Polysaccharide-inspired biomaterials for skeletal muscle regeneration: innovations for volumetric muscle loss treatment.

Volumetric muscle loss (VML) is a severe condition caused by extensive damage to muscle tissue that exceeds the body's intrinsic regenerative capacity, resulting in significant functional deficits and long-term disability. Current clinical treatments, including autologous muscle grafts and physical therapy, often fall short due to donor site morbidity, limited functional recovery, and an inability to fully restore muscle structure and function. Polysaccharides as natural macromolecules are characterized by inherent biocompatibility, degradability, and tunable mechanical properties, making them

Improving the bioactivity and mechanical properties of poly(ethylene glycol)-based hydrogels through a supramolecular support network.

Most synthetic hydrogels are formed through radical polymerization to yield a homogenous covalent meshwork. In contrast, natural hydrogels form through mechanisms involving both covalent assembly and supramolecular interactions. In this communication, we expand the capabilities of covalent poly(ethylene glycol) (PEG) networks through co-assembly of supramolecular peptide nanofibers. Using a peptide hydrogelator derived from the tryptophan zipper (Trpzip) motif, we demonstrate how in situ formation of nanofiber networks can tune the stiffness of PEG-based hydrogels, while also imparting shear t

Rapid Gelation of Anti-Swelling, Self-Healing, and Biocompatible PEG Hydrogels Based on CBT-Cys Click Reaction under Mild Conditions.

Hydrogels, essential biomaterials in bioengineering, face challenges of undesirable swelling in applications requiring stable dimensions. This study presents a novel anti-swelling hydrogel using a cyanobenzothiazole (CBT)-cysteine (Cys) click reaction, inspired by luciferin chemistry. By functionalizing 4-Arm PEG with CBT and synthesizing N-terminal cysteine-terminated ethylenediamine precursors, gels are formed under mild conditions, avoiding harsh reagents or catalysts and ensuring compatibility with physiological environments. The resulting hydrogels benefit from enhanced cross-linking dens

Advanced regenerative biomaterials for ischaemic stroke: a comprehensive therapeutic strategy.

Ischaemic stroke is a major global public health problem, accounting for 62.4% of all strokes, with far-reaching consequences in terms of health burden in all regions. Nonetheless, a therapeutic window that is excessively narrow, coupled with restricted therapy modalities, has hindered the majority of patients from receiving effective treatment. With the rapid advancement of biomaterials technology, stroke treatment is being propelled towards a regenerative medicine era characterized by the tripartite collaboration of "materials, drugs, and cells". This review systematically reviews the cuttin

Rapid and Inexpensive Image-Guided Grayscale Biomaterial Customization via LCD Printing.

Hydrogels are an important class of biomaterials that permit cells to be cultured and studied within engineered microenvironments of user-defined physical and chemical properties. Though conventional 3D extrusion and stereolithographic (SLA) printing readily enable homogeneous and multimaterial hydrogels to be formed with specific macroscopic geometries, strategies that further afford spatiotemporal customization of the underlying gel physicochemistry in a non-discrete manner would be profoundly useful toward recapitulating the complexity of native tissue in vitro. Here, we demonstrate that gr

Study on the biosafety and in vivo metabolic patterns of DNA-inspired Janus Base nanotubes.

Janus base nanotubes (JBNTs), also known as Rosette nanotubes, are synthesized from DNA-inspired base molecular monomers and show promise for biomedical applications. However, their clinical translation has been hindered by a lack of comprehensive biocompatibility and biodistribution studies. In this study, we evaluated the in vitro and in vivo biocompatibility of JBNTs and their biodistribution following intravenous and knee intra-articular injections. Within the concentration range of 40 μg/mL, JBNTs supported excellent cell growth with no significant inflammatory response, and even at a con

Comprehensive overview of biomaterials for bone regeneration in cleft lip and palate.

