Developmental Exposure of Mice to T-2 Toxin Increases Astrocytes and Hippocampal Neural Stem Cells Expressing Metallothionein.

We previously reported that developmental exposure to T-2 toxin caused transient disruption of the hippocampal neurogenesis targeting neural stem cells (NSCs) and early-stage progenitor cells involving oxidative stress on weaning in mouse offspring. The present study examined metallothionein (MT) expression changes and their cellular identity in brain regions of these animals. T-2 toxin at 0, 1, 3, and 9 mg/kg was given in the diet of maternal mice from gestational day 6 to postnatal day (PND) 21 on weaning. Offspring were maintained through PND 77 without T-2 toxin exposure. Male offspring we

Betulinic Acid Ameliorates T-2 Toxin-Induced Neuroinflammation by Suppressing Oxidative Stress via Regulating Nrf2/NLRP3 Axis.

T-2 toxin is widely present in agricultural products and poses a significant neurotoxicity threat. Betulinic acid (BA), a natural triterpenoid, exhibits strong antioxidant and anti-inflammatory properties. However, its protective role against T-2 toxin-induced neuroinflammation remains poorly understood. This study aimed to elucidate the mechanisms underlying T-2 toxin-induced neurotoxicity and evaluate the therapeutic potential of BA. Our results demonstrated that T-2 toxin (1 mg/kg/bw) exposure caused significant pathological damage in the hippocampus and cerebral cortex. T-2 toxin also indu

T-2 toxin induces degradation of chondrocytes extracellular matrix through the methyltransferase-like 3/m6A/Csf1 axis.

T-2 toxin can induce the development of cartilage diseases. N6-methyladenosine (m6A) modification mediated by methyltransferase-like 3 (METTL3) is involved in the occurrence and progression of bone-related diseases. However, the mechanism of action of METTL3 in T-2 toxin-caused cartilage injury has not been fully revealed. It is unclear whether METTL3 regulates cartilage injury by modulating its downstream target macrophage colony-stimulating factor (Csf1). Here, we aimed to investigate the mechanism by which the T-2 toxin induces cartilage damage. We performed histological staining, western b

Knockout of Mlkl alleviates myocardial damage and inflammation induced by T-2 toxin with low selenium conditions.

T-2 toxin is one of the most potent and commonly occurring secondary metabolites of type A trichothecene mycotoxins. Although it has been associated with cardiomyocyte necrosis, the underlying mechanisms of T-2 toxin-induced myocardial injury, particularly combined with low selenium conditions, remain unclear. This study investigates the role of MLKL, the terminal effector of the Receptor interacting protein kinase-1 and -3/mixed lineage kinase domain-like protein (RIP1-RIP3-MLKL) necroptotic pathway, in cardiac injury induced by T-2 toxin alone or in combination with low selenium. Histopathol

Melatonin alleviates T-2 toxin-induced oxidative damage, inflammatory response, and apoptosis in piglet spleen and thymus.

T-2 toxin, an unavoidable contaminant in animal feeds, can induce oxidative stress and damage immune organs. Melatonin (MT), a natural and potent antioxidant, has shown promise as a detoxifier for various mycotoxins. However, the detoxifying effect of MT on T-2 toxin has not been previously reported. In order to investigate the protective effect of MT added to diets on the immune system of T-2 toxin-exposed piglets, twenty piglets weaned at 28d of age were randomly divided into control, T-2 toxin (1 mg/kg), MT (5 mg/kg), and T-2 toxin (1 mg/kg) + MT (5 mg/kg) groups(n = 5 per group). Gene expression profiles and molecular mechanism of cultured human chondrocytes' exposure to T-2 toxin and deoxynivalenol.

T-2 toxin and deoxynivalenol (DON) are secondary metabolites produced by Fusarium fungi and are commonly found on food and feed. Although T-2 toxin and DON have been suggested as the etiology of Kashin-Beck disease (KBD), an endemic osteochondropathy, little is known about the mechanism when human chondrocytes are exposed to T-2 toxin and DON. The purpose of this study is to identify the gene expression differences and underlying molecular changes modulated by T-2 toxin and DON in vitro in human chondrocytes. After the experiments of cell viability, the gene expression profiles were analyzed i

Detoxification of T-2 toxin in human chondrocytes due to its hydroxylation by carboxylesterases and arylacetamide deacetylase in the liver.

