In a solvent-free system, Candida rugosa lipase (CRL) catalyzes the transesterification reaction between ethyl ferulate and triolein to yield feruloyl oleoyl glycerol, a lipophilic food antioxidant. However, this reaction is limited by temperature, time, and water activity, posing significant challenges to improving enzymatic efficiency and reaction yield. Inspired by this, this study added the enzyme protector ectoine to the reaction system. The results showed that low concentrations (2.0 mmol/L) of ectoine can significantly enhance the catalytic performance of CRL, achieving a maximum conver

Biocatalytic conversion of CO-containing C1 gas to formate provides a route for C1 gas valorization, but purified carbon monoxide dehydrogenase (CODH)-formate dehydrogenase (FDH) enzyme cascades require enzyme purification and immobilization steps. As a practical alternative, this study developed an ethyl viologen (EV)-mediated intercellular whole-cell cascade by coupling CODH-expressing Escherichia coli with FDH-expressing Methylobacterium extorquens AM1. In this cascade, the two whole-cell biocatalysts performed the oxidative and reductive half-reactions, respectively, and were functionally

Zearalenone (ZEN) lactonases are promising biocatalysts for ZEN detoxification, yet the catalytic mechanism underlying ZEN hydrolysis remains poorly understood. Here, we combined structural analysis, quantum-mechanical (QM) calculations, and molecular dynamics (MD) simulations to elucidate the catalytic mechanism and guide enzyme engineering. QM and MD analyses identified a near-attack conformation of the catalytic His245 as essential for proton transfer. Crystal structure analysis revealed that mutations within the active pocket enhanced the local hydrophobic microenvironment, thereby optimiz

Enzymatic production of low-molecular-weight chitosan and chitooligosaccharides (COS) with consistent properties is challenging since chitosanases are unstable under acidic, high-temperature conditions. To address this, a recombinant chitosanase was covalently attached to glyoxyl-activated agarose via multipoint attachment to improve stability and influence its hydrolysis pattern. The immobilized enzyme showed increased activity and stability at pH 4.5 and 55 °C, where the free enzyme quickly lost activity. Immobilization also reduced temperature sensitivity and enabled sustained catalysis up

Laccases are biocatalysts with a high potential for developing green procedures to achieve new compounds. This study employed laccases in buffer solution to synthesize dimeric neolignans from honokiol and magnolol. Four laccases (LTV, POXA1b, EV3, and EV4) were first screened for honokiol dimerization in the presence of the mediator, with the variant EV4 achieving with the highest yield and in a shorter timeframe the natural houpulin A and B. Reaction conditions were optimized using a mathematical model to maximize the recovery of the desired products, achieving up to 3.42% yield. Interestingl

Phytosterols have long been used to prevent hypercholesterolemia-related diseases, although their effects on intestinal cholesterol metabolism remain elusive. This study applied comparative proteomics to investigate the effects of various sterols on proteomic changes in differentiated Caco-2 cells. A total of 206, 231, 180, and 197 differentially expressed proteins were identified following treatment with cholesterol, β-sitosterol, stigmasterol, and campesterol, respectively. Functional enrichment analysis revealed that phytosterols significantly altered the proteome of Caco-2 cells, though th

Organophosphorus compounds (OPs) poisoning poses a significant health risk as an insecticide, and its potent toxicity is characterized by rapid onset and a very narrow window for intervention. Prophylactic medication is crucial for managing OPs poisoning, yet effective and safe drugs are still lacking. The enzyme Organophosphorus hydrolase (OPH) shows promise as a bioscavenger but faces challenges due to its short half-life and strong immunogenicity. Our research reveals that N-terminal PEGylation of OPH significantly extends its pharmacokinetic half-life, reduces immunogenicity, and, surprisi

Efficient C1 utilization relies on glycolaldehyde (GALD) as a key intermediate, typically formed via condensation of two formaldehyde (FALD) molecules catalyzed by glycolaldehyde synthase (GALS). While catalytic activity improvements have been achieved, limited attention to thermal stability has constrained industrial application. Building on the high catalytic efficiency (153.3 M-1·s-1) variant GALS M3 (T87A/A416T/W463I), we applied computer-aided design strategy to further enhance enzyme stability. After two rounds of systematic protein engineering, variant GALS M5 (GALS M3-A381P/K290P) was

Electrocatalytic carbon dioxide (CO2) reduction by CO2 reductases is a promising approach for biomanufacturing. Among all known biological or chemical catalysts, hydrogen-dependent carbon dioxide reductase from Thermoanaerobacter kivui (TkHDCR) possesses the highest activity toward CO2 reduction. Herein, we engineer TkHDCR to generate an electro-responsive carbon dioxide reductase considering the safety and convenience. To achieve this purpose, a recombinant Escherichia coli TkHDCR overexpression system is established. The formate dehydrogenase is obtained via subunit truncation and rational d

The self-sufficient cytochrome P450 monooxygenase CYP505E3 from Aspergillus terreus catalyzes the regioselective in-chain hydroxylation of alkanes, fatty alcohols, and fatty acids at the ω-7 position. It is the first reported P450 to give regioselective in-chain ω-7 hydroxylation of C10-C16 n-alkanes, thereby enabling the one step biocatalytic synthesis of rare alcohols such as 5-dodecanol and 7-tetradecanol. It shows more than 70 % regioselectivity for the eighth carbon from one methyl terminus, and displays remarkably high activity towards decane (TTN≈8000) and dodecane (TTN≈2000). CYP505E3