Cleft lip and palate (CL/P) is the most common congenital malformation within the oral and maxillofacial region of human beings, involving abnormal development of lips, palate, and alveolar bone. Bone grafting is a crucial yet challenging part of the treatment for CL/P. Although autologous iliac bone transplantation is the clinical gold standard, additional surgical trauma, donor site complications, bone resorption, and risk of secondary surgical grafting undermine its clinical efficacy. Recently, synthetic bones have attracted increasing attention for bone regeneration, such as calcium phosph

Exploring bioactive bone cement pastes: A promising solution for orthopedic surgeries.

Bone cement is a biomaterial that has been used effectively in orthopedic surgery for almost 50 years. The cement hardens after injection in the defect site without needing any external agent. Bone cement is commonly used in implant fixation, joint replacement, trauma surgery, osteoporosis management, and antibiotic delivery. Since the bone anatomy of patients is different, the final hardening of bone cement in the body corrects the abnormalities and gaps between the implant and the bone. The most universal injectable bone cements are based on ceramic or polymer compounds. The chemical composi

Development of Novel Squid Gladius Biomaterials for Cornea Tissue Engineering.

Cornea tissue engineering is strictly dependent on the development of biomaterials that fulfill the strict biocompatibility, biomechanical, and optical requirements of this organ. In this work, we generated novel biomaterials from the squid gladius (SG), and their application in cornea tissue engineering was evaluated. Results revealed that the native SG (N-SG) was biocompatible in laboratory animals, although a local inflammatory reaction was driven by the material. Cellularized biomaterials (C-SG) demonstrated that the SG provides an adequate substrate for cell attachment and growth, and cor

From Basic Science to Clinical Practice: A Review of Current Periodontal/Mucogingival Regenerative Biomaterials.

Periodontitis is a dysbiosis-driven inflammatory disease affecting the tooth-supporting tissues, characterized by their progressive resorption, which can ultimately lead to tooth loss. A step-wise therapeutic approach is employed for periodontitis. After an initial behavioral and non-surgical phase, intra-bony or furcation defects may be amenable to regenerative procedures. This review discusses the regenerative technologies employed for periodontal regeneration, highlighting the current limitations and future research areas. The search, performed on the MEDLINE database, has identified the av

A new direction in periodontitis treatment: biomaterial-mediated macrophage immunotherapy.

Periodontitis is a chronic inflammation caused by a bacterial infection and is intimately associated with an overactive immune response. Biomaterials are being utilized more frequently in periodontal therapy due to their designability and unique drug delivery system. However, local and systemic immune response reactions driven by the implantation of biomaterials could result in inflammation, tissue damage, and fibrosis, which could end up with the failure of the implantation. Therefore, immunological adjustment of biomaterials through precise design can reduce the host reaction while eliminati

Enantiomer-Dependent Supramolecular Immunosuppressive Modulation for Tissue Reconstruction.

Modulating the properties of biomaterials in terms of the host immune response is critical for tissue repair and regeneration. However, it is unclear how the preference for the cellular microenvironment manipulates the chiral immune responses under physiological or pathological conditions. Here, we reported that in vivo and in vitro oligopeptide immunosuppressive modulation was achieved by manipulation of macrophage polarization using chiral tetrapeptide (Ac-FFFK-OH, marked as FFFK) supramolecular polymers. The results suggested that chiral FFFK nanofibers can serve as a defense mechanism in t

Responsive Supramolecular Polymers for Diagnosis and Treatment.

Stimuli-responsive supramolecular polymers are ordered nanosized materials that are held together by non-covalent interactions (hydrogen-bonding, metal-ligand coordination, π-stacking and, host-guest interactions) and can reversibly undergo self-assembly. Their non-covalent nature endows supramolecular polymers with the ability to respond to external stimuli (temperature, light, ultrasound, electric/magnetic field) or environmental changes (temperature, pH, redox potential, enzyme activity), making them attractive candidates for a variety of biomedical applications. To date, supramolecular res
