The Dog as a Model to Study the Tumor Microenvironment.

Cancer is a complex and dynamic disease with an outcome that depends on a strict crosstalk between tumor cells and other components in tumor microenvironment, namely, tumor-infiltrating immune cells, fibroblasts, cancer stem cells, adipocytes, and endothelial cells. Within the tumor microenvironment, macrophages and T-lymphocytes appear to be key effectors during the several steps of tumor initiation and progression. Tumor cells, through the release of a plethora of signaling molecules, can induce immune tolerance, by avoiding immune surveillance, and inhibit immune cells cytotoxic functions. …

An Organotypic Microcosm for the Pancreatic Tumor Microenvironment.

Pancreatic duct adenocarcinoma (PDAC) is projected to become the second leading cause of cancer-related deaths in the next few years. Unfortunately, the development of novel therapies for PDAC has been challenged by a uniquely complex tumor microenvironment. The development of in vitro cancer organoids in recent years has demonstrated potential to increase therapies for patients with PDAC. Organoids have been established from PDAC murine and human tissues and they are representative of the primary tumor. Further, organoids have been shown beneficial in studies of molecular mechanisms and drug …

The multifaceted effects of metformin on tumor microenvironment.

The efficacy of metformin in treating cancer has been extensively investigated since epidemiologic studies associated this anti-diabetic drug with a lower risk of cancer incidence. Since tumors are complex systems, in which cancer cells coexist and interact with several different types of non-malignant cells, it is not surprising that anti-cancer drugs affect not only cancer cells, but also the abundance and functions of cells of the tumor microenvironment. Recent years have seen a wide collection of reports showing how metformin, as well as other complex I inhibitors, may influence cancer pro…

The Role of Tumor Microenvironment in Cancer Immunotherapy.

The field of tumor immunology and immunotherapy has undergone a renaissance in the past decade do in large part to a better understanding of the tumor immune microenvironment. After suffering countless successes and setbacks in the twentieth century, immunotherapy has now come to the forefront of cancer research and is recognized as an important tool in the anti-tumor armamentarium. The goal of therapy is to aid the immune system in recognition and destruction of tumor cells by enhancing its ability to react to tumor antigens. This traditionally has been accomplished by induction of adaptive i

The role of the microenvironment in tumor immune surveillance.

The evidence appears compelling that the microenvironment, and associated biological cellular and molecular factors, may contribute to the progression of a variety of tumors. The effects of the microenvironment may directly influence the plasticity of T cell lineages, which was recently discussed (O'Shea & Paul, 2010 [4]). To review the putative role of the microenvironment in modulating the commitment of tumor immune surveillance, we use the model of oral premalignant lesions.

The double-positive cells in the tumor microenvironment.

The tumor microenvironment substantially influences cancer progression by mediating complex interactions between immune cells, fibroblasts, endothelial cells, and mesenchymal cells. Recent studies have identified a critical component of this ecosystem, double-positive cells (DPCs), which are characterized by simultaneously expressing two markers that are traditionally confined to completely different cell lineages or cell types. In this review, we demonstrated DPCs' formation principles, characterization, classification, functions, and clinical significance. We underscore the multifaceted cont…

Metabolic regulation of immunity in the tumor microenvironment.

Metabolic-immune crosstalk in the tumor microenvironment (TME) is a critical driver of tumorigenesis, progression, and immune evasion. Tumor cells undergo profound metabolic reprogramming, causing nutrient competition, toxic metabolite accumulation, and the formation of cold niches that gradually exhaust effector immune cells. In contrast, immunosuppressive cells exhibit strong metabolic adaptability, reinforcing the suppressive milieu. Moreover, tertiary lymphoid structures provide nutrient- and oxygen-rich "moats" that sustain the functions of B and T cells. In addition, metabolic-immune int…

Role of the tumor microenvironment in CLL pathogenesis.

Chronic lymphocytic leukemia (CLL) cells extensively interact with and depend on their surrounding tumor microenvironment (TME). The TME encompasses a heterogeneous array of cell types, soluble signals, and extracellular vesicles, which contribute significantly to CLL pathogenesis. CLL cells and the TME cooperatively generate a chronic inflammatory milieu, which reciprocally reprograms the TME and activates a signaling network within CLL cells, promoting their survival and proliferation. Additionally, the inflammatory milieu exerts chemotactic effects, attracting CLL cells and other immune cel…

Integrating tumor microenvironment modulation with nanocarrier engineering for enhanced tumor-targeted delivery.

