Chronic treatment with novel brain-penetrating selective NOP receptor agonist MT-7716 reduces alcohol drinking and seeking in the rat.

Since its discovery, the nociceptin/orphanin FQ (N/OFQ)-NOP receptor system has been extensively investigated as a promising target to treat alcoholism. Encouraging results obtained with the endogenous ligand N/OFQ stimulated research towards the development of novel brain-penetrating NOP receptor agonists with a pharmacological and toxicological profile compatible with clinical development. Here we describe the biochemical and alcohol-related behavioral effects of the novel NOP receptor agonist MT-7716. MT-7716 has high affinity for human NOP receptors expressed in HEK293 cells with a Ki valu

Prevalence of alcohol use before and during pregnancy and predictors of drinking during pregnancy: a cross sectional study in Sweden.

BACKGROUND: There is a paucity of research on predictors for drinking during pregnancy among women in Sweden and reported prevalence rates differ considerably between studies conducted at different antenatal care centres. Since this knowledge is relevant for preventive work the aim of this study was to investigate these issues using a multicenter approach. METHODS: The study was conducted at 30 antenatal care centers across Sweden from November 2009 to December 2010. All women in pregnancy week 18 or more with a scheduled visit were asked to participate in the study. The questionnaire include

What non-alcohol drugs are used by drinking drivers in Sweden? Toxicological results from ten years of forensic blood samples.

INTRODUCTION AND METHOD: Using a forensic toxicology database (TOXBASE), the toxicological results from 10 years of forensic blood samples from people arrested for driving under the influence of alcohol and/or other drugs were reviewed. RESULTS: Alcohol was the only drug identified in blood in N=35,704 cases at a median blood-alcohol concentration (BAC) of 1.63 mg/g. The mean age (±SD) of these offenders was 40±15years and 89% were male. The median BAC was lower (1.30 mg/g) in drivers who had consumed alcohol and used a prescription drug before driving (N=1,251). The mean age of this group of

Moderating effects of a craving intervention on the relation between negative mood and heavy drinking following treatment for alcohol dependence.

OBJECTIVE: Negative affect is a significant predictor of alcohol relapse, and the relation between negative affect and drinking has been shown to be strongly mediated by alcohol craving. Thus, targeting craving during treatment could potentially attenuate the relation between negative affect and drinking. METHOD: The current study is a secondary analysis of data from the COMBINE study, a randomized clinical trial that combined pharmacotherapy with behavioral intervention in the treatment of alcohol dependence. Our goal in the current study was to examine whether a treatment module that target

Economic impact on health-care costs related to major diseases including HIV/AIDS due to alcohol drinking among Thai populations.

OBJECTIVE: The objective of this study was to estimate health-care costs because of diseases caused by alcohol consumption based on health system's perspective. METHODS: Total direct health-care costs of alcohol drinking were the summation of the costs of alcohol drinking in inpatient and outpatient departments due to chronic diseases and acute conditions using prevalence-based approach. The alcohol-attributable fractions, defined as the proportion of a disease or acute condition in a population attributable to alcohol drinking, were calculated to obtain the number of patients in each disease…

Effects of naltrexone, duloxetine, and a corticotropin-releasing factor type 1 receptor antagonist on binge-like alcohol drinking in rats.

A 'binge' is defined by National Institute on Alcohol Abuse and Alcoholism as an excessive pattern of alcohol drinking that produces blood-alcohol levels (BALs) greater than 0.08 g% within a 2-h period and may or may not be associated with dependence. The purpose of this investigation was to explore the effects of several neuropharmacological agents in an animal model in which outbred rats voluntarily and orally self-administer pharmacologically meaningful alcohol doses that produce BALs >or=0.08 g% in daily limited access two-bottle choice and operant drinking sessions. Rats were trained to s…

Alcohol dehydrogenase 2 His47Arg polymorphism influences drinking habit independently of aldehyde dehydrogenase 2 Glu487Lys polymorphism: analysis of 2,299 Japanese subjects.

Although the functional effect of alcohol dehydrogenase 2 (ADH2) His(47)Arg polymorphism has been elucidated, its effect on habitual drinking remains unknown. Here, we conducted a cross-sectional study in 2,299 nonalcoholic Japanese subjects (989 men and 1,310 women). Drinking status, ethanol consumption, and physical reaction to one glass of beer were examined with regard to ADH2 and aldehyde dehydrogenase 2 (ALDH2) polymorphism. Strength of associations were assessed by age-, sex-, smoking status-, and genotype-adjusted odds ratios and their 95% confidence intervals. ADH2 His/Arg and Arg/Arg

Family History of Problem Drinking is Associated With Reduced Sensitivity of Alcohol Demand to Next-Day Responsibilities: A Replication and Extension.

OBJECTIVE: Murphy et al. (2014) found that university students with a family history (FH) of alcohol problems exhibit a reduced sensitivity of alcohol demand to next-day academic responsibilities. Considering the ongoing replication crisis, the objective of this study was to replicate and extend this research in community adults, including a range of non-academic responsibilities. METHOD: We conducted a secondary analysis of 461 adults who reported past-year alcohol consumption (39% women; mean age = 38.42 years), recruited from Amazon Mechanical Turk. Participants were coded as FH+ (40%) or

Associations of Sensitivity to Reward and Punishment with Alcohol Use Severity in a Trauma-Exposed Community Sample: The Role of Drinking Motives.

