Exploring the Nicotine-Parkinson's disease link - Insights from the UK Biobank.

INTRODUCTION: Evidence suggests an inverse correlation between smoking and Parkinson's disease (PD), yet the mechanisms remain unclear. This study examines the changing risk relationship between smoking and PD diagnosis using alcohol consumption, another reward-driven behavior, as a comparative measure. METHODS: A nested case-control study was conducted using the UK Biobank (UKBB) database. Participants in the prediagnostic phase of PD were identified, and self-reported data on tobacco and alcohol use were analyzed employing conditional binary logistic regression. Polynomial and piece-wise re…

Autoimmune diseases and new-onset atrial fibrillation: a UK Biobank study.

AIMS: The underlying mechanisms of atrial fibrillation (AF) are largely unknown. Inflammation may underlie atrial remodelling. Autoimmune diseases, related to increased systemic inflammation, may therefore be associated with new-onset AF. METHODS AND RESULTS: Participants from the population-based UK Biobank were screened for rheumatic fever, gastrointestinal autoimmune diseases, autoimmune diseases targeting the musculoskeletal system and connective tissues, and neurological autoimmune diseases. Between 2006 and 2022, participants were followed for incident AF. Cox proportional hazards regre…

IVIg-exposure and thromboembolic event risk: findings from the UK Biobank.

BACKGROUND: Arterial and venous thromboembolic events (TEEs) have been associated with intravenous Ig use, but the risk has been poorly quantified. We aimed to calculate the risk of TEEs associated with exposure to intravenous Ig. METHODS: We included participants from UK Biobank recruited over 3 years, data extracted September 2020.The study endpoints were incidence of myocardial infarction, other acute ischaemic heart disease, stroke, pulmonary embolism and other venous embolism and thrombosis.Predictors included known TEE risk factors: age, sex, hypertension, smoking status, type 2 diabete…

Investigating Pleiotropy Between Depression and Autoimmune Diseases Using the UK Biobank.

BACKGROUND: Epidemiological studies report increased comorbidity between depression and autoimmune diseases. The role of shared genetic influences in the observed comorbidity is unclear. We investigated the evidence for pleiotropy between these traits in the UK Biobank (UKB). METHODS: We defined autoimmune and depression cases using hospital episode statistics, self-reported conditions and medications, and mental health questionnaires. Pairwise comparisons of depression prevalence between autoimmune cases and controls, and vice versa, were performed. Cross-trait polygenic risk score (PRS) ana…

Neurological diseases and COVID-19: prospective analyses using the UK Biobank.

COVID-19 (Coronavirus disease-19) may present with neurological signs, but whether people already affected by neurological conditions are at a higher risk of contracting COVID-19 is still not known. We, therefore, aimed to investigate the association of previously diagnosed neurological conditions with COVID-19. 502,536 community-dwelling UK Biobank participants (54.4% male, mean age 56.6 ± 10.3 years) were included. Among these, 57,463 participants had a diagnosis of neurological conditions (11.43%) and a total of 1326 COVID-19-positive cases were identified (0.26%). Neurological conditions w…

Clinical and Genetic Characteristics of Covid-19 Patients from UK Biobank.

We conducted an analysis of 669 Covid-19 positive patients within the UK Biobank cohort, a prospective cohort including over 500,000 participants. Our analyses led to several findings. We found that black participants in the cohort were over four times more likely to be diagnosed with Covid-19 than white participants. In order to assess for confounding, we produced - to our knowledge - the first multivariable adjusted estimate of the association of racial characteristics with Covid-19. Our adjusted estimates indicated that black participants remained at over threefold increased risk of Covid-1…

Factors associated with sarcopenia: A cross-sectional analysis using UK Biobank.

INTRODUCTION: The critical sociodemographic, lifestyle and diseases factors influencing sarcopenia, defined by the current European Working Group on Sarcopenia 2 (EWGSOP2) classification and cut-off points, have not yet been fully elucidated. This study aimed, therefore, to determine sociodemographic, anthropometric, lifestyle and health-related factors associated with sarcopenia using the new EWGSOP2 definition. STUDY DESIGN: 396,283 participants (52.8 % women, age 38-73 years) were included in this cross-sectional study. The potential factors associated with sarcopenia were allocated to fou

Sarcopenia, long-term conditions, and multimorbidity: findings from UK Biobank participants.

BACKGROUND: Sarcopenia, the loss of muscle strength and mass, predicts adverse outcomes and becomes common with age. There is recognition that sarcopenia may occur at younger ages in those with long-term conditions (LTCs) as well as those with multimorbidity (the presence of two or more LTCs), but their relationships have been little explored. Our aims were to describe the prevalence of sarcopenia in UK Biobank, a large sample of men and women aged 40-70 years, and to explore relationships with different categories of LTCs and multimorbidity. METHODS: We used data from 499 046 participants in…

A phenome-wide structural variant map from 467,152 UK Biobank genomes.

