Oncologic Outcomes of Sequential Intravesical Gemcitabine and Docetaxel Compared with Bacillus Calmette-Guérin in Patients with Bacillus Calmette-Guérin-Unresponsive Non-Muscle Invasive Bladder Cancer.

BACKGROUND AND OBJECTIVE: Non-muscle-invasive bladder cancer (NMIBC) patients treated with additional bacillus Calmette-Guérin (BCG) may become unresponsive to BCG. Recently, sequential intravesical gemcitabine and docetaxel (gem/doce) are being used for NMIBC. This study aims to compare oncologic outcomes between sequential intravesical gem/doce versus additional BCG in patients with BCG-unresponsive NMIBC. METHODS: Data were collected from ten academic institutions on patients with BCG-unresponsive NMIBC based on the Food and Drug Administration guidelines. Information on high-grade recurre…

Does switching the bacillus Calmette-Guérin strain affect clinical outcome in patients with recurrent non-muscle-invasive bladder cancer after initial bacillus Calmette-Guérin therapy?

PURPOSE: It is still unknown whether switching the bacillus Calmette-Guérin (BCG) strain at the second induction course of BCG therapy has a therapeutic benefit in patients with tumor recurrence after the initial BCG therapy (BCG-relapsing tumor). MATERIALS AND METHODS: We retrospectively reviewed the clinicopathological features of 97 patients treated with a second induction course of BCG therapy between 1986 and 2014. Among the patients initially treated with BCG Tokyo-172, the second course was either BCG Tokyo-172 in 56 (57.8%) or BCG Connaught in 15 (15.5%). Among those who were initiall…

Secondary cell wall polysaccharides from Bacillus cereus strains G9241, 03BB87 and 03BB102 causing fatal pneumonia share similar glycosyl structures with the polysaccharides from Bacillus anthracis.

Secondary cell wall polysaccharides (SCWPs) are important structural components of the Bacillus cell wall and contribute to the array of antigens presented by these organisms in both spore and vegetative forms. We previously found that antisera raised to Bacillus anthracis spore preparations cross-reacted with SCWPs isolated from several strains of pathogenic B. cereus, but did not react with other phylogenetically related but nonpathogenic Bacilli, suggesting that the SCWP from B. anthracis and pathogenic B. cereus strains share specific structural features. In this study, SCWPs from three st…

The potential to manage releases of Bacillus anthracis using bioretention and a high flow media filter: Results of simulated runoff testing with tracer spores Bacillus globigii.

The threat of bioterrorism has spurred research on the decontamination and containment of different agents. Anthrax [causative agent Bacillus anthracis (Ba)] is a disease that can lead to severe infections within human and animals, particularly when inhaled. This research investigated the use of spore-contaminated simulated runoff events into stormwater control measures (SCMs), which are designed to retain and improve the quality of runoff and may have the potential to filter and contain the spores. In this study, the effectiveness of a bioretention cell (BRC) and high flow media filter (HFMF)…

Risk presented to minimally processed chilled foods by psychrotrophic Bacillus cereus.

BACKGROUND: Spores of psychrotrophic Bacillus cereus may survive the mild heat treatments given to minimally processed chilled foods. Subsequent germination and cell multiplication during refrigerated storage may lead to bacterial concentrations that are hazardous to health. SCOPE AND APPROACH: This review is concerned with the characterisation of factors that prevent psychrotrophic B. cereus reaching hazardous concentrations in minimally processed chilled foods and associated foodborne illness. A risk assessment framework is used to quantify the risk associated with B. cereus and minimally p…

From soil to gut: Bacillus cereus and its food poisoning toxins.

Bacillus cereus is widespread in nature and frequently isolated from soil and growing plants, but it is also well adapted for growth in the intestinal tract of insects and mammals. From these habitats it is easily spread to foods, where it may cause an emetic or a diarrhoeal type of food-associated illness that is becoming increasingly important in the industrialized world. The emetic disease is a food intoxication caused by cereulide, a small ring-formed dodecadepsipeptide. Similar to the virulence determinants that distinguish Bacillus thuringiensis and Bacillus anthracis from B. cereus, the…

Enzymatic and biosurfactant production with antimicrobial activity by Bacillus subtilis group.

Biosurfactants are valuable microbial metabolites with broad industrial and environmental applications. In this study, we investigated biosurfactant production by Bacillus subtilis group strains using residual glycerol as the sole carbon source, while also evaluating their enzymatic activity and antimicrobial properties. PCR-based gene detection confirmed the presence of genes encoding surfactin, fengycin, and iturin peptide biosynthesis. Crude glycerol, a byproduct of biodiesel production, supported biosurfactant synthesis by the tested strains. Strains CCMA 0087, 0658, and 2031 exhibited die…

Identification of MscS as a Key L-Glutamate Exporter in Bacillus methanolicus.

