When treated with heat-killed bacterial cells, mosquito cells in culture respond by up-regulating several proteins. Among these is a 66-kDa protein (p66) that is secreted from cells derived from both Aedes aegypti and Aedes albopictus. p66 was degraded by proteolysis and gave a virtually identical pattern of peptide products for each mosquito species. The sequence of one peptide (31 amino acids) was determined and found to have similarity to insect transferrins. By using conserved regions of insect transferrin sequences, degenerate oligonucleotide PCR primers were designed and used to isolate

The relationship of the serum iron-binding capacity (IBC) to the serum proteins and other haematological parameters was studied in 96 elderly subjects. With two exceptions, IBC levels above the normal range were confined to cases of anaemia with transferrin saturation levels of less than 12%. In 13 cases the IBC was below normal, and in 9 it was normal in the presence of iron deficiency; in these groups the IBC correlated with serum albumin, and there was a clinical explanation for protein depletion. In cases with normal serum albumin and normal transferrin saturation, there was no decline in

The lack of experimental models suitable for the study of meningococcal pathogenicity led us to investigate if those actually in use (culture in iron-restricted media and animal models) provide results comparable with the responses observed in vivo during infection. In this work we studied three invasive strains cultured both in laboratory media and in human plasma, analysing the immune responses elicited in mice against membrane antigens and comparing them with those seen using homologous human convalescent sera. Outer membrane protein profiles observed after culture in plasma were different

A series of new iron(III) and copper(II) complexes of bovine serum transferrin (BTf), with carbonate and/or oxalate as the synergistic anion, are presented. The complexes [Fe(2)(CO(3))(2)BTf], [Fe(2)(C(2)O(4))(2)BTf], [Cu(2)(CO(3))(2)BTf] and [Cu(C(2)O(4))BTf] were prepared by standard titrimetric techniques. The oxalate derivatives were also obtained from the corresponding carbonate complexes by anion-displacement. The site-preference of the transition metal-oxalate synergism has facilitated the preparation and isolation of the mononuclear complex [Cu(C(2)O(4))BTf], the mixed-anion complexes

The hypotransferrinemic (hpx) mouse is a model of inherited transferrin deficiency that originated several decades ago in the BALB/cJ mouse strain. Also known as the hpx mouse, this line is almost completely devoid of transferrin, an abundant serum iron-binding protein. Two of the most prominent phenotypes of the hpx mouse are severe anemia and tissue iron overload. These phenotypes reflect the essential role of transferrin in iron delivery to bone marrow and regulation of iron homeostasis. Over the years, the hpx mouse has been utilized in studies on the role of transferrin, iron and other me

Oxidative protein folding remains a subject of intense investigation, with ongoing debate surrounding the specific molecular actors driving this process. In this context, the involvement of oxidized glutathione (GSSG) is supported by the recent discovery of an enigmatic, previously undescribed hyper-reactivity toward this disulfide. This phenomenon is exhibited by structural cysteines within the reduced, molten-globule intermediates of several proteins, including bovine serum albumin, hen-egg lysozyme, human lysozyme, ribonuclease A, trypsinogen, chymotrypsinogen, and lactoferrin. Notably, fou

Abnormal iron metabolism, characterized by low serum iron concentration and diminished saturation of iron-binding capacity, is known to occur in patients with malignant or inflammatory diseases. In this study, serum iron concentration and iron-binding capacity were measured in 21 patients with malignant disease, trauma or inflammatory illness before and during intravenous nutritional support without iron supplementation. Prior to nutritional support, the serum iron value averaged 26.0 +/- 3.5 micrograms per deciliter and the iron-binding capacity, 160.0 +/- 14.0 micrograms per deciliter. After

Transferrin plays an important role in immune response of vertebrates. In the present study, a transferrin cDNA with a partial 5' UTR of 7 bp and a complete 3' UTR of 345 bp was obtained from the liver of roughskin sculpin, Trachidermus fasciatus, which encodes a deduced 681 amino acid protein containing an N-terminal signal peptide and two conserved lobes. In the N-terminal lobe, the anion-binding residue Arg was substituted with Lys, which represents a common feature in fish and implies a selective preference in the transferrin evolutionary process. In contrast to mammalian transferrin, the

The modulation of cellular iron metabolism depends on the expression of ferritin and transferrin receptor (TfR). These proteins are regulated post-transcriptionally by interaction of specific sequences, termed iron responsive elements (IRE) which are located in the 5' untranslated region (UTR) of ferritin mRNA and 3'UTR of TfR mRNA and a specific cytosolic protein, termed iron responsive element-binding protein (IRE-BP). The liver plays an important role in iron metabolism as a major organ of iron storage. In hepatocytes iron is taken up in the form of transferrin and intercellular iron levels

Haemophilus influenzae is a heme-dependent bacterium. However, little is known of the heme-iron uptake mechanism in this organism. By using a batch ligand affinity chromatography method, a hemin-binding protein of 39,500 molecular weight was isolated from total membranes derived from H. influenzae type b grown under iron-depleted but not under iron-sufficient conditions. Detection of the hemin-binding protein in a whole-cell binding assay demonstrated a surface-exposed location. Competition binding experiments indicated that this hemin-protein interaction was specific, since only hemin or heme

