Ref: Submission ID 0e636778-60d8-4db7-8504-1b5c099437d9 Preliminary In Vitro Evaluation of Dodecyl Trimethylammonium Bromide (DDTAB) as a Selective Inhibitor of Theileria annulata-Infected Lymphocyte Proliferation.

PURPOSE: Tropical theileriosis, caused by the protozoan parasite Theileria annulata, is an economically significant constraint to cattle production, with disease control increasingly compromised by emerging resistance to the primary chemotherapeutic agent, buparvaquone. This study aimed to screen candidate compounds for in vitro anti-theilerial activity and host cell safety. METHODS: Four compounds - hesperidin methyl chalcone (HMC), p-coumaric acid, dodecyl trimethylammonium bromide (DDTAB), and diclofop-methyl - were evaluated for anti-theilerial activity using a resazurin-based viability a

Nutritional and technological potential of umbu-caja and soursop co-product flours.

Umbu-caja and soursop from the Northeast region of Brazil are rich in nutrients and bioactive compounds and are widely processed by the fruit agroindustry. However, there is a lack of research examining the composition and nutritional/technological potential of these co-product fruits. The present study evaluated the nutritional and technological characteristics of umbu-caja and soursop co-product flours (UCF and SCF, respectively), in addition to cytotoxicity in healthy cells. The results demonstrated that they are rich in dietary fiber (approximately 53 %), low in protein (approximately 8.0

Biochemical and Structural Analysis of Substrate Specificity of a Phenylalanine Ammonia-Lyase.

Phenylalanine ammonia-lyase (PAL) is the first enzyme of the general phenylpropanoid pathway catalyzing the nonoxidative elimination of ammonia from l-phenylalanine to give trans-cinnamate. In monocots, PAL also displays tyrosine ammonia lyase (TAL) activity, leading to the formation of p-coumaric acid. The catalytic mechanism and substrate specificity of a major PAL from sorghum (Sorghum bicolor; SbPAL1), a strategic plant for bioenergy production, were deduced from crystal structures, molecular docking, site-directed mutagenesis, and kinetic and thermodynamic analyses. This first crystal str

An In Vivo C57BL/6 Mouse Study Demonstrating Fine-Tuning of Serum and Brain-Derived Exosomes Reveal Therapeutic Potential of Phytochemical Ferulic Acid Against Alzheimer's Disease.

Alzheimer's disease (AD) is a neurodegenerative disorder that is characterized by memory loss, synaptic impairment, and biochemical changes. The study aimed to investigate the neuroprotective role of ferulic acid (FA) in an AD mouse C57BL/6 model induced by the Aβ(25-35) peptide. Cognitive deficits, which include memory and spatial learning deficits caused by Aβ, were evaluated using a set of behavioural tests. Transmission electron microscopy (TEM), dynamic light scattering (DLS), and Western blot analysis were used to characterize the exosomes from the brain and the serum. The activity of ac

Cytoplasmic sequestering of a fungal stress-activated MAPK in response to a host plant phenolic acid.

Fungal pathogens employ conserved signaling pathways to survive in the host. The stress-activated MAP kinase Hog1 of the maize pathogen Cochliobolus heterostrophus undergoes dephosphorylation upon exposure to ferulic acid, a phenolic compound abundant in the host plant. Unlike its nuclear localization during osmotic stress, Hog1 forms cytoplasmic foci in response to FA, indicating its sequestering to a compartment or condensate. FA prevents several characteristic responses of the Hog1 pathway to osmotic stress: hyperphosphorylation of Hog1, nuclear localization, and expression of a monosacchar

Inhibitory effects of ferulic acid on the contraction responses of porcine coronary arteries: a comparison with diltiazem.

Ferulic acid (FA) exerts protective effects against cardiovascular-related diseases; however, its role in coronary artery dilation is unclear. Here, we examined the effects and underlying mechanisms of FA in response to contractions induced by spasmogenic candidates in porcine coronary arteries (PCAs). FA (3 × 10-4-3 × 10-3 M) inhibited high-KCl-induced contractions in a concentration-dependent manner, and FA (3 × 10-4-10-3 M) inhibited high-KCl-induced increases in intracellular Ca2+ concentrations in A7r5 cells. FA (3 × 10-3 M) showed greater inhibition than diltiazem (3 × 10-5 M) against ch

Ferulic acid suppresses guinea pig ileal longitudinal smooth muscle contractions by inhibiting voltage-dependent Ca2+ channels.

We investigated the effects of ferulic acid (FA) on contractions in guinea pig ileal longitudinal smooth muscle (ILSM). FA (3 × 10-4-3 × 10-3 M) inhibited ILSM contractions induced by acetylcholine, histamine, prostaglandin F2α, and serotonin concentration-dependently, reversibly, and noncompetitively with pD'2 values of ∼3. FA also concentration-dependently and reversibly inhibited KCl-induced ILSM contractions. FA (10-3 M), which inhibited ILSM contractions by ∼50 %, also reduced verapamil-sensitive, KCl-induced intracellular Ca2+ increases in A7r5 cells by ∼35 %. These results suggest that

Biocompatible sinapic acid-loaded chitosan nanoparticles as a topical nanotherapy for uveitis in rats: Formulation, characterization, and anti-inflammatory efficacy.