Ribose-5-phosphate isomerase (Rpi, EC 5.3.1.6) is widespread in microorganisms, animals, and plants. It has a pivotal role in the pentose phosphate pathway and responsible for catalyzing the isomerization between D-ribulose 5-phosphate and D-ribose 5-phosphate. In recent years, Rpi has received considerable attention as a multipurpose biocatalyst for production of rare sugars, including D-allose, L-rhamnulose, L-lyxose, and L-tagatose. Besides, it has been thought of as a potential drug target in the treatment of trypanosomatid-caused diseases such as Chagas' disease, leishmaniasis, and human

Carotenoids are a large class of structures that are important in human health and include both provitamin A and nonprovitamin A compounds. Vitamin A deficiency is a global health problem that can be alleviated by enriching provitamin A carotenoids in a range of food crops. Suitable plants for biofortification are those with high levels of the provitamin A biosynthetic precursor, lycopene, which is enzymatically converted by lycopene β-cyclase (LCYB) to β-carotene, a provitamin A carotenoid. Crops, such as citrus, naturally accumulate high levels of provitamin A and other health-promoting caro

We report the functional and structural characterization of trehalose-6-phosphate phosphatase (TPP), from the Gram-negative bacterium B. pseudomallei that causes melioidosis, a severe infectious disease endemic in Southeast Asia and Northern Australia. TPP is a key enzyme in the trehalose biosynthesis pathway, which plays an important role in bacterial stress responses. Due to the absence of this biosynthetic pathway in mammals, TPP has drawn attention as a potential drug target, to combat antibiotic resistance. In this context, we present a detailed biochemical analysis of purified recombinan

Transglutaminase 2 (TG2) is a ubiquitous but enigmatic mammalian protein to which a number of biological functions have been ascribed but not definitively proven. As a member of the transglutaminase family, TG2 can catalyze deamidation or alternatively transamidation of selected Gln residues in proteins and peptides. It is also known to harbor other enzymatic properties, including protein disulfide isomerase, GTP-dependent signal transduction, and ATP dependent protein kinase activity. Given its multifunctional chemistry, it is unsurprising that a long list of proteins from the mammalian prote

In a one-way cross-over study, we investigated the effect of Khat, a natural amphetamine-like psychostimulant plant, on catalytic activities of five major drug-metabolizing cytochrome P450 (CYP) enzymes. After a one-week Khat abstinence, 63 Ethiopian male volunteers were phenotyped using cocktail probe drugs (caffeine, losartan, dextromethorphan, omeprazole). Phenotyping was repeated after a one-week daily use of 400 g fresh Khat leaves. Genotyping for CYP1A2, CYP2C9, CYP2C19, CYP2D6, CYP3A5 were done. Macrolactonization of natural product analogs presents a significant challenge to both biosynthetic assembly and synthetic chemistry. In the preceding paper , we identified a thioesterase (TE) domain catalytic bottleneck processing unnatural substrates in the pikromycin (Pik) system, preventing the formation of epimerized macrolactones. Here, we perform molecular dynamics simulations showing the epimerized hexaketide was accommodated within the Pik TE active site; however, intrinsic conformational preferences of the substrate resulted in predominately unproductive conformations, in agreement w

Hypoxanthine-guanine phosphoribosyltransferase (HGPRT) (EC 2.4.2.8) reversibly catalyzes the transfer of the 5-phophoribosyl group from 5-phosphoribosyl-alpha-1-pyrophosphate (PRPP) to hypoxanthine or guanine to form inosine monophosphate (IMP) or guanosine monophosphate (GMP) in the purine salvage pathway. To investigate the catalytic mechanism of this enzyme in the intracellular pathogen Legionella pneumophila, we determined the crystal structures of the L. pneumophila HGPRT (LpHGPRT) both in its apo-form and in complex with GMP. The structures reveal that LpHGPRT comprises a core domain and

Recent work has shown that engineered variants of cytochrome P450BM3 (CYP102A1) efficiently catalyze non-natural reactions, including carbene and nitrene transfer reactions. Given the broad substrate range of natural P450 enzymes, we set out to explore if this diversity could be leveraged to generate a broad panel of new catalysts for olefin cyclopropanation (i.e., carbene transfer). Here, we took a step towards this goal by characterizing the carbene transfer activities of four new wild-type P450s that have different native substrates. All four were active and exhibited a range of product sel

The aim of the present study was to identify cytochrome P450 isoenzymes (CYPs) involved in the 5-sulfoxidation and N-demethylation of the aliphatic-type phenothiazine neuroleptic levomepromazine in human liver. Experiments were performed in vitro using cDNA-expressed human CYP isoforms (Supersomes 1A2, 2A6, 2B6, 2C8, 2C9, 2C19, 2D6, 2E1, 3A4), liver microsomes from different donors and CYP-selective inhibitors. The obtained results indicate that CYP3A4 is the main isoform responsible for levomepromazine 5-sulfoxidation (72%) and N-demethylation (78%) at a therapeutic concentration of the drug
