Transmission of influenza A viruses.

Influenza A viruses cause respiratory infections that range from asymptomatic to deadly in humans. Widespread outbreaks (pandemics) are attributable to 'novel' viruses that possess a viral hemagglutinin (HA) gene to which humans lack immunity. After a pandemic, these novel viruses form stable virus lineages in humans and circulate until they are replaced by other novel viruses. The factors and mechanisms that facilitate virus transmission among hosts and the establishment of novel lineages are not completely understood, but the HA and basic polymerase 2 (PB2) proteins are thought to play essen…

Viruses and viral proteins.

For more than 30 years X-ray crystallography has been by far the most powerful approach for determining the structures of viruses and viral proteins at atomic resolution. The information provided by these structures, which covers many important aspects of the viral life cycle such as cell-receptor recognition, viral entry, nucleic acid transfer and genome replication, has extensively enriched our vision of the virus world. Many of the structures available correspond to potential targets for antiviral drugs against important human pathogens. This article provides an overview of the current know…

Overview of influenza viruses.

The influenza virus (IV) is still of great importance as it poses an immanent threat to humans and animals. Among the three IV-types (A, B, and C) influenza A viruses are clinically the most important being responsible for severe epidemics in humans and domestic animals. Aerosol droplets transmit the virus that causes a respiratory disease in humans that can lead to severe pneumonia and ultimately death. The high mutation rate combined with the high replication rate allows the virus to rapidly adapt to changes in the environment. Thereby, IV escape the existing immunity and become resistant to…

Viruses and multiple sclerosis.

Multiple sclerosis (MS) is a heterogeneous disease that develops as an interplay between the immune system and environmental stimuli in genetically susceptible individuals. There is increasing evidence that viruses may play a role in MS pathogenesis acting as these environmental triggers. However, it is not known if any single virus is causal, or rather several viruses can act as triggers in disease development. Here, we review the association of different viruses to MS with an emphasis on two herpesviruses, Epstein-Barr virus (EBV) and human herpesvirus 6 (HHV-6). These two agents have genera…

The detection of hepatitis viruses.

The hepatitis viruses, named A-E, cause acute and chronic liver disease depending on the virus. Laboratory-based diagnosis relies on the detection of specific markers of infection for each virus, through the use of appropriate serological tests. In recent years, molecular tests that rely on the amplification of viral nucleic acids have also been employed, not only in confirming active viral replication but also in monitoring antiviral treatment efficacy. Some of these tests that are currently in use in routine diagnostic laboratories are outlined in this chapter.

Viruses and lymphoma/leukaemia.

Viruses of the retrovirus and herpesvirus families are aetiological agents of human leukaemias and lymphomas. The human T-cell leukaemia virus type 1 causes adult T-cell leukaemia and the Epstein-Barr virus is associated with Burkitt's lymphoma, lymphomas in immunosuppressed people, and Hodgkin lymphoma. The discovery of human herpesvirus type 8 has led to the identification of a rare and unusual group of virus-associated lymphoproliferative diseases. Individuals infected with the human immunodeficiency virus are at greatly increased risk of developing lymphoma but here the mechanism of lympho…

Vaccines, viruses, and voodoo.

Vaccinations are invaluable in protection from a wide variety of diseases that can cause substantial morbidity and mortality. Although a rare complication of vaccination, autoimmune disorders represent one of these morbidities. Recently, widespread public concern has arisen from case reports suggesting that--similar to what has been observed after natural viral infections--there might be an association between specific immunizations and autoimmune diseases. Herein we address the biological plausibility of such a connection, focusing particularly on the examples of hepatitis B, rubella, and mea

Respiratory viruses and asthma.

Viral infections have become increasingly recognized as a significant cause of asthma exacerbations, mainly because of improved viral detection techniques. Unfortunately, the ability to specifically treat viral infections and to limit the asthma morbidity associated with these agents has not kept pace with diagnostic technology. This article focuses on current concepts of the epidemiology of viruses in asthma exacerbations, investigations studying the physiologic and immunologic consequences of viral infection, and potential therapies to minimize virally-induced airway hyperresponsiveness. To …

Viruses and childhood arthritis.

The role of viral infections in the aetiology of acute and chronic arthritides of childhood is incompletely understood. The fact that some viruses cause acute arthritis is certain, although in most instances of presumed viral arthritis no agent is identified. The associations of viruses with diseases such as juvenile chronic arthritis (JCA) are limited, and have been difficult to prove with certainty. Rubella, parvovirus B19 and influenza AH2N2 have been shown by culture, serology or epidemiology to be related to at least some cases of JCA in some studies, but not in others. A rationale for pu…

[Viruses of parasitic protozoa].

The authors present the actual review on several publications concerning the molecular characterizations of the viruses found in parasitic protozoa such as Giardia, Trichomonas, Leishmania and Entamoeba histolytica. All of the RNA viruses observed in parasitic protozoa showed several similarities and did not considerably differ from the viruses found in simple eukaryotic cells; they closely correspond to dsRNA viruses of yeast. The supposition that the protozoan symbionts detected in laboratories transfer to their hosts in natural conditions seemed to be rational, though, there are no evidence…

Airborne bacteria and viruses.

Coughing, sneezing, talking, bed-making, turning pages of books, etc. all generate microbial aerosols which are carried and dispersed by air movements. Inhalation of these particles may cause allergic responses but whether or not infectious disease ensues depends in part on the viability and infectivity of the inhaled microbes and their landing sites. Desiccation is experienced by all airborne microbes; gram-negative bacteria and lipid-containing viruses demonstrate phase changes in their outer phospholipid bilayer membranes owing to concomitant changes in water content and/or temperature. The…

Viruses and human cancer.

The cause of human cancer is probably multifactorial and the role of viruses is unclear. The study of retroviruses has led to the identification of oncogenes responsible for transformation and tumor induction. Human viruses associated with malignancies include the JC virus (associated with progressive multifocal leukoencephalopathy) and some adenoviruses. No human malignancies have been associated with the latter group. A number of herpes viruses of lower animals have been associated with malignancies and herpes simplex virus type 2 has been associated with carcinoma of the cervix and vulva. T…
