Tumour-brain crosstalk restrains cancer immunity via a sensory-sympathetic axis.

Body-brain communication has emerged as a key regulator of tissue homeostasis1-5. Solid tumours are innervated by different branches of the peripheral nervous system and increased tumour innervation is associated with poor cancer outcomes6-8. However, it remains unclear how the brain senses and responds to tumours in peripheral organs, and how tumour-brain communication influences cancer immunity. Here we identify a tumour-brain axis that promotes oncogenesis by establishing an immune-suppressive tumour microenvironment. Combining genetically engineered mouse models with neural tracing, tissue

Inhibitory Circuits Can Restore OFF Pathway Responses in Retinal Prostheses.

One of the first steps in processing visual information is to split the light signal captured by photoreceptors into complementary ON and OFF pathways, which separately encode increases and decreases in luminance. In blind patients with retinal degeneration, optoelectronic prostheses can successfully activate the ON pathway and evoke bright percepts; however, patients do not perceive dark features. This indicates that the OFF pathway is not being activated by existing prosthetics. To quantify OFF pathway deficits, we stimulated retinal ganglion cells (RGCs) in a mouse model of retinitis pigmen

Electroacupuncture-based vagal stimulation attenuates epileptic seizures through a body-brain circuit.

Vagus nerve stimulation offers a promising strategy for seizure control but remains limited by its invasive delivery. Here, we reveal electroacupuncture (EA), an ancient neuromodulatory technique rooted in traditional Chinese medicine, at specific somatic acupoints linked to the vagal-brain axis to treat epilepsy and delineate the precise underlying neural mechanisms. We demonstrate that EA at the Dazhui (GV14) acupoint exhibits broad-spectrum antiseizure efficacy across multiple seizure models. Anatomical and functional analyses reveal that GV14 activates the vagal afferents and recruits neur

CRH neurons in the dorsal bed nucleus of the stria terminalis (dBNST) mediate contextual fear generalization.

Contextual fear generalization is a hallmark of psychiatric disorders including post-traumatic stress disorder (PTSD), generalized anxiety disorder and panic disorder. Among these disorders, PTSD is particularly associated with contextual fear generalization. However, the mechanisms by which fear responses extend to similar contexts remain unclear. Here we tested whether corticotropin-releasing hormone (CRH) neurons in the dorsal bed nucleus of the stria terminalis (dBNST) contribute to contextual fear generalization. Using a contextual fear generalization paradigm with variable shock timing,

Role of CaMKIIa reticular neurons of caudal medulla in control of posture.

Terrestrial quadrupeds stabilize dorsal-side-up body orientation (the vertical orientation of its dorso-ventral axis) through the postural control system, with supraspinal inputs, including those from the reticular formation, playing a central role. The contribution of specific molecularly identified reticular neuron populations to posture, however, has remained unclear. We investigated CaMKIIa-expressing reticular neurons (CaMKIIa-RNs) in the caudal medulla and their role in postural regulation. Using chemogenetic activation and inactivation in mice, we found that unilateral activation of CaM

Potentiation of Nigra-Striatal Dopaminergic Projection Underpins Core Autism-Like Behaviors in Valproate-Exposed Mice.

Autism is characterized by two key diagnostic criteria including social deficits and repetitive behaviors. However, the underlying neural circuit dysfunction that accounts for these coexisting symptoms in autism remains poorly understood. Here we revealed that prenatal valproate exposure induced functional alterations of dopaminergic projections from substantia nigra pars compacta (SNc) to dorsomedial striatum (DMS). Specifically, we observed enhanced excitatory input and increased excitability in SNc→DMS dopamine (DA) neurons, resulting in a basal state of potentiation. This potentiated basel

Enhanced Microglial Engulfment of Dopaminergic Synapses Induces Parkinson's Disease-Related Executive Dysfunction in an Acute LPC Infusion Targeting the mPFC.

The dysfunction of the dopaminergic projection from the ventral tegmental area (VTA) to the medial prefrontal cortex (mPFC) is believed to play a key role in the pathophysiology of Parkinson's disease (PD) accompanied by executive dysfunction (EDF). In this study, we identified an abnormal increase in lysophosphatidylcholine (LPC) levels in PD patients, which closely correlates with the severity of cognitive impairment. LPC disrupts the miR-2885/TDP-43 signaling pathway in microglia, driving dopaminergic presynaptic engulfment. In LPC-exposed mice, microglial activation via miR-2885/TDP-43/p65

Microglia drive synaptic and functional connectivity deficits in the Ts65Dn mouse model of Down syndrome by affecting inhibition.

