a natural osmotic decrease (e.g., in mouse, from ~415 mOsm in the cauda epididymis to ~310 mOsm in the uterine cavity). Sperm have evolved to utilize this hypotonic exposure for motility activation, meanwhile efficiently silence the negative impact of hypotonic cell swelling. Previous physiological and pharmacological studies have shown that ion channel-controlled water influx/efflux is actively involved in the process of sperm volume regulation; however, no specific sperm proteins have been found responsible

Supplementation of quercetin for advanced DNA integrity in bull semen cryopreservation.

The aim of this study was to identify the effects of adding quercetin (Q) to Tris extender in order to identify levels of oxidative stress in bull sperm after freeze thawing. Ejaculates were collected via artificial vagina from Holstein bulls. Semen was divided into five tools and diluted to a final concentration of 15 × 106 spermatozoa/ml with the Tris extender containing Q (25, 50, 100 and 200 μg/ml) and no additive (control; C). All examples were equilibrated at 4°C during 4 hr then were loaded into 0.25-ml straws and frozen using a controlled rate. Sperm motility and motility characteristi

Importance of sperm morphology during sperm transport and fertilization in mammals.

After natural or artificial insemination, the spermatozoon starts a journey from the site of deposition to the place of fertilization. However, only a small subset of the spermatozoa deposited achieves their goal: to reach and fertilize the egg. Factors involved in controlling sperm transport and fertilization include the female reproductive tract environment, cell-cell interactions, gene expression, and phenotypic sperm traits. Some of the significant determinants of fertilization are known (i.e., motility or DNA status), but many sperm traits are still indecipherable. One example is the infl

The ultrastructural characteristics of the spermatozoa stored in the cauda epididymidis in Chinese soft-shelled turtle Pelodiscus sinensis during the breeding season.

The ultrastructure of spermatozoa in cauda epididymidis of soft-shelled turtle, P. sinensis during breeding season was investigated by light microscopy (LM) and electron microscopy (TEM and SEM). The mature spermatozoa appeared elongated and filiform. In general, the turtle spermatozoon contains a characteristic head, midpiece and tail, similar in morphology to that of birds, amphibians and other reptiles. However, several features are unique. These include (1) three intranuclear tubules containing dense core extend from the subacrosomal cone through the rostral nucleus and deep into the nucle

The cause of infertility of male c-ros tyrosine kinase receptor knockout mice.

Male homozygous transgenic c-ros knockout mice are sterile by natural mating, lack a part of their epididymis, and the epididymal sperm exhibit tail angulation in vivo and in vitro. To ascertain if this abnormal tail form caused the infertility, the number and nature of sperm in the tract of females mated to knockout and wild-type mice were determined. Percentage motility and numbers of sperm in the uterus 1 h after mating were similar between genotypes. The majority of the uterine sperm from the wild-type males had straight flagella, whereas 46-86% of knockout sperm were bent at the cytoplasm

[Outer dense fiber 2 and sperm function: Progress in studies].

Oligoasthenozoospermia, teratozoospermia or low sperm motility is the main cause of male infertility. Low sperm motility can be induced by abnormalities of the sperm tail structure and sperm function. The outer dense fiber protein 2 (ODF2) is a protein fiber maintaining cytoskeleton, as a major component of the mammalian sperm tail and centrosome, and its abnormality is closely related to asthenospermia. Recent studies indicate that ODF2 includes many proteins of the same name and homologous splices located in the sperm centrosomes and spindles of cleaved-embryos, necessary for animal ciliogen…

How to increase the motility of spermatozoa from the epididymis of bulls and alloplastic spermatoceles in minipigs.

The motility of spermatozoa from the head and tail of the epididymis in bulls was studied. Qualitatively and quantitatively, the motility of spermatozoa from the cauda was distinctly better than that from the caput. It was possible to achieve a highly significant increase in the motility of epididymal spermatozoa from the caput as well as the cauda area using caffeine or a caffeine-kallikrein mixture. Above all, motility stimulants improved the local motility of the epididymal spermatozoa as compared to twitching and progressive motility. The motility of caudal spermatozoa was increased by 100

Deep-Learning-Based Profiling of Mouse Sperm Head and Neck Morphology Reveals Coordinated Structural Variation.

BACKGROUND: Mammalian sperm head morphology varies across species and dictates fertilization capability. Rodent sperm have asymmetric, hook-shaped heads, complicating analysis. Despite widespread biomedical use, current tools poorly characterize these complex variations. OBJECTIVES: This study aimed to establish a robust framework for the precise, noninvasive, and quantitative analysis of mouse sperm morphology. MATERIALS AND METHODS: We developed ADAM-net (Anomaly-aware Deep-learning Architecture for Morphology), a machine-learning system for mouse sperm head and neck structures. Integratin

Reorganization of the flagellum scaffolding induces a sperm standstill during fertilization.

Mammalian sperm delve into the female reproductive tract to fertilize the female gamete. The available information about how sperm regulate their motility during the final journey to the fertilization site is extremely limited. In this work, we investigated the structural and functional changes in the sperm flagellum after acrosomal exocytosis (AE) and during the interaction with the eggs. The evidence demonstrates that the double helix actin network surrounding the mitochondrial sheath of the midpiece undergoes structural changes prior to the motility cessation. This structural modification i

Dissecting the signaling pathways involved in the function of sperm flagellum.

The mammalian flagellum is a specific type of motile cilium required for sperm motility and male fertility. Effective flagellar movement is dependent on axonemal function, which in turn relies on proper ion homeostasis within the flagellar compartment. This ion homeostasis is maintained by the concerted function of ion channels and transporters that initiate signal transduction pathways resulting in motility changes. Advances in electrophysiology and super-resolution microscopy have helped to identify and characterize new regulatory modalities of the mammalian flagellum. Here, we discuss what
