Timing-dependent protective and therapeutic effects of ethyl pyruvate in trinitrobenzene sulfonic acid-induced experimental colitis.

Ulcerative colitis (UC) involves excessive inflammation, extracellular matrix remodelling and oxidative stress. Ethyl pyruvate (EP) exhibits anti-inflammatory effects, but the comparative efficacy of prophylactic versus therapeutic administration and timing-dependent molecular responses remains unclear. The objective of this work was to evaluate timing-dependent protective and therapeutic effects of EP in trinitrobenzene sulfonic acid (TNBS)-induced experimental colitis, focusing on inflammatory mediators, matrix metalloproteinases (MMPs), oxidative status and gene expression. Rats were random

Perfluorohexane Sulfonic Acid Disrupts the Immune Microenvironment for Spermatogenesis by Damaging the Structure of the Blood-Testis Barrier in Mice.

Perfluorohexane sulfonic acid (PFHxS) is extensively used in waterproof coatings and fire-fighting foams, and several studies have found it to be a potential health hazard, but there is still unknown about its effects on spermatogenesis. Our results showed that PFHxS-treated mice have significant reproductive toxicity, including a decrease in sperm count and motility, and the levels of sex hormones (P < 0.05). Concurrently, structural abnormalities are observed in sperm, affecting ≈60-75% of those in the PFHxS-treated group. Additionally, it is found that the structure of the blood-testis barr

Drug interactions with perfluorooctanoic acid and perfluorooctane sulfonate in cytotoxic activity against prostate cancer - in vitro studies.

Poly- and perfluoroalkyl substances (PFAS), including perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS), are persistent environmental pollutants with potential toxicological effects on human health. The aim of this study was to investigate the impact of PFOS and PFOA on the effectiveness of selected drugs used in the treatment of prostate cancer based on in vitro tests on cell lines. Three cell lines were used in the study: two human prostate cancer cells (DU-145 and PC3) and one human normal prostate cell line (PNT1A). Using dose-response experiments, it was observed that PFA

New insight on 8-anilino-1-naphthalene sulfonic acid interaction with TgFNR for hydrophobic exposure analysis.

The exposed hydrophobic patches of protein are widely detected through the binding by the fluorescent probes such as 1-anilino-8-naphthalene sulfonate (ANS), Nile Red (NR) and 1-(N-phenylamino) naphthalene, N-(1-Naphthyl) aniline (1NPN). Interestingly, at pH4, where the Toxoplasma gondii Ferredoxin-NADP(+) reductase (TgFNR) is stable, an exclusive binding and fluorescence emission was observed for ANS. To understand the underlying difference in the binding of ANS, NR and 1NPN; their effect on the protein structure was studied in detail. ANS was found to interact with TgFNR via electrostatic as

LN-1-255, a penicillanic acid sulfone able to inhibit the class D carbapenemase OXA-48.

OBJECTIVES: Carbapenemases are the most important mechanism responsible for carbapenem resistance in Enterobacteriaceae. Among carbapenemases, OXA-48 presents unique challenges as it is resistant to β-lactam inhibitors. Here, we test the capacity of the compound LN-1-255, a 6-alkylidene-2'-substituted penicillanic acid sulfone, to inhibit the activity of the carbapenemase OXA-48. METHODS: The OXA-48 gene was cloned and expressed in Klebsiella pneumoniae and Escherichia coli in order to obtain MICs in the presence of inhibitors (clavulanic acid, tazobactam and sulbactam) and LN-1-255. OXA-48 w

Anti-inflammatory effects of carbon monoxide-releasing molecule on trinitrobenzene sulfonic acid-induced colitis in mice.