Microglia, the resident immune cells of the brain, play a crucial role in sculpting neuronal circuits during development, and their dysfunction is increasingly implicated in neurodevelopmental disorders such as Down syndrome (DS). Here, we reveal a previously unrecognized pathological mechanism whereby microglia contribute to synaptic and neuronal activity deficits in DS: a selective disruption of microglia–interneuron interactions. Using primary neuron–microglia co-cultures from Ts65Dn mice, we show that while trisomy in neurons drives excitatory synaptic deficits and major microglial morphol

Inhibition of Ephrin receptor signaling in the paraventricular nucleus attenuates psoriasis-like dermatitis via synaptic plasticity and immune homeostasis modulation.

Psoriasis pathophysiology involves dysregulated neuroimmune crosstalk, yet the central mechanisms involved remain incompletely understood. Here, we show that the hypothalamic paraventricular nucleus (PVN) orchestrates cutaneous inflammation via a transsynaptic brain-skin circuit. Using neural tracing and chemogenetic approaches, we revealed functional connectivity between the PVN and both sympathetic neurons and psoriatic skin. Reactivation of imiquimod (IMQ)-induced PVN-transgenic targeted recombination in active population (TRAP) neurons (which form a specific "inflammatory memory") is essen

Environmental noise-induced changes to the IC-SNc circuit promotes motor deficits and neuronal vulnerability in a mouse model of Parkinson's Disease.

Emerging clinical evidence suggests a link between environmental noise and the severity of Parkinson's disease (PD). However, the effects of high-decibel noise exposure on PD and its underlying mechanisms remain unclear. In this study, we demonstrate that acute noise exposure induces reversible motor deficits in subacute low-dose 6-hydroxydopamine (6-OHDA) mice, a model of presymptomatic early-stage PD, while chronic noise exposure results in irreversible motor deficits and significant loss of substantia nigra compacta (SNc) dopaminergic (DA) neurons. Additionally, noise exposure activates the

Adapter protein 2-modulated μ -opioid receptor trafficking in paraventricular thalamus contributes to fentanyl contextual addiction memory in mice.

Fentanyl and its analogues are the most commonly used synthetic opioid analgesics in clinical practice, but their abuse is a significant concern. Drug-paired environmental cues often trigger memory retrieval, leading to relapse, complicating treatment and overdose prevention. In this study we investigated μ-opioid receptor-related molecular mechanisms underlying the retrieval of fentanyl contextual addiction memory in mice. A conditional place preference (CPP) model was established in mice by citrate injections of fentanyl (0.1 mg/kg) for 4 days. By performing whole-brain screening using c-Fos

The gut vagal sensory pathway drives postprandial sleep via activation of PVH-projecting GABAergic neurons in the NTS.

Feeding status bidirectionally modulates sleep; however, the neural circuitry that integrates the sensing of gastrointestinal (GI) state and sleep remains poorly understood. The afferent fibers of the vagus nerve extensively innervate the GI tract, transmitting postprandial satiety signals to the brain. This study investigates the key role of the upper gut-innervating vagal sensory neurons in modulating sleep-wake states and promoting postprandial sleep, uncovering the underlying circuit mechanisms. Both feeding and activation of stomach/duodenum-innervating vagal sensory neurons reduce wakefu

A coordinated and enhanced brain network supports the persistence of long-term cocaine memory.

The persistence of drug memories accounts for the high risk of drug relapse, which is a major challenge in the treatment of substance use disorders. However, the neurobiological underpinnings, especially the dynamic changes of brain networks underlying long-term drug memories, remain unclear. Here we utilized cocaine conditioned place preference (CPP) in rats combined with c-Fos mapping in multiple brain regions and network analysis to assess dynamic patterns of neural activity and functional memory networks following the recall of short-term and long-term cocaine memory. Furthermore, we emplo

Cortical kappa opioid receptors integrate negative affect and sleep disturbance.