Sinapic acid (SA), a naturally occurring polyphenol, exhibits diverse biological properties, including anti-inflammatory activity. Its potential in treating ocular inflammations remains underexplored. Ocular protective barriers and hydrophobicity of SA hinder its effective ocular delivery. To overcome these limitations, SA-loaded chitosan nanoparticles (SA-CSNPs) were developed. Characterization revealed nano-sized (234.7 nm), positively charged (+ 32.4 mV) SA-CSNPs with a narrow polydispersity index (0.304). XRD and FTIR analyses confirmed the amorphous state of encapsulated SA without advers

In vivo reduction of skin inflammation using ferulic acid-loaded lipid vesicles derived from Brewer's spent grain.

Breweŕs spent grain (BSG) is the main by-product of the brewing industry, and due to its rapid decomposition, it generates serious environmental problems such as malodors and greenhouse gases emissions. On the other hand, this lignocellulosic compound contains a large number of antioxidants, being ferulic acid (FA) the most abundant. FA is a powerful antioxidant molecule that has demonstrated significant protective effects on key components of the skin, including keratinocytes, fibroblasts, collagen, and elastin. FA inhibits melanogenesis, promotes angiogenesis and accelerates the wound healin

Structural elucidation, distribution and antioxidant activity of bound phenolics from whole grain brown rice.

Chemical profiles, distribution, and antioxidant activity of bound phenolics from brown rice were investigated. Four new dehydrodiferulic acid dimers (DFA) along with eighteen known phenolics were isolated from brown rice bound phenolic extracts and their structures were determined by multiple spectroscopic methods. Among them, ferulic acid and 8-5' DFA were the most abundant monomeric and dimeric bound phenolics in brown rice, rice bran and polished rice. In whole brown rice, polished rice contributed more than 50% of three phenolic monomers and six phenolic dimers, while rice bran contribute

Xylanase from Aspergillus tamarii shows different kinetic parameters and substrate specificity in the presence of ferulic acid.

A 22 kDa xylanase (AtXyl1) from Aspergillus tamarii was purified by two chromatographic steps and presented preference for oat spelt (OSX), birchwood (BrX) and beechwood (BeX) xylans respectively, as substrates. AtXyl1 displays the highest activity at pH 5.5 and 55 °C and showed tolerance over a range of different phenolic compounds. The activity of AtXyl1 was not inhibited when the enzyme was incubated with ferulic acid (FA) using OSX or BrX as substrate. On the other hand, the incubation of AtXyl1 with BeX and FA resulted in an increase in enzyme activity. The molecular docking of a GH11 xyl

Ferulic acid lowers body weight and visceral fat accumulation via modulation of enzymatic, hormonal and inflammatory changes in a mouse model of high-fat diet-induced obesity.

Previous studies have reported on the glucose and lipid-lowering effects of ferulic acid (FA) but its anti-obesity potential has not yet been firmly established. This study investigated the possible anti-obesitogenic effects of FA in mice fed a high-fat diet (HFD) for 15 weeks. To assess the antiobesity potential of FA, 32 male Swiss mice, weighing 20-25 g (n=6-8 per group) were fed a normal diet (ND) or HFD, treated orally or not with either FA (10 mg/kg) or sibutramine (10 mg/kg) for 15 weeks and at the end of this period, the body weights of animals, visceral fat accumulation, plasma levels

Dissociation kinetics of singly protonated leucine enkephalin investigated by time-resolved photodissociation tandem mass spectrometry.

The yields of post-source decay (PSD) and time-resolved photodissociation (PD) at 193 and 266 nm were measured for singly protonated leucine enkephalin ([YGGFL + H](+)), a benchmark in the study of peptide ion dissociation, by using tandem time-of-flight mass spectrometry. The peptide ion was generated by matrix-assisted laser desorption ionization (MALDI) using 2,5-dihydroxybenzoic acid as the matrix. The critical energy (E(0)) and entropy (DeltaS(++) at 1000 K) for the dissociation were determined by Rice-Ramsperger-Kassel-Marcus fit of the experimental data. MALDI was done for a mixture of

Antioxidant activity of ferulic acid alkyl esters in a heterophasic system: a mechanistic insight.

The antioxidant activity of some esters of ferulic acid with the linear fatty alcohols C7, C8 (branched and linear), C9, C11, C12, C13, C15, C16, and C18 has been studied in homogeneous and heterogeneous phases. Whereas in homogeneous phase all of the alkyl ferulates possessed similar radical-scavenging abilities, in rat liver microsomes they showed striking differences, the more effective being C12 (7) (IC50 = 11.03 M), linear C8 (3) (IC50 = 12.40 microM), C13 (8) (IC50 = 18.60 microM), and C9 (5) (IC50 = 19.74 microM), followed by C7 (2), C15 (9), C11 (6), branched C8 (4), C16 (10), and C18

Biosensor-guided evolution of chalcone synthase enhances biosynthesis of natural and non-natural flavanones.

Flavonoids constitute a large class of natural products widely investigated for their bioactive properties, with microbial production offering a potentially scalable alternative to plant extraction. However, achieving structural diversification of these compounds in microbial systems remains challenging, as modification of the flavonoid B-ring typically relies on downstream tailoring enzymes. An alternative strategy is to exploit the intrinsic promiscuity of the canonical flavanone biosynthesis pathway to introduce structural variation at an early stage. Here, we sought to improve microbial pr
