Carcinogenicity of vinyl chloride in Sprague-Dawley rats after prenatal and postnatal exposure.

Vinyl chloride was administered by inhalation, 7 hours daily, 5 days weekly, at concentrations of 2500 and 0 ppm, to Sprague-Dawley rats. The treatment was started on 13-week-old breeders and male and female offspring (12-day embryos). The breeders and part of the offspring were exposed for 104 weeks; the other part of the offspring was exposed for 15 weeks only. Under the experimental conditions, vinyl chloride caused an exceptionally high incidence of brain neuroblastomas, liver angiosarcomas, and hepatocarcinomas. The age at start and/or length of treatment may affect the onset of these tum…

Vinyl-chloride-induced liver disease. From idiopathic portal hypertension (Banti's syndrome) to Angiosarcomas.

Histologic examination of liver tissue (eight autopsy and 18 biopsy specimens) and five spleens from 20 workers with vinyl chloride polymerization showed hepatic angiosarcomas in 15. In addition, a peculiar pattern of progressive portal-tract, inconspicuous intralobular and conspicuous capsular fibrosis was observed in the five workers without angiosarconma, in all the seven patients with angiosarcoma from whom tumor-free portions of the liver were available, and in two tumor-free biopsies from patients subsequently found to have angiosarcoma. The fibrosis was accompanied by splenomegaly. Hype…

Hepatotoxicity of vinyl chloride and 1,1-dichloroethylene. Role of mixed function oxidase system.

Vinyl chloride, an occupational carcinogen, produces acute liver injury in rats pretreated with phenobarbital or Aroclor 1254. Injury appears related to morphologic changes in the endoplasmic reticulum. The degree of injury, as indicated by elevation of serum enzymes derived from the liver, correlates with the magnitude of induction of cytochrome P-450 and its reduction by NADPH. Hepatic injury following 1,1-dichloroethylene exposure differs strikingly from that caused by vinyl chloride and appears to involve plasma membranes, mitochondria, and chromatin and spares endoplasmic reticulum. Induc…

Friedel-Crafts Reaction with Polyolefins as Alkylation Reagents and Poly(vinyl chloride) as Initiator.

Chemical upcycling of waste plastic has attracted substantial attention. While current upcycling methods are typically successful in converting single-component plastic waste, conversion of mixed plastics resembling real-world plastic waste with a single catalytic system remains a challenge. In this manuscript, we report an upcycling method suitable for a range of polyolefins (including mixtures of PP, LDPE, HDPE, and PVC, the four most produced plastics), which selectively produces toluene, ethylbenzene, cumene, and propylbenzene in the presence of benzene. The generation of ethylbenzene and

Biodegradation of vinyl chloride and cis-dichloroethene by a Ralstonia sp. strain TRW-1.

An aerobic bacterium, Ralstonia sp. strain TRW-1, that assimilates vinyl chloride (VC) or ethene (ETH) as the sole carbon source was isolated from a chloroethene-degrading enrichment culture. Phylogenetic analysis of 16S rDNA sequence of the isolate revealed almost 99% sequence similarity to Ralstonia pickettii. To our knowledge, this is the first report describing the isolation of a member of Ralstonia that can degrade VC as the growth substrate. The measured growth yield values for VC and ETH were 11.27 and 18.90 g protein/mole, respectively. The estimated half-velocity constant K (m) values…

Effect of polymer concentration on partitioning and molecular recognition in plasticized poly(vinyl chloride).

Mixtures of poly(vinyl chloride) (PVC) with plasticizers have been used in ion-selective electrodes for many years. The same material has proven useful in solid-phase microextraction (SPME), both with and without artificial receptors. We hypothesized that by changing the polymer concentration in plasticized PVC membranes containing artificial receptor from the standard 33 wt %, the selectivity of the extraction of barbiturates over similar molecules could be improved. Partition coefficients and receptor-substrate formation constants of a target species, phenobarbital, in membranes with various…

Gas chromatographic determination of trace amounts of vinyl chloride and dichloroethenes in landfill-gas.

