BACKGROUND: A homozygous mutation of the endothelin B receptor (EDNRB) gene in spotting lethal (sl/sl) rats leads to Hirschsprung's disease … r of nerve cells per ganglia, and the diameter of nerve fibre strands within both the myenteric and submucous plexus.
RESULTS: Sl/sl rats were characterised by complete aganglionosis resembling the same histopathological features observed in patients with HSCR. +/sl rats revealed more subtle abnormalities of the ENS: the submucous plexus was characterised by a si …

Detailed investigations were made of the microvasculature of the mucous membrane, especially the transverse palatine plicae, of the hard palate of the Japanese monkey (Macaca fuscata) utilizing the plastic injection method. The microvascular patterns obtained were compared with those of the cat and other mammals. The incisive papilla was located at the anterior end of the median line of the hard palate, and seven or eight transverse palatine plicae were observed symmetrically from this papilla posteriorly at similar intervals. Each plica arched bilaterally in a wide M. The heights of the plica

Hypogenesis of intestinal ganglion cells is a rare cause of functional intestinal obstruction showing both diminished numbers of ganglion cells per plexus and immature ganglion cells. We report a case of hypogenesis extending from anus to mid small intestine.

A series of 93 rectal biopsies performed for diagnosis of suspected progressive neurometabolic disease between 1967 and 1973 is reviewed, and the results of this and of two previously published series totalling 307 biopsies are analysed. In GM1 and GM2 gangliosidosis, Batten's disease, and certain other neuronal storage diseases rectal biopsy is a reliable diagnostic alternative to brain biopsy. However, the need for biopsy has diminished with improvement in other diagnostic methods, particularly enzyme assay, the availability of which should determine the extent to which biopsy is used. It is

On a review of 15 cases of Hirschsprung's disease of the colon we have encountered 4 cases of an atypical variant. These 4 cases had a zone of aganglionosis of both intermyenteric and submucous plexuses in the sigmoid colon and, in 1 case, a further zone of aganglionosis in the rectum. Ganglion cells reappeared distal to the sigmoid zone of aganglionosis. A transition zone was not invariably present. The clinical features demonstrated a variability of presentation typical of the more usual form of the disease. The importance of atypical Hirschsprung's disease is discussed from a diagnostic and…

Intestinal neuronal dysplasia (IND) is a malformation of the enteric plexus and clinically resembles Hirschsprung's disease. The pathogenesis of IND is not known. There is no information available regarding the nature of the neuromuscular junction in this disease. The authors examined immunohistochemically full-thickness biopsy specimens from 14 patients (aged 3 weeks to 26 months) with IND and 10 age-matched controls, using monoclonal antibodies to growth associated protein-43 (GAP-43), synaptophysin, and neural-cell adhesion molecule (NCAM) as neuromuscular junction markers. In the normal bo

Autopsies of 12 consecutively born infants with anencephaly showed varying degrees of aganglionosis and lateralization defects in four of them. This seemingly regular occurrence of these three defects together suggests that they are caused by an aberration of blastogenesis that results in a polytopic field defect.

We investigated the distributions and targets of nitrergic neurons in the rat stomach, using neuronal nitric oxide synthase (NOS) immunohistochemistry and nicotinamide adenine dinucleotide phosphate (NADPH) diaphorase histochemistry. Nitrergic neurons comprised similar proportions of myenteric neurons, about 30%, in all gastric regions. Small numbers of nitrergic neurons occurred in submucosal ganglia. In total, there were ~ 125,000 neuronal nitric oxide synthase (nNOS) neurons in the stomach. The myenteric cell bodies had single axons, type I morphology and a wide range of sizes. Five targets

This study reports on changes in the somatostatin-like immunoreactive (SOM-LI) nerve structures of the enteric nervous system (ENS) in the porcine descending colon, caused by chemically driven inflammation, proliferative enteropathy (PE), which is a "natural" inflammation with proliferative changes and nerve injury (axotomy). The distribution pattern of SOM-LI structures was studied using the immunofluorescence technique in the circular muscle layer, the myenteric (MP), outer submucous (OSP) and inner submucous plexuses (ISP) and also in the mucosal layer. Under physiological conditions SOM-LI…

A rat with a congenital aganglionosis develops a mega-small intestine and megacolon. This rat mutant has been used as an animal model for Hirschsprung's disease. The myenteric and submucosal plexuses of the rats were exposed by the trypsin-HCl digestion method and examined by scanning electron microscopy. In the control rats the myenteric and submucosal plexuses were present throughout the gut. However, in the mutant rats the myenteric plexus of duodenum was different from those of control rats; the mesh of plexuses varied both in size and shape. There were no myenteric or submucosal plexuses

A case of adult type mucolipidosis with beta-galactosidase and sialidase deficiency is described. This patient, a woman aged 20, had mental retardation, macular cherry-red spots, corneal clouding, gargoyle-like face, cerebellar ataxia, myoclonus and convulsions beginning at the age of 14. Bony deformities, vacuoles in the peripheral lymphocyte and foamy cells in the bone marrow were also noted. Biopsy study of the sural nerve and vermiform appendix disclosed many vacuoles in almost every kind of cells, although the accumulated substance in these vacuoles could not be characterized histochemica

Intestinal neuronal dysplasia type B (IND B) is currently considered to be a subtle malformation of the submucosal plexus, leading to an increased proportion of over-sized ganglia and potentially accompanied by a mild, chronic gastrointestinal motility disturbance. The diagnosis of IND B is morphologically based and involves the demonstration of an increased proportion of giant ganglia in the submucous plexus related to the patient's age. Giant ganglia are physiologically frequent in the neonatal period. Therefore, IND B should not be diagnosed prior to 1 year of age. Morphological features of…

RESULTS: A significantly higher proportion of submucous plexus neurones was trk C immunoreactive in control infant than adult colon (mean(s.e.m.) 73(9) versus 16(3) per cent of the total; P < 0.001), in accord with a role in development. The enteric submucosal plexus regulates essential digestive functions, yet its neuronal composition remains incompletely understood. We identified two putative secretomotor neuron classes and a previously unrecognized submucosal intrinsic primary afferent neuron class through single-cell RNA sequencing in the mouse small intestine. Using viral-mediated labeling of each class, we uncovered their morphologies and neural projections in the submucosa-mucosa context, finding connections among all classes and an unexpected close association with enterochromaffin cells. Further transcriptome analysis

Combined nicotinamide adenine dinucleotide phosphate-diaphorase (NADPH-d) histochemistry, nitric oxide synthase (NOS) and vasoactive intestinal polypeptide (VIP) immunocytochemistry were used to study the distribution of NOS- and VIP-containing nerve elements in the feline pylorus. A large number of stained multipolar neurons was found in the myenteric plexus. However, some NADPH-d and NOS positive neurons were also observed in the submucous plexus and in the internal part of muscular layer. A few stained perikarya were found in the tunica mucosa, in a very close situation to the blood vessels…