Sleep disruption and negative affect are attendant features of many psychiatric and neurological conditions that are often co-morbid including major depressive disorder, generalized anxiety disorder and chronic pain. Whether there is a causal relationship between negative affect and sleep disruption remains unclear. We therefore asked if mechanisms promoting negative affect can disrupt sleep and whether inhibition of pathological negative affect can normalize disrupted sleep. Signaling at the kappa opioid receptor (KOR) elicits dysphoria in humans and aversive conditioning in animals. We teste

Indole induces anxiety-like behaviour in mice mediated by brainstem locus coeruleus activation.

The gut microbiota produces metabolites that enrich the host metabolome and play a part in host physiology, including brain functions. Yet the biological mediators of this gut-brain signal transduction remain largely unknown. In this study, the possible role of the gut microbiota metabolite indole, originating from tryptophan, was investigated. Oral administration of indole to simulate microbial overproduction of this compound in the gut consistently led to impaired locomotion and anxiety-like behaviour in both C3H/HeN and C57BL/6J mice. By employing c-Fos protein expression mapping in mice, w

PV and SST neurons in the anterior cingulate cortex regulate social disorders in adulthood induced by sensory abnormalities in childhood.

OBJECTIVE: Childhood sensory abnormalities experience has a crucial influence on the structure and function of the adult brain. The underlying mechanism of neurological function induced by childhood sensory abnormalities experience is still unclear. Our study was to investigate whether the GABAergic neurons in the anterior cingulate cortex (ACC) regulate social disorders caused by childhood sensory abnormalities experience. METHODS: We used two mouse models, complete Freund's adjuvant (CFA) injection mice and bilateral whisker trimming (BWT) mice in childhood. We applied immunofluorescence, c

Glutamatergic neurons in ventral pallidum modulate heroin addiction via epithalamic innervation in rats.

Glutamatergic neurons in ventral pallidum (VPGlu) were recently reported to mediate motivational and emotional behavior, but its role in opioid addiction still remains to be elucidated. In this study we investigated the function of VPGlu in the context-dependent heroin taking and seeking behavior in male rats under the ABA renewal paradigm. By use of cell-type-specific fiber photometry, we showed that the calcium activity of VPGlu were inhibited during heroin self-administration and context-induced relapse, but activated after extinction in a new context. The drug seeking behavior was accompan

The plasticity of neuropeptide Y-Y1 receptor system on Tac2 neurons contributes to mechanical hyperknesis during chronic itch.

Rationale: In the physiological states, the act of scratching protects the person from harmful substances, while in certain pathological conditions, the patient suffers from chronic itch, both physically and mentally. Chronic itch sufferers are more sensitive to mechanical stimuli, and mechanical hyperknesis relief is essential for chronic itch treatment. While neuropeptide Y-Y1 receptor (NPY-Y1R) system is known to play a crucial role in modulating mechanical itch in physiological conditions, it is elusive how they are altered during chronic itch. We hypothesize that the negative regulatory e

Cerebellar contributions to a brainwide network for flexible behavior in mice.

The cerebellum regulates nonmotor behavior, but the routes of influence are not well characterized. Here we report a necessary role for the posterior cerebellum in guiding a reversal learning task through a network of diencephalic and neocortical structures, and in flexibility of free behavior. After chemogenetic inhibition of lobule VI vermis or hemispheric crus I Purkinje cells, mice could learn a water Y-maze but were impaired in ability to reverse their initial choice. To map targets of perturbation, we imaged c-Fos activation in cleared whole brains using light-sheet microscopy. Reversal

Hippocampal area CA2 controls seizure dynamics, interictal EEG abnormalities and social comorbidity in mouse models of temporal lobe epilepsy.

UNLABELLED: Temporal lobe epilepsy (TLE) is characterized by spontaneous recurrent seizures, abnormal activity between seizures, and impaired behavior. CA2 pyramidal neurons (PNs) are potentially important because inhibiting them with a chemogenetic approach reduces seizure frequency in a mouse model of TLE. However, whether seizures could be stopped by timing inhibition just as a seizure begins is unclear. Furthermore, whether inhibition would reduce the cortical and motor manifestations of seizures are not clear. Finally, whether interictal EEG abnormalities and TLE comorbidities would be im