A method for the determination of vinyl chloride (VC) and dichloroethenes (DCE) in gas samples is presented. The analytes are preconcentrated from a gas-volume of up to 20 l on an adsorption tube filled with 1.0 g of a carbon molecular sieve at a flow rate of 80 l/h and are subsequently desorbed with carbon disulfide. Vinyl bromide is added as internal standard to the extract. The analytes are determined as their 1,2-dibromo-derivatives by capillary gas chromatography with electron capture detection. The detection limits have been found to be 82 ng/m(3) = 32 ppt (VC), 190 ng/m(3) = 48 ppt (1,1…

[Various aspects of the clinical and electroencephalographic studies in workers chronically exposed to vinyl chloride].

The paper presents evaluation of the nervous system and bioelectric functioning of the brain (EEG) in 114 workers aged 20-62, employed in significant exposure to vinyl chloride for 1-28 years, on average--7.5 +/- 4.0. Clinical symptoms of the nervous system occurred in form of peripheral--vegetative syndrome with accompanying vasomotor disturbances of Raynaud syndrome type EEG yielded 39 (34,2%) correct and 75 (65,8%) incorrect records. Among incorrect records most frequent (32.5%) were low-voltage and flat records; of this--those with fast spindled activity and frequent changes typical for re…

Chlorinated Poly(vinyl chloride) Stamps with High Adhesion for Origami Folding of 2D Materials.

We demonstrate that a chlorinated poly(vinyl chloride) (CPVC) film containing a high-viscosity plasticizer enables stable three-dimensional (3D) manipulation of two-dimensional materials, including atomic-layer origami. In clear contrast to conventional PVC films, the CPVC film prevents flake detachment and allows precise control of folding, realizing diverse 3D morphologies such as multiply and corrugated structures. To clarify why CPVC outperforms PVC, we quantitatively investigated adhesion behavior using adhesive force measurements during tensile detachment. The CPVC film exhibits both str…

Interaction of vinyl chloride monomer exposure and hepatitis B viral infection on liver cancer.

Vinyl-chloride monomer (VCM), a human carcinogen, has caused angiosarcoma of the liver. Recent studies have shown that VCM exposure is associated with hepatocellular cancer. In Taiwanese studies, the majority of VCM-exposed workers with liver cancer had history of hepatitis B virus (HBV) infection. To determine the role of HBV on the development of liver cancer in the VCM-exposed workers, we conducted a case-control study from a previously established polyvinyl chloride (PVC) cohort consisting of 4096 male workers from six PVC polymerization plants. A total of 18 patients with liver cancer, an…

p53 mutations at A:T base pairs in angiosarcomas of vinyl chloride-exposed factory workers.

Mutations in the p53 tumor suppressor gene are commonly found in the major human cancers and the mutational spectrum in some cancer types is consistent with the genotoxic effects of the associated environmental risk factors. Thus far there is little information on p53 mutations in cancers of factory workers with a history of carcinogen exposure in the workplace. Occupational exposure to vinyl chloride causes liver angiosarcomas (ASL) and also increases the risk of several other cancers. Loss of p53 function in osteo- and fibrosarcomas can occur by two different mechanisms, p53 mutation and amp…

Biotransformation of tetrachloroethylene to trichloroethylene, dichloroethylene, vinyl chloride, and carbon dioxide under methanogenic conditions.

Tetrachloroethylene (PCE) and trichloroethylene (TCE), common industrial solvents, are among the most frequent contaminants found in groundwater supplies. Due to the potential toxicity and carcinogenicity of chlorinated ethylenes, knowledge about their transformation potential is important in evaluating their environmental fate. The results of this study confirm that PCE can be transformed by reductive dehalogenation to TCE, dichloroethylene, and vinyl chloride (VC) under anaerobic conditions. In addition, [14C]PCE was at least partially mineralized to CO2. Mineralization of 24% of the PCE occ…

Soft Artificial Muscle of Poly(vinyl chloride) Gel with an Imperceptible Liquid Metal Electrode.

Electrodes with high electrical conductivity, yet good flexibility and mechanical compliance, are critical for electroactive artificial muscles. Herein, a promising liquid metal (LM) electrode is proposed by transforming eutectic gallium-indium (EGaIn) LM with high surface tension into paste-like LM with solid Ga2O3 shells. The developed compliant LM electrode not only shows high conductivity and negligible additional stiffness but also displays excellent electrical stability during cyclic actuation. Based on the LM electrode, the poly(vinyl chloride) gel (PVCG) artificial muscle developed exh…

Adipose tissue-liver crosstalk during pathologic changes caused by vinyl chloride metabolites in mice.

