WGS reveals high-risk clones of Pseudomonas aeruginosa harbouring extensive antimicrobial and predicted anti-phage defense systems recovered from diabetic foot ulcer patients in Egypt.

Diabetic foot ulcers (DFUs) are a major complication of diabetes mellitus and a leading cause of morbidity, amputation, and mortality among affected patients. In this study, five Pseudomonas aeruginosa isolates recovered from DFUs of diabetic patients in Egypt were investigated. Antimicrobial susceptibility was evaluated using the disc diffusion method and broth microdilution assay, while biofilm-forming ability was phenotypically assessed using the crystal violet assay. All isolates exhibited multidrug-resistant (MDR) phenotypes, with two isolates classified as pan-drug resistant (PDR). Moreo

Phage cocktail amikacin combination as a potential therapy for bacteremia associated with carbapenemase producing colistin resistant Klebsiella pneumoniae.

The increasing occurrence of hospital-associated infections, particularly bacteremia, caused by extensively drug-resistant (XDR) carbapenemase-producing colistin-resistant Klebsiella pneumoniae highlights a critical requirement to discover new therapeutic alternatives. Bacteriophages having host-specific bacteriolytic effects are promising alternatives for combating these pathogens. Among 12 phages isolated from public wastewater in Thailand, two phages-vB_kpnM_05 (myovirus) and vB_kpnP_08 (podovirus) showed broad-host range, producing bacteriolytic activities against 81.3% (n = 26) and 78.1%

Bacteriophage therapy as an alternative technique for treatment of multidrug-resistant bacteria causing diabetic foot infection.

Diabetic foot ulcer (DFU) represented the most feared diabetic complication that caused the hospitalization of the diabetic patient. DFU was usually characterized with delayed healing as the diabetic neuropathy, angiopathy, and ulcer concomitant infections, among them, are multidrug-resistant (MDR) bacteria that emphasized the clinical importance for developing new therapeutic strategy with safe and effective alternatives for the antibiotics to overcome DFU-MDR bacterial infection. Bacteriophage therapy was considered a novel approach to eradicate the MDR, but its role in the polymicrobial inf

Detection of SARS-CoV-2 RNA in bivalve mollusks and marine sediments.

The presence of SARS-CoV-2 in wastewater pose the question of whether this new pandemic virus could be released into watercourses and potentially continue to finally reach coastal waters. In this study, we employed two bivalve molluscan species from the genus Ruditapes as sentinel organisms to investigate the presence of SARS-CoV-2 signals in the marine coastal environment. Estuarine sediments from the natural clam banks were also analyzed. Viral RNA was detected by RT-qPCR, targeting IP4, E and N1 genomic regions. Positive samples were also subjected to a PMAxx-triton viability RT-qPCR assay

Incorporation of membrane-anchored flagellin into Salmonella Gallinarum bacterial ghosts induces early immune responses and protection against fowl typhoid in young layer chickens.

The present study aimed to investigate whether the incorporation of flagellin, a TLR5 agonist, in the bacterial ghosts (BGs) of Salmonella Gallinarum can enhance protective immune responses against fowl typhoid, a septicemic disease of poultry, in chickens. BGs are empty cell envelopes derived from Gram-negative bacteria through the bacteriophage phiX174 gene E mediated lysis. In this study, the S. Gallinarum ghosts carrying flagellin were genetically constructed utilizing a lysis plasmid pJHL184-flagellin, designed for the coexpression of the flagellin and the lysis protein E. The adjuvant ef

Analyses of the Distribution Patterns of Burkholderia pseudomallei and Associated Phages in Soil Samples in Thailand Suggest That Phage Presence Reduces the Frequency of Bacterial Isolation.

BACKGROUND: Burkholderia pseudomallei is a soil saprophytic bacterium that causes melioidosis. The infection occurs through cutaneous inoculation, inhalation or ingestion. Bacteriophages (phages) in the same ecosystem may significantly impact the biology of this bacterium in the environment, and in their culturability in the laboratory. METHODS/PRINCIPAL FINDINGS: The soil samples were analysed for the presence of bacteria using culture methods, and for phages using plaque assays on B. pseudomallei strain 1106a lawns. Of the 86 soil samples collected from northeastern Thailand, B. pseudomalle

Perspectives on super-shedding of Escherichia coli O157:H7 by cattle.