Small-conductance mechanosensitive channels (MscS) are established l-glutamate exporters in industrially relevant bacteria, yet their role in the methylotrophic bacterium Bacillus methanolicus, a promising platform for sustainable methanol-based l-glutamate production, remains unexplored. Our research on B. methanolicus MGA3 identifies its MscS as the sole mechanosensitive channel in this organism and a key exporter of the amino acid l-glutamate, providing valuable insights into its potential for industrial applications. Transcriptomic analysis of B. methanolicus wild type cultured on an l-glu…

Detection of Bacillus production strains and contaminants in food enzyme products.

Shotgun metagenomics enables taxonomic analysis of microbial communities by aligning sequencing reads to reference genomes, for which interpretation of alignment results often lacks standardization and relies on arbitrary abundance thresholds. This can bias species detection, especially for low-abundance or taxonomically complex genera like Bacillus, where closely related species may differ in safety and function, and their co-occurrence increases misclassification risk. This study presents a bioinformatics framework for defining detection thresholds of biological contaminations in samples usi…

Bioremediation and Recovery of Lead and Cadmium by Spores of Bacillus subtilis C1.

Spores of a hot spring isolated strain of Bacillus subtilis were tested as a biotechnological tool to be used for the detoxification and bioremedition of heavy metals. Lead and cadmium were efficiently adsorbed by B. subtilis spores with those of C1 more efficient than those of the lab collection strain PY79. Metal-adsorption did not alter the functionality of C1 spores that were still fully resistant to heat, ethanol or chloroform and able to germinate after the interaction with Cd2+ or Pb2+. The spore-adsorbed metals were released upon disruption of the spore coat layers, suggesting that the…

RagB stimulates the activity of the peptidoglycan polymerase RodA in Bacillus subtilis.

The bacterial cell wall is primarily composed of peptidoglycan (PG), a polymer essential for its protective envelope function, and any defect in its synthesis or repair can potentially result in bacterial lysis. Class A Penicillin-Binding Proteins (aPBPs) and Shape, Elongation, Division, and Sporulation (SEDS) proteins are PG polymerases acting in concert to ensure bacterial cell wall growth. Here, we identify the first regulator of the SEDS protein RodA in the Gram-positive model bacterium Bacillus subtilis. In the presence of the antibiotic moenomycin, which specifically inhibits glycosyltra…

Biosensors and Bioassays for the Medical Detection of Bacillus Anthracis and Diagnosis of Anthrax.

Bacillus anthracis is a causative agent of the highly mortal disease anthrax. This zoonosis is present in nature, but it is also considered one of the most powerful biological warfare agents. A timely diagnosis is necessary for proper therapy and setting of epidemiological countermeasures. Current diagnostic methods should be used in specialized laboratories or medical facilities because there are only a limited number of methods suitable as hand-held assays or even point-of-care tests for detecting B. anthracis or anthrax diagnosis. The lateral flow tests are an exception in this regard, but …

Effects of dipicolinic acid on Bacillus anthracis spore biology and cytotoxicity.

UNLABELLED: Bacillus anthracis is a gram-positive spore-forming bacterium that causes lethal inhalation anthrax. The use of B. anthracis as a bioweapon is predicated in its ability to form dormant and resistant infective spores that can be used as agents. B. anthracis spores contain large concentrations of dipicolinic acid complexed with cations, especially calcium (Ca-DPA). Following phagocytosis by alveolar macrophages, spores germinate inside the phagolysosome and excrete the Ca-DPA depot into the phagosomal space. In this study, we assessed the effects of DPA on B. anthracis spore biology …

Characterization and Molecular Insights of a Chromium-Reducing Bacterium Bacillus tropicus.

Environmental pollution from metal toxicity is a widespread concern. Certain bacteria hold promise for bioremediation via the conversion of toxic chromium compounds into less harmful forms, promoting environmental cleanup. In this study, we report the isolation and detailed characterization of a highly chromium-tolerant bacterium, Bacillus tropicus CRB14. The isolate is capable of growing on 5000 mg/L Cr (VI) in an LB (Luria Bertani) agar plate while on 900 mg/L Cr (VI) in LB broth. It shows an 86.57% reduction ability in 96 h of culture. It can also tolerate high levels of As, Cd, Co, Fe, Zn,…
