An in situ hybridization study of syndecan family during the late stages of developing mouse molar tooth germ.

Expression of syndecan-1, 2, 3, and 4 mRNAs during the late stages of tooth germ formation was investigated by in situ hybridization, using [35S]-UTP-labeled cRNA probes. Syndecan-1 mRNA was mainly expressed in the stellate reticulum and stratum intermedium as well as at the cervical region of dental papilla/dental follicle during E18.5-P3.0. Expression in the dental epithelium was enhanced during the postnatal periods, which was supported by real-time RT-PCR analysis. These spatiotemporal expression patterns may suggest specific roles of syndecan-1 in tooth formation such as tooth eruption or…

[Establishment of permanent tooth germ missing animal model and study on root resorption of the corresponding deciduous teeth].

OBJECTIVE: To establish the permanent tooth germ missing animal model for future research on the root resorption of deciduous tooth in the absence of permanent tooth germ. METHODS: The permanent tooth germ missing animal model was established by surgical removal of the permanent tooth buds in a male 11-week-old Beagle dog. Root resorption of the deciduous teeth without permanent successors was observed by taking periapical films periodically, and compared with physiological root resorption. Once the sign of root resorption of the deciduous teeth without permanent successors was detected on rad…

MFAP5 Activates ITGA5 to Drive Tooth Germ Mineralization Through the MAPK/ERK Pathway: Insights from Single-Cell Transcriptomics.

Tooth germ development is a precisely orchestrated process dependent on integrated cellular interactions and molecular signals, yet its regulatory mechanisms remain incompletely defined. Here, we constructed a high-resolution cellular atlas of miniature pig tooth germs using 10× single-cell RNA sequencing to investigate the molecular mechanisms underlying tooth mineralization. By leveraging cellular heterogeneity and dynamic gene expression trajectories in epithelial and mesenchymal populations, we identified microfibril-associated protein 5 (MFAP5) as a previously unrecognized regulator of th…

Changes of cytochemical properties in the Golgi apparatus during in vivo differentiation of the ameloblast in developing rat molar tooth germs.

The cytochemical changes of the Golgi stacks occurring concomitantly with cell differentiation were examined in ameloblasts of developing rat molar tooth germs using osmium impregnation and cytochemistry with nicotinamide adenine dinucleotide phosphatase (NADPase), thiamine pyrophosphatase (TPPase), and acid phosphatase (Acpase). NADPase, TPPase, and Acpase activities were already present in the Golgi stacks of the inner enamel epithelial cells, the undifferentiated form of the ameloblast: NADPase activity existed in the medial Golgi cisternae, TPPase activity in the trans Golgi cisternae, and…

Dentin sialoprotein: biosynthesis and developmental appearance in rat tooth germs in comparison with amelogenins, osteocalcin and collagen type-I.

A non-collagenous protein, extracted from rat incisor dentin, is a dentin sialoprotein (DSP). We examined immunohistochemically the developmental appearance and tissue distribution of DSP in 1 to 3-day-old rat molar and incisor tooth germs. The earliest staining for DSP was observed in newly differentiated odontoblasts. In more advanced stages, immunostaining for DSP gradually increased in pre-dentin, odontoblasts and dentin, and appeared in many cells of the dental papilla. In early stages of development before the breakdown of the dental basement membrane, pre-ameloblasts were also positive …

Changes of lectin staining pattern of the Golgi stack during differentiation of the ameloblast in developing rat molar tooth germs.

Changes of lectin staining patterns in the Golgi stack during cell differentiation were examined in the ameloblasts of developing rat molar tooth germs, using HRP-labeled lectins: Canavalia ensiformis (Con A), Griffonia simplicifolia I (GS-I), Glycine max (SBA), Ulex europeus I (UEA-I), Triticum vulgaris (WGA), and Arachis hypogaea (PNA). The Golgi stacks of the inner enamel epithelial cells and the presecretory ameloblasts were stained with the lectins, although the staining strength and pattern varied among the stacks with each lectin. In some cases, the reaction products for the lectins wer…

Budding of small vesicles from the rough-surfaced endoplasmic reticulum in secretory ameloblasts of rat molar tooth germs.

The budding of small vesicles from the rough-surfaced endoplasmic reticulum (rER) was examined in the secretory ameloblast of rat molar tooth germs by ordinary fixation or prolonged osmium fixation. The budding of small vesicles from the rER was observed not only at the special region (transitional region) of the rER system, which abutted on the cis-face of the Golgi apparatus, but also at other regions of the rER in the secretory ameloblast. Small vesicles (presumed to be transitional vesicles) were adjacent to the rER, which also showed budding of vesicles. After prolonged osmium treatment, …

Three-dimensional localization of tooth germs without ionizing radiation : Proof-of-concept study using magnetic resonance imaging.