Neisseria are pathogenic bacteria that cause gonorrhea, septicemia, and meningitis. Like other pathogenic bacteria, Neisseria must acquire iron for survival from their local environment within the human host. Instead of secreting siderophores to scavenge iron, Neisseria steal iron from human iron binding proteins such as hemoglobin, transferrin and lactoferrin for survival. Recently we reported the crystal structures of the Neisseria meningitidis transferrin receptors TbpA and TbpB, as well as the structures of apo and holo human transferrin. We also analyzed these proteins using small angle X

Neisseria meningitidis acquires iron through the action of the transferrin (Tf) receptor, which is composed of the Tf-binding proteins A and B (TbpA and TbpB). Meningococci can be classified into isotype I and II strains depending on whether they harbor a type I or II form of TbpB. Both types of TbpB have been shown to differ in their genomic, biochemical, and antigenic properties. Here we present a comparative study of isogenic mutants deficient in either or both Tbps from the isotype I strain B16B6 and isotype II strain M982. We show that TbpA is essential in both strains for iron uptake and

Naegleria fowleri causes acute and fulminant primary amoebic meningoencephalitis. This microorganism invades its host by penetrating the olfactory mucosa and then traveling up the mesaxonal spaces and crossing the cribriform plate; finally, the trophozoites invade the olfactory bulbs. During its invasion, the protozoan obtains nutrients such as proteins, lipids, carbohydrates, and cationic ions (e.g., iron, calcium, and sodium) from the host. However, the mechanism by which these ions are obtained, particularly iron, is poorly understood. In the present study, we evaluated the ability of N. fo

Human transferrin is a single-chain bilobal protein with each of the two similar but not identical lobes in turn composed of two domains. Each lobe may assume one of two stable structural conformations, open or closed, determined by a rigid rotation of the domains with respect to each other. In solution, the transformation of a lobe between open and closed conformations is associated with the release or binding of an Fe(III) ion. The results of the present study indicate that encapsulation of transferrin within a porous sol-gel matrix allows for a dramatic expansion, to days or weeks, of this

Human serum transferrin is an essential bilobal protein that transports iron in the circulation for delivery to iron-requiring cells. Obtaining the C-terminal lobe of human transferrin in verified native conformation has been problematic, possibly because its 11 disulfide bonds lead to misfolding when the lobe is expressed without its accompanying N-lobe. A recently reported method for preparing the C-lobe free of extraneous residues, with normal iron-binding properties and capable of delivering iron to cells, makes use of a Factor Xa cleavage site inserted into the interlobal connecting stran

IgG, IgA, IgM, and transferrin levels were measured in sera from a group of children who had been subjected to splenectomy in the previous 10 years. In those children splenectomized for hereditary spherocytosis, idiopathic thrombocytopenic purpura, or traumatic rupture, mean IgM levels were significantly lower and mean transferrin levels were significantly higher than in sera from control children. Children splenectomized for thalassaemia major had a significantly raised mean IgG level, while children splenectomized for portal hypertension had a raised mean IgA level. However, IgG and IgA leve

The transferrin-binding proteins TbpA and TbpB enable Neisseria gonorrhoeae to obtain iron from human transferrin. The lipoprotein TbpB facilitates, but is not strictly required for, TbpA-mediated iron acquisition. The goal of the current study was to determine the contribution of two conserved regions within TbpB to the function of this protein. Using site-directed mutagenesis, the first mutation we constructed replaced the lipobox (LSAC) of TbpB with a signal I peptidase cleavage site (LAAA), while the second mutation deleted a conserved stretch of glycine residues immediately downstream of

Isolated rat liver mitochondria accumulate iron from fully saturated transferrin at neutral pH. With 5 microM iron as diferric transferrin, accumulation at 30 degrees C amounts to approx. 40 pmol/mg protein per h. With access to a suitable porphyrin substrate, 70-80% of the amount of iron accumulated is recovered in heme. Mobilization of iron and synthesis of heme both depend on a functioning respiratory chain. Vacant iron-binding sites on mono- and apotransferrin compete with the mitochondria for iron mobilized from transferrin. Pyrophosphate at concentrations in the range 10-50 microM enhanc

Worldwide stocks of actinides and lanthanide fission products produced through conventional nuclear spent fuel are increasing continuously, resulting in a growing risk of environmental and human exposure to these toxic radioactive metal ions. Understanding the biomolecular pathways involved in mammalian uptake, transport and storage of these f-elements is crucial to the development of new decontamination strategies and could also be beneficial to the design of new containment and separation processes. To start unraveling these pathways, our approach takes advantage of the unique spectroscopic

Chronic alcoholism has been reported to be associated with a reduced carbohydrate content of transferrin (TF), particularly, its reduced sialylation state. Low sialylation state of TF now serves as an objective marker of chronic alcohol abuse. To investigate the pathophysiological significance of this finding in relation to hepatic siderosis, also commonly associated with chronic alcoholism, the authors have investigated the effect of ethanol on the uptake, binding, and desialylation of transferrin by isolated rat liver endothelium in vitro. In pulse-chase experiments, transferrin labeled with