BACKGROUND AND AIM: Recent findings indicate that carbon monoxide (CO) in non-toxic doses exerts a beneficial anti-inflammatory action in various experimental models. However, the precise anti-inflammatory mechanism of CO in the intestine remains unclear. Here, we assessed the effects of a novel water-soluble CO-releasing molecule, CORM-3, on trinitrobenzene sulfonic acid (TNBS)-induced colitis in mice. METHODS: To induce colitis, C57BL/6 male mice received an enema of TNBS. CORM-3 or its inactive compound, iCORM-3, were administered intraperitoneally, once immediately before, and twice daily

Validation and optimization of experimental colitis induction in rats using 2, 4, 6-trinitrobenzene sulfonic acid.

Trinitrobenzene sulfonic acid (TNBS)-induced colitis is one of the most common methods for studying inflammatory bowel disease in animal models. Several factors may, however, affect its reproducibility, rate of animal mortality, and macroscopic and histopathological outcomes. Our aim was to validate the main contributing factors to this method and compare the effects of different reference drugs upon remission of resultant colon injuries. TNBS was dissolved in 0.25 ml of ethanol (50% v/v) and instilled (25, 50, 100 and 150 mg/kg) intracolonically to the male Wistar rats. After determination of

The role of mast cells in the development of 2, 4, 6-trinitrobenzene sulfonic acid-induced colitis in rats.

BACKGROUND: The role of mast cells in Crohn disease (CD) remains to be established. The aim of this study was to elucidate this in the development of CD-like colitis in rats by the use of mast-cell-deficient Ws/Ws and their control W+/W+ rats. METHODS: CD-like colitis was induced in both groups by an enema of 10 mg of 2,4, 6-trinitrobenzene sulfonic acid (TNBS) in 50% ethanol. Colonic damage, adhesion and colonic weight were measured at 7 and 14 days after the TNBS/ethanol enema. Rat mast cell protease-2 (RMCP-2) in the colonic tissue was also measured at 7 days after the enema. RESULTS: The

Dermal Permeability of Perfluorohexane Sulfonic Acid (PFHxS) and Perfluorohexanoic Acid (PFHxA) in Varying Vehicles and the Effects on Skin Integrity in a Reconstructed Human Epidermis Model.

Per- and polyfluoroalkyl substances (PFAS) are widespread environmental contaminants associated with adverse health effects, yet dermal exposure remains poorly characterized. This study evaluated the influence of vehicle and chemical structure on dermal absorption and skin barrier integrity using a reconstructed human epidermis model (EpiDermFT). Two C6 PFAS, perfluorohexane sulfonate (PFHxS) and perfluorohexanoic acid (PFHxA), were applied in three vehicles (acetone, water, and diethylene glycol monobutyl ether [DEGME]) at concentrations of 0-0.015% for 4 or 24 h. PFAS concentrations were qua

Antidotal effects of 2,3-dimercaptopropane-1-sulfonic acid (DMPS) and meso-2,3-dimercaptosuccinic acid (DMSA) on the organotoxicity of dibutyltin dichloride (DBTC) in rats.

1. Dialkyltin compounds have been widely used in industry and agriculture, mainly as biocides, catalysts and plast stabilizer. In dependence on the length of the alkyl chains these organotins exert toxic effects on the immune system, the bile duct, liver and pancreas. It has been supposed that similar to organoarsenic the toxicity of the dialkyltin compounds is related to reactions with biological dithiol groups. Therefore, in the present study, the antidotal effects of 2,3-dimercapto-propane-1-sulfonic acid (DMPS) and meso-2,3-dimercaptosuccinic acid (DMSA) on the organotoxic effects of dibut

Core-shell alginate-sulfonic acid networks crosslinked by multivalent cations (Ca2+/Al3+) for atmospheric water harvesting.