Background: Sensitivity to reward (SR), or the likelihood of engagement in positively reinforcing experiences, and sensitivity to punishment (SP), or the tendency to avoid behaviors associated with negative outcomes, are risk and maintenance factors for problematic alcohol use, particularly when proximal motivations for drinking are present. Trauma-exposed adults are at increased risk of engaging in problematic alcohol use, particularly if they are using alcohol to cope with negative emotions. Present Study: Thus, the present study examined, among trauma-exposed community adults, the indirect

Evaluating a drink-counting and a breathalyzer-coupled app for monitoring alcohol use: A comparison with timeline followback and peth biomarker.

AIM: Accurately measuring alcohol consumption remains a challenge. This study aimed to evaluate two app-based methods, one using drink-count logging and one using a breathalyzer, by comparing them to retrospective self-report (Timeline Followback, TLFB) and a biomarker of alcohol use (Phosphatidylethanol, PEth). METHODS: Data were acquired from a randomized controlled trial involving alcohol-dependent adults (n = 110). Standard drinks, drinking days, and heavy drinking days reported via the drink-counting app or breathalyzer, were compared with TLFB data over the 12-week period using Lin's co

On The Ground: Methodological and Ethical Considerations of a Field Study on Alcohol Intoxication and Sexual Misperception Among Drinking Venue Patrons.

OBJECTIVE: Decades of research have been dedicated to the study and prevention of sexual aggression perpetration. Despite robust evidence linking acute intoxication and sexual aggression perpetration, few studies have examined this association in naturalistic contexts like drinking settings. Field studies are needed to inform etiological models and interventions that generalize to the naturalistic environment. The goal of this study is to guide researchers on the logistical and ethical considerations involved in conducting an alcohol field study by detailing the methodology used in a field stu

Brain responses and approach bias to social alcohol cues and their association with drinking in a social setting in young adult males.

Alcohol is mainly consumed in social settings, in which people often adapt their drinking behaviour to that of others, also called imitation of drinking. Yet, it remains unclear what drives this drinking in a social setting. In this study, we expected to see stronger brain and behavioural responses to social compared to non-social alcohol cues, and these responses to be associated with drinking in a social setting. The sample consisted of 153 beer-drinking males, aged 18-25 years. Brain responses to social alcohol cues were measured during an alcohol cue-exposure task performed in an fMRI scan

Drink wise, age well; reducing alcohol related harm among people over 50: a study protocol.

BACKGROUND: Evidence suggests that the use of alcohol among older adults (defined as those aged 50+) has increased in recent years, with people aged 55-64 now more likely to exceed the recommended weekly guidelines than any other age group. METHODS/ DESIGN: This is a quasi-experimental study with a before-after design. A postal questionnaire will be sent to 76,000 people aged 50 and over registered with a general practice in five different 'demonstration' (intervention) and control areas in the UK. Multiple interventions will then be delivered in demonstration areas across the UK. At the end

Co-ingestion of energy drinks with alcohol and other substances among a sample of people who regularly use ecstasy.

INTRODUCTION AND AIMS: Despite the potential harms of mixing unregulated drugs with energy drinks (ED), research to date has primarily been focused on EDs co-ingested with alcohol. Consequently, the aim of the present study was to explore the rate of use, harms and correlates of EDs co-ingested with alcohol and other drugs among a sample of people who regularly use illicit stimulant drugs. DESIGN AND METHODS: In 2010, 693 Australians who regularly used ecstasy completed a 1-h interview about their past six-month ED and drug use. RESULTS: Three-quarters of the sample (77%) had recently consum

The small G protein H-Ras in the mesolimbic system is a molecular gateway to alcohol-seeking and excessive drinking behaviors.

Uncontrolled consumption of alcohol is a hallmark of alcohol abuse disorders; however, the central molecular mechanisms underlying excessive … tra-NAc infusion of the farnesyltransferase inhibitor, FTI-276, produced a robust decrease of rats' alcohol drinking; however, sucrose consumption was unaltered. Finally, intra-NAc infusion of FTI-276 also resulted in an attenuation of seeking for alcohol. Together, these results position H-Ras as a central molecular mediator of alcohol's actions within the mesoli …

Effects of neuropeptide Y on appetitive and consummatory behaviors associated with alcohol drinking in wistar rats with a history of ethanol exposure.

BACKGROUND: Neuropeptide Y (NPY) reduces ethanol intake under free access conditions in Wistar rats with a history of prolonged ethanol vapor exposure. The current study was designed to determine whether NPY differentially alters ethanol-associated appetitive behavior (i.e., lever pressing) or ethanol consumption in Wistar rats with a history of ethanol vapor exposure. METHODS: Wistar rats were first trained to self-administer 10% ethanol in a paradigm that provided 25 min of free access to 10% ethanol after completing a 20-lever press response requirement (i.e., an RR20 schedule). After stab

Fetal exposure to alcohol as evidenced by fatty acid ethyl esters in meconium in the absence of maternal drinking history in pregnancy.

The detection of fatty acid ethyl esters (FAEE) in neonatal meconium has been proposed as a novel screening method for intrauterine exposure to alcohol. We investigated the potential use of meconium FAEE screening in a high-risk neonatal population in the absence of maternal drinking history. One hundred forty-two meconium samples of neonates suspected of intrauterine illicit substance exposure and referred to the Motherisk Laboratory were analyzed for the existence of drugs by enzyme-linked immunosorbent assay (ELISA) and FAEE by gas chromatography-flame ionization detection (GC-FID).