OBJECTIVES: This study aimed to evaluate the feasibility and accuracy of using magnetic resonance imaging (MRI) for the three-dimensional (3D) localization of tooth germs. A MRI 3D dataset was compared to a cone-beam computed tomography (CBCT) dataset, which is the current gold standard method in the localization of tooth germs. In this proof-of-concept study, the potential of eliminating the exposure of patients to ionizing radiation for tooth germ localization was evaluated. METHODS: A halved pig head (Sus scrofa domestica) was imaged with MRI and CBCT. The MRI data were manually segmented …

A histological study of the short-term effects of fluoride on enamel and dentine formation in hamster tooth-germs in organ culture in vitro.

First maxillary molars in early stages of secretory amelogenesis were exposed in vitro to fluoride (F-) concentrations ranging between 2.63 microM and 2.63 mM for up to 3 days. In contrast to the dentine papilla, which seemed unaffected by F- in concentrations up to 1.31 mM, the enamel organ was dose-dependently extremely sensitive for F-. Young ameloblasts which became secretory in vitro were in particular sensitive and did not secrete enamel adjacent to new, in vitro-formed dentine. Hypermineralization of the dentine at the enamel-dentine junction suggested that these ameloblasts still trans

Apical tooth germ cell-conditioned medium enhances the differentiation of periodontal ligament stem cells into cementum/periodontal ligament-like tissues.

BACKGROUND AND OBJECTIVE: Limitations of current periodontal regeneration modalities in both predictability and extent of healing response, especially on new cementum and attachment formation, underscore the importance of restoring or providing a microenvironment that is capable of promoting the differentiation of periodontal ligament stem cells (PDLSCs) towards cementoblast-like cells and the formation of cementum/periodontal ligament-like tissues. The aim of this study was to investigate the biological effect of conditioned medium from developing apical tooth germ cells (APTG-CM) on the diff…

Antagonism of fluoride toxicity by high levels of calcium but not of inorganic phosphate during secretory amelogenesis in the hamster tooth germ in vitro.

Whether the interference by fluoride (F-) with secretory amelogenesis in vitro could be modulated by altering the levels of calcium (Ca) and inorganic phosphate (P) in the medium was investigated. Hamster first upper molar tooth germs in the secretory phase of amelogenesis were exposed to 10 microM-1.31 mM (0.2-25 parts/10(6)) of F- in vitro for 2 days in the presence of either low (1.2 mM), moderate (2.1 mM) or high (4.1 mM) levels of Ca, or moderate (1.6 mM) and high (3.6 mM) levels of P. The biosynthesis and secretion of enamel matrix proteins under each of the experimental conditions were …

Cytochemical study of the Golgi apparatus and related organelles of the secretory ameloblasts of rat molar tooth germs cultured with and without colchicine.

The Golgi apparatus and Golgi-associated endoplasmic reticulum lysosome (GERL) were examined in the ameloblasts with a cytochemical marker, … ow that Golgi stacks of ameloblasts consist of several distinct compartments. In colchicine-treated tooth germs, there were morphological and cytochemical changes in both Golgi stacks and GERL. The Golgi apparatus was fragmented and its stacks were scattered throughout the supranuclear region. In some stacks, the number of osmium-positive cisternae was great …

Accelerated bone formation and increased osteoblast number contribute to the abnormal tooth germ development in parathyroid hormone-related protein knockout mice.

Our previous study showed that tooth germs at late embryonic stage [later than embryonic day 17.5 (E17.5)] and neonatal homozygous parathyroid hormone-related protein (PTHrP)-knockout mice are compressed or penetrated by the surrounding alveolar bone tissue. In vivo and in vitro studies have shown that the development of the tooth germ proper is not disturbed, but insufficient alveolar bone resorption, due to the decreased number and hypofunction of osteoclasts, is the main cause of this abnormality. In addition to the insufficient alveolar bone resorption, progressive bone formation toward to…

Chondroitin sulfates in developing mouse tooth germs. An immunohistochemical study with monoclonal antibodies against chondroitin-4 and chondroitin-6 sulfates.

The role of glycosaminoglycans and proteoglycans during ontogenesis is not known. The developing tooth offers a potentially important model for studies of structure-function relationships. In this study, we have analysed the temproal and spatial expression of chondroitins of differing sulfation patterns in embryonic molars and incisors. For this purpose, we have used monoclonal antibodies (Mabs) specific for unsulfated, 4-sulfated, and 6-sulfated forms of chondroitin in conjunction with indirect immunofluorescence or immunoperoxidase labeling. Unsulfated chondroitin was not detected in embryon
