Vaccinia Virus Vector Bivalent Norovirus Vaccine.

Norovirus is a major etiological agent of nonbacterial gastroenteritis around the world. Due to its in vitro culture complexity, high genome diversity, and the lack of cross-reactive immunity between genogroups, there is an unmet urgent need for polyvalent norovirus vaccines that provide broad-spectrum protection, and no vaccine has gained global approval to date. In this study, we constructed a bivalent norovirus vaccine, based on the highly attenuated poxvirus [strain VG9] vector, expressing the major capsid protein VP1 from genotypes GII.4 and GII.17. VG9-NOR exhibited a comparable replicat

Palisade structure in intact vaccinia virions.

Vaccinia virus assembly in the cytoplasm of infected cells involves the formation of a biconcave viral core inside the maturing viral particle. The boundary of the core is defined by a pseudohexagonal palisade layer, composed of trimers projecting from an inner wall. To understand the assembly of this complex core architecture, we obtained a subnanometer structure of the palisade trimer by cryo-electron tomography and subtomogram averaging of purified intact virions. Using AlphaFold2 structure predictions, we determined that the palisade is formed from trimers of the proteolytically processed …

Counselling of workers handling vaccinia virus.

Vaccination with smallpox vaccine for workers handling vaccinia virus was universally recommended in the United Kingdom until 1990. Since 1990, occupational physicians and other doctors acting as supervisory medical officers have been advised to consider the need for vaccination on a case-by-case basis taking into account factors such as the virulence of the organism and the nature of the proposed work. This paper describes the practice in one organization for the counselling of prospective vaccinia workers and covers the hazards and risks of the work, the main control measures and the advanta…

Modified vaccinia virus Ankara immunization protects against lethal challenge with recombinant vaccinia virus expressing murine interleukin-4.

Recent events have raised concern over the use of pathogens, including variola virus, as biological weapons. Vaccination with Dryvax is associated with serious side effects and is contraindicated for many people, and the development of a safer effective smallpox vaccine is necessary. We evaluated an attenuated vaccinia virus, modified vaccinia virus Ankara (MVA), by use of a murine model to determine its efficacy against an intradermal (i.d.) or intranasal (i.n.) challenge with vaccinia virus (vSC8) or a recombinant vaccinia virus expressing murine interleukin-4 that exhibits enhanced virulenc…

Use of a recombinant vaccinia virus expressing interferon gamma for post-exposure protection against vaccinia and ectromelia viruses.

Post-exposure vaccination with vaccinia virus (VACV) has been suggested to be effective in minimizing death if administered within four days of smallpox exposure. While there is anecdotal evidence for efficacy of post-exposure vaccination this has not been definitively studied in humans. In this study, we analyzed post-exposure prophylaxis using several attenuated recombinant VACV in a mouse model. A recombinant VACV expressing murine interferon gamma (IFN-γ) was most effective for post-exposure protection of mice infected with VACV and ectromelia virus (ECTV). Untreated animals infected with …

Persistence of vaccinia at the site of smallpox vaccination.

Persistence of vaccinia at vaccination sites may help determine the risk associated with secondary transmission. Culture, PCR, and antigen detection were performed on serial vaccination site swab specimens. On day 21 after vaccination, 37% of volunteers were culture positive, most of whom had received vaccine for the first time. Vaccinia is detectable at least through day 21 after vaccination.

Risks associated with vaccinia virus in the laboratory.

Vaccinia virus (VACV) is used commonly in research laboratories. Non-highly attenuated strains of VACV are potentially pathogenic in humans, and VACV vaccination and biosafety level 2 facilities and protocols are currently recommended for vaccinated laboratory workers in the United States who handle non-highly attenuated strains of the virus. Despite this, laboratory-related VACV exposures continue to occur and a number of recent instances of VACV infection in non-vaccinated laboratory workers have been documented. We provide a discussion of the usage and risks associated with VACV in laborato…

Delay of chemotherapy to prevent progressive vaccinia.

OBJECTIVE: The goal was to discuss the potential risk of progressive vaccinia in the setting of smallpox vaccination with immunosuppression and to present strategies to avoid progressive vaccinia. METHODS: A case report and literature review are presented. RESULTS: A 21-year-old, male, military member received smallpox vaccination and was coincidentally diagnosed as having osteosarcoma approximately 2 weeks later. His recent vaccination was recognized, and chemotherapy was subsequently delayed until separation of the scab at the vaccination site. The patient received neoadjuvant chemotherapy…

Generalized vaccinia in a deployed military member.

This case report demonstrates the clinical progression of a case of lesions consistent with generalized vaccinia after primary vaccination in an otherwise healthy adult. The photographs document the appearance and natural course. The progression of the lesions is discussed, documenting the natural progression of the disease. This case report includes photographs of the oral lesions consistent with generalized vaccinia. Oral generalized vaccinia lesions are not well documented in the current medical literature.

[Generalized vaccinia in a child with acute leukemia].

The authors present a girl suffering from acute lymphoblastic leukaemia, which was diagnosed, after she was given the antivariolic vaccina. The clinical flow was very rapid, and was in the fulminant shape, which was presented in from of generalized vaccinia. Although the adequate therapy was given, according to the protocol for the treatment of ALL(08LA74), the child died on for the eight day of hospitalization. There was the evident immunodeficiency in our patient, and it is know that the vaccina with the alive viruses is contraindicated in the imunodeficietic diseases.

Mechanism of poly(A) synthesis by vaccinia virus.

Data are presented which indicate that vaccinia DNA does not contain poly(dT) sequences the size of poly(A) sequences (50 to 200 nucleotides in length) found in vaccinia RNA. A hybridization experiment and polyacrylamide gel electrophoresis and DEAE-Sephadex chromatography of pyrimidine tracts show that poly(dT) sequences can account for no more than 0.1% of vaccinia DNA. Ultraviolet irradiation (which causes thymine dimer formation) and phleomycin (which binds to thymidine) both inhibit RNA synthesis but not poly(A) synthesis by vaccinia cores. These data are consistent with a nontranscriptiv…

Necropsy findings in a case of progressive vaccinia.

A case of progressive vaccinia complicating chronic lymphocytic leukaemia is reported. At necropsy a vaccinial pneumonia, focal pancreatitis, and evidence of disseminated intravascular coagulation were found. Epithelial proliferation was noted in sweat glands, bronchi, and pancreatic ducts associated with lesions in these sites. The significance of these findings is discussed.