Volatile organic compounds (VOCs), such as vinyl chloride (VC), can be directly toxic at high concentrations. However, we have shown that 'nontoxic' exposures to VC and its metabolite chloroethanol (CE) enhances experimental non-alcoholic fatty liver disease (NAFLD), suggesting an unpredicted interaction. Importantly, VOC exposure has been identified as a potential risk factor for the development of obesity and its sequelae in humans. As there is a known axis between adipose and hepatic tissue in NAFLD, the impact of CE on white adipose tissue (WAT) inflammation and lipolysis was investigated.…

Acute liver injury by vinyl chloride: involvement of endoplasmic reticulum in phenobarbital-pretreated rats.

A single 6-hr exposure to vinyl chloride monomer (5%) produces extensive vacuolization of centrolobular liver parenchyma and focal midzonal necrosis in the hepatic lobuole in phenobarbital-pretreated rats. Ultrastructurally, vacuolization consists of dilation of cysternae of rough endoplasmic reticulum and in the same cells smooth endoplasmic reticulum coalesces into discreet aggregates resembling denatured membranes. The findings support the hypothesis that vinyl chloride is hepatotoxic because it is converted into a toxic metabolite by components of the mixed function oxidase system of liver…

Mechanism of subchronic vinyl chloride exposure combined with a high-fat diet on hepatic steatosis.

Vinyl chloride (VC) is a common industrial organic chlorine and environmental pollutant. In recent years, the dietary structure of residents especially Chinese has gradually shifted to western dietary patterns. VC aggravates dietary fatty acid-induced hepatic steatosis, but its mechanism is still unclear. And if the risk factors for steatosis persist, more severe diseases such as fibrosis and cirrhosis will occur. Therefore, we studied the effects and mechanisms of VC (160 and 800 mg/m3 ) and its metabolite (chloroacetaldehyde, 2.25, 4.5, and 9 μM) on hepatic steatosis of high-fat diet (HFD)-f…

Bioaugmentation potential of a vinyl chloride-assimilating Mycobacterium sp., isolated from a chloroethene-contaminated aquifer.

An aerobic bacterium, Mycobacterium sp. strain TRW-2 that assimilated vinyl chloride (VC) or ethene (ETH) as the sole carbon source was isolated from a chloroethene-degrading enrichment culture. The strain TRW-2 also degraded cis-dichloroethene (cis-DCE) in mineral salts medium, but only when VC was present as the primary carbon source. However, no degradation of trans-dichloroethene or trichloroethene occurred in either the presence or absence of added VC. The measured growth yield values were 6.53 and 14.1g protein/mol of VC and ETH utilized, respectively. Inoculation by strain TRW-2 in micr…

Epithelioid hemangioendothelioma of the liver following contact with vinyl chloride. Recurrence after orthotopic liver transplantation.

Malignant epithelioid hemangioendothelioma of the liver is a recently recognized and uncommon neoplasm of vascular origin. Few cases have been reported and the treatment remains therefore difficult to define. A relationship to oral contraceptive use has been suggested but no other toxic agents have been incriminated. We report here the first case occurring after a close contact with vinyl chloride. Because of wide hepatic destruction and serious portal hypertension with bleeding varices, the patient underwent orthotopic liver transplantation. After a disease-free interval of 20 months, he died…

Cu(I)-Catalyzed Radical Coupling of Alkenes with Sulfonimidoyl Chlorides: Synthesis of Vinyl Sulfoximines.

A Cu(I)-catalyzed radical coupling was reported for the synthesis of vinyl sulfoximines from sulfonimidoyl chlorides and alkenes under mild, light-free conditions. This method proceeds via a sulfonimidoyl radical intermediate, generated through Cu(I) reduction, and demonstrates broad substrate tolerance, delivering products in up to 84% yield. The transformation could also be efficiently conducted in a practical one-pot manner by generating the sulfonimidoyl chloride in situ from the S(IV) sulfonamide. Moreover, the addition of tBuOCl further improved the yields of vinyl sulfoximines for sever
