Hypothalamic CART and serum leptin levels are reduced in the anorectic (anx/anx) mouse.

Cocaine- and amphetamine-regulated transcript (CART) is expressed in the hypothalamus, and putative peptides encoded by CART potently inhibit feeding when administered centrally. CART is strongly down-regulated in the lateral hypothalamic area and the arcuate nucleus in animal models of obesity with disrupted leptin signaling. Here we have used in situ hybridization and immunohistochemistry to study CART expression in mice homozygous for the anorexia (anx) mutation which are characterized by a much reduced food intake and premature death. anx/anx mice had significantly decreased levels of CART

Experimental studies of osteopetrosis in laboratory animals.

Osteopetrosis is a metabolic bone disease characterized by a systemic increase in skeletal mass. It results from a defect in the production or function of osteoclasts and is inherited in nine genetically distinct osteopetrotic animal mutations and man. Studies of these mutations have revealed that osteopetrosis is a complex, heterogeneous disorder in its expression, etiology, and response to treatment by bone marrow transplantation or by hormone/growth factor therapy. These animal mutations have been valuable tools for probing the pathogenesis and treatment of osteopetrosis, and information ob

Biologic effects of auto emissions. I. Exhaust from engine with and without catalytic converter.

This paper relates to the efficacy of a catalytic converter in reducing the levels of certain pollutants emitted from an automobile engine and to the reduction and/or elimination of gross biologic damages in animals exposed to emissions from an exhaust system containing such catalysts. Groups of rats were exposed to diluted emissions from an automobile engine with and without catalyst. Concomitantly, a comparative experiment was conducted by exposing a group of rats to carbon monoxide alone (575 mg/m3). The parameters measured included hematocrit, serum LDH, GOT, and lysozyme. An elevation in

Broad-Specificity Aptamer of Sulfonamides: Isolation and Its Application in Simultaneous Detection of Multiple Sulfonamides in Fish Sample.

Sulfonamide antibiotics (SAs) are widely used in animal husbandry and aquaculture, and the excess residues of SAs in animal-derived foods will harm the health of consumers. In reality, various SAs were alternately used in animal husbandry and aquaculture, and thus, it is urgent need to develop simple and high-throughput methods for simultaneously detecting multiple SAs or groups of SAs in order to realize rapid screening of total SAs residues in animal-derived foods. We herein isolated a broad-specificity aptamer for SAs by using a multi-SAs systematic evolution of ligands by exponential enric

Mice with experimental antiphospholipid syndrome display hippocampal dysfunction and a reduction of dendritic complexity in hippocampal CA1 neurones.

AIMS: The antiphospholipid syndrome (APS) is an autoimmune disease characterized by high titres of auto-antibodies (aPL) leading to thrombosis and consequent infarcts. However, many affected patients develop neurological symptoms in the absence of stroke. Similarly, in a mouse model of this disease (eAPS), animals consistently develop behavioural abnormalities despite lack of ischemic brain injury. Therefore, the present study was designed to identify structural alterations of hippocampal neurones underlying the neurological symptoms in eAPS. METHODS: Adult female Balb/C mice were subjected t

Evaluation of a novel access and closure device for NOTES applications: a transcolonic survival study in the porcine model (with video).

BACKGROUND: Natural orifice transluminal endoscopic surgery (NOTES) is a novel, potentially less invasive alternative to laparoscopic surgery. However, the problems of transluminal access and closure represent significant obstacles to its successful introduction in humans. OBJECTIVE: Our purpose was to evaluate the feasibility and safety of a novel device designed for transluminal access and closure. DESIGN: Experimental endoscopic study of transcolonic incision and closure with use of a prototype device in a survival porcine model. SUBJECTS: Four adult female Yorkshire pigs were used in th

Retrievable puncture anchor traction method for endoscopic ultrasound-guided gastroenterostomy: A porcine study.

BACKGROUND: Endoscopic ultrasound-guided gastroenterostomy (EUS-GE) is an alternative method for the surgical treatment of gastric outlet obstruction, but it is regarded as a challenging technique for endoscopists as the bowel is highly mobile and can tent away. Thus, the technique requires superb skill. In order to improve EUS-GE, we have developed a retrievable puncture anchor traction (RPAT) device for EUS-GE to address the issue of bowel tenting. AIM: To evaluate the feasibility of RPAT-assisted EUS-GE using an animal model. METHODS: Six Bama mini pigs each weighing between 15 and 20 kg

Unraveling the potential and challenges of photosynthetic microalgae for oxygenating engineered tissues.