The tumor microenvironment (TME) imposes vascular, stromal, metabolic, and immune barriers that restrict nanoparticle (NP) extravasation, interstitial penetration, cellular access, and therapeutic activity in solid tumors. Because the dominant barrier and the delivery step most directly constrained vary across tumor types, platforms, and experimental models, neither TME modulation nor nanocarrier optimization can be treated as a universal solution. This review critically examines vascular, extracellular matrix (ECM), hypoxia/acidosis, and immune-modulating strategies together with nanocarrier …

Exploring the role of the tumor microenvironment in refractory pituitary tumor pathogenesis.

PURPOSE: Treatment-refractory pituitary tumors demonstrate characteristics resembling those of highly aggressive tumors, in which the local tumor microenvironment (TME) plays a dominant role in promoting aggressiveness and refractoriness. However, role of the TME in pituitary tumors is not well studied. METHODS: Literature on the TME and development of refractory pituitary tumors was reviewed RESULTS: TME harbors tumorigenic immune cells, cancer-associated fibroblasts (CAF), extracellular matrix, and other factors that have been shown to affect behavior of tumor tissue. For example, tumor-ass…

Myeloid-derived suppressor cells: Roles in the tumor microenvironment and tumor radiotherapy.

Cancer progression is closely related to the tumor microenvironment in which the tumor exists, including surrounding blood vessels, immune cells, fibroblasts, bone marrow-derived inflammatory cells, signaling molecules and the extracellular matrix. Tumors can influence the microenvironment by releasing extracellular signals, promoting tumor angiogenesis and inducing peripheral immune tolerance, while the immune cells in the microenvironment can impact the growth and evolution of cancerous cells. One of major cell components in the tumor microenvironment is myeloid-derived suppressor cells (MDS…

Nanozymes in Tumor Microenvironment Modulation for Cancer Therapy.

The tumor microenvironment (TME) plays a central regulatory role in malignant tumor progression and therapeutic resistance, and its complexity and dynamic heterogeneity markedly limit the efficacy of current treatment strategies. Therefore, the development of novel functional materials capable of effectively remodeling the TME and alleviating immunosuppression and therapeutic tolerance has become an important research direction in cancer therapy. In recent years, nanozymes, as a class of nanomaterials with enzyme-mimicking catalytic activity, have shown broad potential in TME intervention and …

ARF beyond p53: Shaping the Pancreatic Tumor Microenvironment.

ARF (p14ARF) is a tumor suppressor classically known for stabilizing and activating p53. In this issue of Cancer Research, Ferreira and colleagues provide insights into the role of ARF in the development of pancreatic ductal adenocarcinoma, showing its p53-independent tumor microenvironment regulatory function. The findings reveal a significant association of ARF loss with stromal remodeling characterized by increased extracellular matrix deposition and fibroblast infiltration. Although further studies are needed to explore the underlying mechanisms and to determine clinical significance, this…

T Cell Dysfunction in the Acidic Tumor Microenvironment.

Solid tumors are characterized by profound metabolic and vascular abnormalities that generate a hostile tumor microenvironment (TME) marked by extracellular acidosis, hypoxia, and nutrient deprivation. While the consequences of these conditions for cancer cell behavior have been extensively studied, their impact on anti-tumor immune responses-particularly T cell function-has only recently gained attention. In this review, we summarize and critically discuss current knowledge on how acidic TME conditions affect the cytotoxic CD8+ T cells which are essential for anti-tumor immunity, and the prot…

Immune Microenvironment of Hair Follicles in Tumor Immunotherapy.

Although tumor immunotherapy has shown significant efficacy against solid tumors, it frequently causes cutaneous and follicular adverse events resulting from disruption of the hair follicle immune microenvironment. This microenvironment is maintained through careful regulation by immune cells and cytokine networks to support normal hair cycling. Immune checkpoint inhibitors disturb this balance by activating T cells, promoting macrophage polarization toward pro-inflammatory phenotypes, upregulating inflammatory cytokines such as tumor necrosis factor-alpha, and activating Toll-like receptor/nu

Profiling tumor immune microenvironment of epithelial ovarian carcinoma.

BACKGROUND: Epithelial ovarian carcinoma (EOC) comprises five main histological subtypes: high-grade serous (HGSOC), low-grade serous (LGSOC), clear cell (CCOC), mucinous (MOC), and endometrioid (ENOC). Each histotype harbors specific genomic alterations and clinical outcome. Few studies systematically compared the tumor immune microenvironment across the five subtypes. METHODS: We performed 7-plex (CD45, CD8, CD68, CD163, FoxP3, CD20, and cytokeratin) sequential immunohistochemistry on a clinically annotated tissue microarray including 139 EOC representing the five subtypes and 26 borderline
