Brain diseases are a major cause of death and disability worldwide and contribute significantly to years of potential life lost. Although there have been considerable advances in biological mechanisms associated with brain disorders as well as drug discovery paradigms in recent years, these have not been sufficiently translated into effective treatments. This Special Article expands on Keystone Symposia's pre- and post-pandemic panel discussions on translational neuroscience Исследование. In the article, we discuss how lessons learned from the COVID-19 pandemic can catalyze critical progress in tr

INTRODUCTION: Glaucoma, a leading cause of irreversible blindness in the world, is a chronic neurodegenerative disease of multifactorial origin. Extensive Исследование is ongoing to better understand, prevent, and treat progressive degeneration of retinal ganglion cells in glaucoma. While experimental models of glaucoma and postmortem tissues of human donors are analyzed for pathophysiological comprehension and improved treatment of this blinding disease, clinical samples of intraocular biofluids and blood collected from glaucoma patients are analyzed to identify predictive, diagnostic, and progno

This is a report from the 21st Meeting of the European Society of Gynaecological Oncology (ESGO 2019) held in Athens, Greece, November 2-5, 2019. The conference offered state of the art educational sessions, and oral and poster abstract presentations. The general sessions throughout the meeting focused not only on prevention, screening, diagnosis, treatment, and translational Исследование but also on emerging trends. Current innovations in gynecological cancers were also discussed. The new rare tumor guidelines project, a joint initiative with the ESGO-Gynecologic Cancer InterGroup, was officially

Various forms of early onset non-autoimmune diabetes are recognized as monogenic diseases, each subtype being caused by a single highly penetrant gene defect at the individual level. Monogenic diabetes (MD) is clinically and genetically heterogeneous, including maturity onset diabetes of the young and infancy-onset and neonatal diabetes mellitus, which are characterized by functional defects of insulin-producing pancreatic β-cells and hyperglycemia early in life. Depending on the genetic cause, MD differs in the age at diabetes onset, the severity of hyperglycemia, long-term diabetic complicat

Neuromuscular disorders (NMDs) encompass a diverse group of genetic diseases characterized by loss of muscle functionality. Despite extensive efforts to develop therapies, no curative treatment exists for any of the NMDs. For multiple disorders, however, therapeutic strategies are currently being tested in clinical settings, and the first successful treatments have now entered clinical practice (e.g. spinraza for spinal muscular atrophy). Successful clinical translation depends on the quality and translatability of preclinical findings and on the predictive value of the experimental models use

Pneumocystis pneumonia (PcP) remains a significant cause of morbidity and mortality in immunocompromised persons, especially those with human immunodeficiency virus (HIV) infection. Pneumocystis colonization is described increasingly in a wide range of immunocompromised and immunocompetent populations and associations between Pneumocystis colonization and significant pulmonary diseases such as chronic obstructive pulmonary disease (COPD) have emerged. This mini-review summarizes recent advances in our clinical understanding of Pneumocystis and PcP, describes ongoing areas of clinical and trans

Triplex-forming oligonucleotides (TFOs) are programmable nucleic acid ligands that recognize duplex DNA through Hoogsteen or reverse Hoogsteen interactions, providing a unique molecular recognition strategy beyond conventional Watson-Crick hybridization. Recent advances in nucleic acid engineering, chemical modification, nanotechnology, and signal amplification have transformed TFOs from classical antigene agents into versatile platforms for biosensing and bioanalysis. In this review, we highlight the chemical principles governing triplex formation and discuss recent developments in TFO-enable

Alzheimer’s disease (AD) is the most common neurodegenerative disorder, affecting millions worldwide. The clinical presentation of AD is heterogenous and often complicated by the presence of multiple co-pathologies, which can overlap with features of other neurodegenerative disorders, underscoring the need for tools to aid accurate and timely diagnosis. Biomarkers for AD span multiple modalities and are being increasingly used in clinical settings to aid diagnosis, monitor disease progression, and assess eligibility and response to disease-modifying therapies. Here, we provide an overview of c

The successful translation of a stem cell-derived product from the laboratory into an approved medicine requires specific scientific, technical, and regulatory understanding. Since this knowledge base is fragmented across websites and publications, the ISSCR has developed a new resource, The Best Practices for the Development of Pluripotent Stem Cell-Derived Therapies. This comprehensive, interactive document, designed for academia and industry, addresses critical areas of the translational pipeline and key decision points for the successful translation to a licensed therapy. Uniquely, it prov

The rising rates of antimicrobial resistance (AMR) have become a significant concern, especially as untreatable infections spiral out of control. This has opened new opportunities for developing novel therapeutic approaches. The therapeutic application of phage therapy, utilizing bacteriophages, has gained importance for combating multidrug-resistant (MDR) pathogens, either as an adjuvant or as an alternative to antibiotics. This review evaluates recent advancements in bacteriophage purification technologies in the context of phage therapy against antibiotic resistance, including their modes o

Hydrogen sulfide (H2S), an endogenous gaseous signaling molecule, plays crucial roles in physiological regulation. However, efficient delivery and controlled release of H2S remain major challenges for clinical application. Hydrogels, with the unique three-dimensional network structure, provide a versatile platform for H2S loading and controlled release, offering a promising strategy to address these limitations. This article reviews the fabrication strategies of H2S-releasing hydrogels, including H2S donors, hydrogel properties, and incorporation strategies. In addition, biomedical application

The rapid expansion of immuno-oncology (I-O) and other advanced therapies is reshaping the complexity of early-phase clinical development. While core Phase I principles remain fundamental across oncology, emerging modalities introduce additional requirements for translational integration, specialized safety management, and therapeutic-specific infrastructure. However, the organizational functions of Phase I units have not evolved at the same pace and remain largely centered on conventional operational responsibilities. Drawing on the practices of leading Phase I units across the USA, Europe, a

Eosinophil basic biology, including immunoregulatory functions, plays a complex role in a myriad of disorders where eosinophils play a role. Established in vivo and in vitro models and novel emerging techniques for studying eosinophils are integral to parsing the involvement of eosinophils in the pathogenesis of disorders including vasculitis. Knowledge of translational approaches from eosinophil-associated diseases and their application to eosinophilic vasculitis are highlighted. It is becoming increasingly evident that the roles of eosinophils in disease are complex. The use of targeted biol

BACKGROUND: Arthrogryposis multiplex congenita (AMC) is a group of rare disorders characterized by multiple joint contractures present at birth. Early rehabilitation is essential to minimize joint contractures and maximize autonomy and participation among individuals with AMC. However, there is little robust scientific evidence to inform best practice. This project aimed to develop consensus-based recommendations for the rehabilitation management of children with AMC in the following priority areas: early intervention and motor development, muscle and joint function, orthotics, mobility, parti

The most common genetic cause of intellectual and developmental disability is trisomy of human chromosome 21 (trisomy 21) or Down syndrome. Relative to the general population, individuals with Down syndrome heterogeneously experience atypical morphogenesis, a distinct neurocognitive profile, and a unique spectrum of diverse medical conditions that impact every major organ system. How trisomy 21 results in the highly variable manifestations of Down syndrome remains largely unknown and an active area of heavy investigation with therapeutic implications. For example, common inflammatory and metab