T-2 toxin, a trichothecene mycotoxin produced by Fusarium species, is a global cereal contaminant and an important environmental risk factor for cartilage damage and Kashin-Beck disease (KBD). Although hydrolysis is a major metabolic pathway of T-2 toxin, the human enzymes responsible and their toxicological relevance remain poorly understood. In this study, human liver microsomes, recombinant hydrolases, and selective inhibitors were used to identify the key enzymes mediating T-2 hydrolysis, and LC-MS/MS was applied to quantify T-2 and its major metabolites. Human chondrocytes were further us

Investigating the effects of thymol and cinnamaldehyde in T-2 toxin-treated hepatic cell cultures of chicken origin.

Trichothecenes are among the most significant mycotoxins, causing serious agricultural problems worldwide. One representative of these is T-2 toxin, which is often found in poultry feed. Due to its harmful effects, it is important to investigate substances that can counteract it. Such compounds include thymol and cinnamaldehyde, whose anti-inflammatory and antioxidant activities are well-documented. In this study, the effects of 24 h exposure to T-2 toxin (100 nM), thymol, and cinnamaldehyde (10 and 100 μg/mL in both cases) and their combinations with the toxin were examined in chicken-derived

The nephrotoxicity of T-2 toxin in mice caused by oxidative stress-mediated apoptosis is related to Nrf2 pathway.

T-2 toxin is an inevitable environmental and grain pollutant, which can cause kidney damage, but the mechanism is not clear. In this study, male mice were administered with T-2 toxin at 0, 0.5, 1.0, 2.0 mg/kg body weight (BW) for 28 days. We found that T-2 toxin induced renal structural damage, downregulated BW and kidney coefficient, impaired renal function accompanied by oxidative stress and apoptosis. Meanwhile, T-2 toxin increased nuclear Nrf2 protein expression and the mRNA expressions of its downstream target genes. The correlation analysis indicated that apoptosis and Nrf2 pathway were

The decreased expression of integrin αv is involved in T-2 toxin-induced extracellular matrix degradation in chondrocytes.

T-2 toxin is one of the most toxic and common mycotoxins in grains and related products. It is considered a risk factor for Kashin-Beck disease (KBD), an endemic osteoarthritis. Both in vitro and in vivo studies have shown that T-2 toxin can cause extracellular matrix degradation; however, the underlying mechanism is unclear. Integrins have been found to regulate the expression of matrix metalloproteinases (MMPs), the 'scissors' of matrix proteins. In this study, we investigated whether integrin αv played a role in T-2 toxin-induced matrix degradation. Results from our study showed that the ex

Synergistic toxicity of T-2 toxin and diniconazole in zebrafish: Involvement of gut-liver homeostasis dysregulation and ferroptosis.

Mycotoxin and pesticide co-contamination represents a critical threat to aquatic ecosystem integrity, yet the toxicological effects and underlying mechanisms of T-2 toxin (T-2) and diniconazole (DIN), two widely coexisting agricultural contaminants, remain elusive. Here, adult male and female zebrafish were exposed to T-2 (4.25 μg L⁻¹), DIN (32.79 μg L⁻¹), or their combination for 60 days to investigate gut-liver homeostasis dysregulation and related toxic effects. Results demonstrated that co-exposure significantly perturbed gut microbial community structure and disrupted gut-liver balance. T

The critical role of p16/Rb pathway in the inhibition of GH3 cell cycle induced by T-2 toxin.

T-2 toxin is a worldwide trichothecenetoxin and can cause various toxicities.T-2 toxin is involved in G1 phase arrest in several cell lines but molecular mechanism is still not clear. In present study, we used rat pituitary GH3 cells to investigate the mechanism involved in cell cycle arrest against T-2 toxin (40 nM) for 12, 24, 36 and 48 h as compared to control cells. GH3 cells showed a considerable increase in reactive oxygen species (ROS) as well as loss in mitochondrial membrane potential (△Ym) upon exposure to the T-2 toxin. Flow cytometry showed a significant time-dependent increase in

Tert-Butylhydroquinone Mitigates T-2-Toxin-Induced Testicular Dysfunction by Targeting Oxidative Stress, Inflammation, and Apoptosis in Rats.

Tert-butylhydroquinone (tBHQ) has emerged as a promising candidate for mitigating the adverse effects of T-2-induced reproductive toxicity. The protective effects of tBHQ on rat sperm quality, testicular injury, apoptosis, and inflammation induced by T-2 toxin exposure were investigated. Histopathological examination of testicular tissues revealed severe damage in the T-2-treated group, characterized by disorganized germ cell arrangement, thinning of the convoluted seminiferous tubule walls, and significant cellular necrosis. However, tBHQ administration, either as a preventive or therapeutic