A ternary core-shell hydrogel composite (Alg/CaAl@PSS) was synthesized via a two-step ionotropic gelation and interfacial polymerization strategy, combining a Ca2+/Al3+ cross-linked alginate core with a polystyrene sulfonic acid (PSS) shell mediated by an intermediate polyaniline layer, for application in atmospheric water harvesting. The effect of gelation pH (3-8) on core morphology and elemental composition was systematically investigated using SEM and EDS, revealing that pH = 7 produced the most homogeneous cross-linked network, with Al/Ca ratios of 2.90, 2.55, and 8.08 measured at pH 5, 7

Stability and Biotransformation of 6:2 Fluorotelomer Sulfonic Acid, Sulfonamide Amine Oxide, and Sulfonamide Alkylbetaine in Aerobic Sludge.

The 6:2 fluorotelomer sulfonamide (6:2 FTSAm)-based compounds signify a prominent group of per- and polyfluoroalkyl substances (PFAS) widely used in contemporary aqueous film-forming foam (AFFF) formulations. Despite their widespread presence, the biotransformation behavior of these compounds in wastewater treatment plants remains uncertain. This study investigated the biotransformation of 6:2 FTSAm-based amine oxide (6:2 FTNO), alkylbetaine (6:2 FTAB), and 6:2 fluorotelomer sulfonic acid (6:2 FTSA) in aerobic sludge over a 100-day incubation period. Soluble Protein Hydrolysate Ameliorates Gastrointestinal Inflammation and Injury in 2,4,6-Trinitrobenzene Sulfonic Acid-Induced Colitis in Mice.

Inflammatory bowel diseases (IBD) are chronic, recurring gastrointestinal diseases that severely impair health and quality of life. Although therapeutic options have significantly expanded in recent years, there is no effective therapy for a complete and permanent cure for IBD. Well tolerated dietary interventions to improve gastrointestinal health in IBD would be a welcome advance especially with anticipated favorable tolerability and affordability. Soluble protein hydrolysate (SPH) is produced by the enzymatic hydrolysis of commercial food industry salmon offcuts (consisting of the head, bac

Perfluorooctane sulfonic acid (PFOS) exposures interfere with behaviors and transcription of genes on nervous and muscle system in zebrafish embryos.

Perfluorooctane sulfonic acid (PFOS) has been widely detected in environment and organisms. PFOS has been identified as the driving agent for the behavioral changes of zebrafish larvae, while the underlying molecular mechanism remains unclear. In this study, zebrafish embryos/larvae were exposed to 0, 0.04, 0.1, 0.4 and 1 μM PFOS for 166 h. The locomotor behaviors and the mRNA transcription of genes in neuromuscular system were detected. Exposure to PFOS did not affect the hatching/death rates and body length, but increased the heart beat rates and frequency of spontaneous tail coiling. Locomo

Magnetic solid sulfonic acid decorated with hydrophobic regulators: a combinatorial and magnetically separable catalyst for the synthesis of α-aminonitriles.

A three-component, Strecker reaction of a series of aldehydes or ketones, amines, and trimethylsilyl cyanide for the synthesis of α-aminonitriles in the presence of a catalytic amount of a magnetic solid sulfonic acid catalyst, Fe3O4@SiO2@Me&Et-PhSO3H under solvent-free conditions have been investigated. This catalyst, with a combination of hydrophobicity and acidity on the Fe3O4@SiO2 core-shell of the magnetic nanobeads, as well as its water-resistant property, enabled easy mass transfer and catalytic activity in the Strecker reaction. The catalyst was easily separated by an external magnet a

Ecabet sodium promotes the healing of trinitrobenzene-sulfonic-acid-induced ulceration by enhanced restitution of intestinal epithelial cells.

BACKGROUND AND AIMS: Ecabet sodium (ES) is a gastric mucosal protective and ulcer-healing agent. Recently enema therapy with ES was found to be effective for the treatment of human ulcerative colitis as well as experimental colitis in an animal model. Whereas ES possesses potential as a novel treatment for ulcerative colitis, its precise mechanism of action remains to be elucidated. In this study, we investigated the therapeutic efficacy of ES in an experimental rat model of colitis, and evaluated the restitution of intestinal epithelial cells treated with ES in vitro. METHODS: Acute colitis