Structural variants (SVs) are large (>50 base pairs) genomic rearrangements that can significantly influence human phenotypes, yet their contribution to health and disease remains undercharacterized. We conducted a phenome-wide association study (PheWAS) of 1.6 million SVs identified in whole-genome sequencing data from 467,152 UK Biobank participants, across 13,322 binary, 2,018 quantitative, and 2,923 plasma protein traits. Of these, 82.25% of SVs were outside the detectable range of prior copy-number variant studies. Protein quantitative trait loci analysis linked 3,467 SVs to 1,690 protein…

Fibromyalgia Subgroups Identified Using Model-Based Clustering: Results from UK Biobank.

Fibromyalgia (FM) is a heterogeneous chronic condition with wide variability in symptom burden, psychosocial distress, and functional impact. This study aimed to identify latent FM subgroups reflecting multidimensional patient profiles using comprehensive biopsychosocial variables and model-based clustering methods within UK Biobank. Individuals meeting the 2016 American College of Rheumatology FM criteria (n = 4,172) were analysed using Gaussian mixture clustering. Twenty-eight variables across sociodemographic, clinical symptoms, and psychological/behavioural domains were included. Cluster s…

Cardiovascular Health Protects against Thyroid Dysfunction: Findings from the UK Biobank.

BACKGROUND: Few studies have investigated the association between cardiovascular health (CVH) and thyroid dysfunction, and the modifying role of depression or anxiety remains unclear. METHODS: This longitudinal study included 360,332 UK Biobank participants without thyroid disease at baseline and with complete CVH scores. We examined the associations between CVH and the risks of hyperthyroidism and hypothyroidism using Cox proportional hazards regression models, quantified the relative contributions of CVH components using weighted quantile sum regression, and evaluated the mediating role of …

Self-Report Alzheimer's Disease Statuses in UK Biobank Distort Downstream Analyses.

Genetic studies have identified Alzheimer's disease (AD)-linked variants through genome-wide association studies (GWAS) and proxy-based GWAS (GWAX), yet inconsistent causal inferences still persist. Here, we systematically evaluated how self-reported AD diagnoses in the UK Biobank (UKB) distort Mendelian randomization (MR) analyses. Using seven AD datasets (four GWAS and three GWAX, including IGAP, N = 63 926; FinnGen R10, N = 191 061; UKB G30, N = 420 531; UKB2024, N = 434 286, and GWAX2017, N = 74 366; GWAX2018, N = 548 955; GWAX2021, N = 408 691) and six education subtypes (including years …

Sex disparities in bladder cancer diagnosis: evidence from the UK Biobank.

PURPOSE: To evaluate sex- and age-specific differences in the predictive value of hematuria and urinary tract infections (UTIs) for bladder cancer, and their association with disease invasiveness and mortality. METHODS: We analyzed 4,848 bladder cancer cases and matched controls from the UK Biobank cohort. Logistic regression with Firth penalization was used to assess hematuria and UTIs as predictors of bladder cancer across sex and age groups (< 50, 50–64, and ≥ 65 years). Secondary analyses evaluated associations with tumor invasiveness and all-cause mortality among cases. RESULTS: Hematuria…

Glucosamine Use and Risk of Colorectal Cancer: Results from UK Biobank.

BACKGROUND: Use of the dietary supplement glucosamine has been associated with reduced risk of colorectal cancer; however, it remains unclear if the association varies by screening status, time, and other factors. METHODS: We therefore evaluated these questions in UK Biobank. Multivariable-adjusted HRs and 95% confidence intervals (95% CI) were estimated using Cox proportional hazards regression. RESULTS: No association was observed between use of glucosamine and risk of colorectal cancer overall (HR = 0.94; 95% CI, 0.85-1.04). However, the association varied by screening status (Pinteractio…

Polygenic risk in Type III hyperlipidaemia and risk of cardiovascular disease: An epidemiological study in UK Biobank and Oxford Biobank.

BACKGROUND: Type III hyperlipidaemia (T3HL) is characterised by equimolar increases in plasma triglycerides (TG) and cholesterol in <10% of APOE22 carriers conveying high cardiovascular disease (CVD) risk. We investigate the role of a weighted triglyceride-raising polygenic score (TG.PS) precipitating T3HL. METHODS: The TG.PS (restricted to genome-wide significance and weighted by published independent effect estimates) was applied to the Oxford Biobank (OBB, n = 6952) and the UK Biobank (UKB, n = 460,037), to analyse effects on plasma lipid phenotypes. Fasting plasma lipid, lipoprotein bioch…

Mild ketosis, metabolic syndrome and genetic-lifestyle interactions in the UK Biobank.

The relationship between mild ketosis and metabolic syndrome (MetS) remains unclear. We aimed to investigate the association between serum ketone levels and MetS and to examine how genetic and lifestyle factors influence this relationship. We conducted a cross-sectional observational study using data from the UK Biobank, comprising 269 178 participants. Participants were categorised into low and high serum ketone groups based on β-hydroxybutyrate levels (cut-off: 0·12 mM). Dietary patterns were assessed using validated questionnaires, and a polygenic risk score (PRS) was generated to examine g…