Hypoxia remains a major barrier to the viability and function of engineered large tissue constructs. Conventional strategies such as oxygen-releasing biomaterials and pre-vascularization have shown partial success, often constrained by scalability and long-term sustainability. Co-culturing photosynthetic microalgae and animal cells offers an alternative by establishing living oxygen factories that locally convert carbon dioxide into oxygen and thus mitigate hypoxia. Despite the promise of this symbiotic approach, inherent challenges remain, including physiological incompatibilities between mic

Current Models to Study the Sporothrix-Host Interaction.

Sporotrichosis is a worldwide distributed subcutaneous mycosis that affects mammals, including human beings. The infection is caused by members of the Sporothrix pathogenic clade, which includes Sporothrix schenckii, Sporothrix brasiliensis, and Sporothrix globosa. The fungus can be acquired through traumatic inoculation of conidia growing in vegetal debris or by zoonotic transmission from sick animals. Although is not considered a life-threatening disease, it is an emergent health problem that affects mostly immunocompromised patients. The sporotrichosis causative agents differ in their virul

Pain Management and Analgesics Used in Small Mammals during Post-Operative Period with an Emphasis on Metamizole (Dipyrone) as an Alternative Medication.

Metamizole (dipyrone) is a widely used non-opioid analgesic in both human and animal medicine. Metamizole's safety has been the topic of numerous opposing debates, given the fact that in certain countries metamizole is frequently used as an over-the-counter (OTC) medicine, while in others it is banned due to the risk of agranulocytosis. Further, small mammals such as rabbits, ferrets, rodents, and hedgehogs have become some of the most common pets present in veterinary practice, and each of these species has specific analgesic needs due to their anatomy and physiology. The key to providing app

Perturbation of the intestinal microbiota of mice infected with Cryptosporidium parvum.

Understanding the interaction between the intestinal microbiota (microbiome) and enteric pathogens is of interest in the development of alternative treatments that do not rely on chemotherapy and do not lead to drug resistance. We undertook research in a rodent model of cryptosporidiosis to assess whether the bacterial gut microbiota is impacted by infection with the protozoan pathogen Cryptosporidium parvum. The profile of the faecal bacterial microbiota in infected and uninfected animals was compared using 16S amplicon sequencing. In four independent experiments, the intestinal microbiota of

Evaluation of Antioxidant and Anthelmintic Properties of Tithonia diversifolia Extracts Against Gastrointestinal Nematode Eggs Using In Vitro Assays.

Parasitic diseases caused by gastrointestinal nematodes (GIN) are considered important limiting factors for livestock production worldwide, as they significantly compromise animal welfare and increase production costs. For years, parasite control has relied exclusively on the use of synthetic drugs. Resistance of GIN to commercial anthelmintics has been increasingly reported throughout the world. Therefore, alternative approaches have emerged for controlling GIN in farm animals, including the exploration of plants with bioactive potential. Tithonia diversifolia can be harvested year-round, and

The Vaginal Microbiota in Pig-Tailed Macaques and Colonization with Neisseria gonorrhoeae.

BACKGROUND: Pig-tailed macaques ( Macaca nemestrina ) may serve as a useful animal model of Neisseria gonorrhoeae (GC) infection, but no such models are currently available. To aid in the development of a macaque model of gonorrhea, we characterized the macaque vaginal microbiota and evaluated the effect of streptomycin treatment on GC colonization. METHODS: Six female pig-tailed macaques were observed for 96 days, with cervicovaginal samples collected every 2 to 3 days. Samples were taken for 1 month before streptomycin treatment to characterize the vaginal microbiota using 16S rRNA gene seq

Palatability of Goat's versus Cow's Milk: Insights from the Analysis of Eating Behavior and Gene Expression in the Appetite-Relevant Brain Circuit in Laboratory Animal Models.

Goat's (GM) and cow's milk (CM) are dietary alternatives with select health benefits shown in human and animal studies. Surprisingly, no systematic analysis of palatability or preference for GM vs. CM has been performed to date. Here, we present a comprehensive investigation of short-term intake and palatability profiles of GM and CM in laboratory mice and rats. We studied consumption in no-choice and choice scenarios, including meal microstructure, and by using isocaloric milks and milk-enriched solid diets. Feeding results are accompanied by qPCR data of relevant genes in the energy balance-

BCG vaccination for bovine tuberculosis; conclusions from the Jerusalem One Health workshop.

The global burden of bovine tuberculosis (bTB) remains poorly characterized, with spill-over impacts on multiple species. The "One Health" concept is especially relevant given the bidirectional risk of cattle infecting humans with Mycobacterium bovis and humans infecting cattle with Mycobacterium tuberculosis. "Test and cull" is the traditional bTB control method, but the strategy may not be economically feasible or culturally acceptable where cattle are highly prized or their killing is a religious taboo; it is also less effective when there are wildlife reservoirs of infection. Vaccination w