Escherichia coli O157:H7 is a foodborne pathogen that causes illness in humans worldwide. Cattle are the primary reservoir of this bacterium, with the concentration and frequency of E. coli O157:H7 shedding varying greatly among individuals. The term "super-shedder" has been applied to cattle that shed concentrations of E. coli O157:H7 ≥ 10⁴ colony-forming units/g feces. Super-shedders have been reported to have a substantial impact on the prevalence and transmission of E. coli O157:H7 in the environment. The specific factors responsible for super-shedding are unknown, but are presumably media

Cholera toxin - a foe & a friend.

After De΄s pivotal demonstration in 1959 of a diarrhoeogenic exo-enterotoxin in cell-free culture filtrates from Vibrio cholerae (of classical biotype), much insight has been gained about cholera toxin (CT), which is arguably now the best known of all microbial toxins. The subunit structure and function of CT, its receptor (the GM1 ganglioside), and its effects on the cyclic AMP system and on intestinal secretion were defined in the 1970s, and the essential aspects of the genetic organization in the 1980s. Recent findings have generated additional perspectives. The 3D-crystal structure of CT h

Epitope detection in the envelope of intracellular naked orthopox viruses and identification of encoding genes.

Monoclonal antibodies (MAbs) were generated against vaccinia virus, cowpox virus KR2 Brighton, monkeypox virus Copenhagen, or ectromelia virus. Pairwise epitope specificity studies by competition ELISAs identified 23 distinct antigenic sites in 19 different orthopox virus strains. Six epitopes were completely independent of each other, and 17 closely related antigenic sites formed three separate epitope complexes. As shown by immunogold electron microscopy (ELMI), all MAbs reacted with epitopes in the envelope of intracellular naked virus, 16 MAbs recognized proteins of 32, 30, 16 or 14 kDa in

Interventions targeting gut colonization by multi-drug-resistant organisms in healthcare settings: a systematic review and meta-analysis of randomized controlled trials.

Gut colonization with multi-drug-resistant organisms (MDROs) increases risks of infection, transmission and mortality in healthcare settings. Although decolonization strategies have been attempted to reduce the impact of gut MDROs, no consensus exists on their effectiveness. Our study evaluates the effectiveness of these strategies. We searched PubMed, Embase, CENTRAL, CINAHL and Web of Science for randomized controlled trials (RCTs) published from January 2005 to December 2024. Eligible studies included patients in healthcare settings with baseline MDRO colonization confirmed by rectal swab o

PEGylation Overcomes Pharmacological Barriers to Improve Systemic Pharmacokinetics and Therapeutic Efficacy of Phages against MDR Escherichia coli.

Systemic bacteriophage therapy against multidrug-resistant (MDR) Escherichia coli is fundamentally limited by rapid immune-mediated clearance, complement activation, and phagocytic sequestration, collectively constituting pharmacological barriers that restrict systemic bioavailability, shorten circulation half-life, and attenuate therapeutic efficacy. We hypothesized that PEGylation, by sterically shielding phage capsids from host immune clearance mechanisms, would enhance systemic stability, improve pharmacokinetic (PK) behavior, and augment therapeutic efficacy in vivo. Four lytic E. coli ph

Minimal PEGylation Improves Pharmacokinetics, Immunocompatibility, and Efficacy of a Lytic Bacteriophage against Multidrug-Resistant Acinetobacter baumannii.

Multidrug-resistant (MDR) Acinetobacter baumannii presents a critical therapeutic challenge due to its extensive antibiotic resistance and the paucity of effective alternatives. This study evaluated whether minimal PEGylation could enhance the pharmacokinetic performance, immune compatibility, and antibacterial efficacy of the lytic phage vB_AbaSt_W16 in an immunocompetent murine model of systemic infection. The phage vB_AbaSt_W16 was conjugated with methoxy polyethylene glycol succinimidyl ester (mPEG-S-NHS, MW 5000) at a low concentration (4.2 pM), experimentally defined as the minimal PEGyl

Isolation and characterization of bacteriophage against clinical isolates of AmpC beta lactamase-Producing Klebsiella pneumoniae from hospital wastewater.

BACKGROUND: The increasing incidence of AmpC β-lactamase producing by K. pneumoniae has raised global alarm. Consequently, there is a crucial need for effective methods to inactivate pathogenic bacteria and mitigate the associated risks. Bacteriophage therapy has been demonstrated to be an effective and alternative approach for targeting and inactivating K. pneumoniae that produces AmpC. This study aimed to isolate and characterize the Klebsiella pneumoniae AmpC-specific phages from hospital wastewater. METHODS: The hospital wastewater samples were collected from the sewage water effluent of
