Modes of death and types of cardiac diseases in opiate addicts: analysis of 168 necropsy cases.

One hundred sixty-eight opiate addicts, whose hearts were submitted for necropsy study, were examined with prime focus on modes of death and types of cardiac abnormalities. Twenty various modes of death were identified: active infective endocarditis or its consequences in 67 (40%), drug overdose in 39 (24%), coronary artery disease in 14 (8%), pulmonary granulomatosis in 7 (4%) and 15 various diseases (7 cardiac and 8 noncardiac) in the remaining 41 (24%) patients. Of the 168 hearts examined, only 7 (4%) were normal. Although infective endocarditis (active, healed or both) was most common (80

Tumor-produced secreted form of binding of immunoglobulin protein elicits antigen-specific tumor immunity.

Binding of immunoglobulin protein (BiP) is a major molecular chaperone localized in endoplasmic reticulum (ER). It has been demonstrated to interact with nascent Ig. However, contrary to other ER-resident heat shock proteins such as gp96, calreticulin, and ORP150, it is not clear whether tumor-derived BiP plays a role in inducing antitumor immunity. In this study, we show that the tumor-derived secreted form of BiP is capable of inducing antitumor CD8(+) T cell responses. We constructed an ER-retention signal KDEL-deleted mutant of BiP cDNA and transfected it to tumor cells, which resulted in

Mycobacterium tuberculosis and TLR2 agonists inhibit induction of type I IFN and class I MHC antigen cross processing by TLR9.

Dendritic cells (DCs) cross process exogenous Ags and present them by class I MHC (MHC-I) molecules to CD8(+) T cells specific for Ags from viruses and bacteria such as Mycobacterium tuberculosis. Unmethylated CpG DNA signals through TLR9 to induce type I IFN (IFN-alpha/beta), which enhances MHC-I Ag cross processing, but lipoproteins that signal through TLR2 do not induce IFN-alpha/beta. In these studies we observed that M. tuberculosis, which expresses agonists of both TLR9 and TLR2, did not induce production of IFN-alpha/beta or cross processing by murine DCs. Furthermore, M. tuberculosis a

Investigation of immunogenic effect of the BCG priming and Ag85A- GM-CSF boosting in Balb/c mice model.

Mycobacterium tuberculosis (M. tuberculosis) has once again become a major public health threat owing to the combined effects of a worldwide anti-tuberculosis drug resistance and the emergence of the human immunodeficiency virus pandemic. The Bacille Calmette-Guérin (BCG)-based vaccine has displayed inconsistent efficacy in different trials although it continues to be used to prevent tuberculosis in many countries. In current work, we have developed DNA vaccines expressing M. tuberculosis antigen 85A (Ag85A) and cytokine granulocyte macrophage colony stimulating factor (GM-CSF) and aimed to in

GIFT4 fusokine converts leukemic B cells into immune helper cells.

BACKGROUND: Chronic lymphocytic leukemia (CLL) remains incurable with standard therapy, and is characterized by excessive expansion of monoclonal abnormal mature B cells and more regulatory immune properties of T cell compartment. Thus, developing novel strategies to enhance immune function merits further investigation as a possible therapy for CLL. METHODS: We generated a fusion cytokine (fusokine) arising from the combination of human GM-CSF and IL-4 (named GIFT4). Primary CLL cells were treated with GIFT4 or GM-CSG and IL-4 in vitro. GIFT4-triggered STAT5 signaling in CLL cells was examine

Murine liver-resident group 1 innate lymphoid cells regulate optimal priming of anti-viral CD8+ T cells.

The liver contains 2 transcriptionally distinct group 1 ILC subsets: CD49a+ ILC1s and CD49b+ NK cells. However, little is known about how group 1 ILCs contribute to hepatic immune responses. Therefore, we characterized murine liver-resident group 1 ILCs and found that CD49a+ ILC1s express high levels of the inhibitory receptor NKG2A and localize near DCs in perivascular spaces surrounding the portal triads. Upon hepatic viral infection, NKG2A signaling in group 1 ILCs, especially in CD49a+ ILC1s, inhibits CXCL9 expression required for robust accumulation of IFN-γ+CD49b+ NK cells. As a conseque

T cell-associated immunity induced by heterologous recombinant BCG and purified protein vaccination confers cross-variant protection against SARS-CoV-2.

Previously, we have developed a recombinant BCG-based vaccine (rBCG) expressing a chimeric protein with spike and nucleocapsid epitopes (rChimera) to combine BCG's innate immune training capacity with SARS-CoV-2-specific adaptive immune responses. The heterologous prime-boost rBCG/rChimera+Alum regimen induced strong humoral and cellular immunity, conferring protection against the ancestral Wuhan strain in K18-hACE2 mice. Here, we used knockout mice lacking functional B cells, CD4+, or CD8+ T lymphocytes expressing hACE2 to uncover the mechanisms of anti-viral immune protection, while also eva

Validation-Aware Retrospective EEG Treatment-Response Modelling Using Chaotic Pattern of Prime Numbers Features: Segment-Level Separability and Subject-Wise Generalisation.

This study presents a validation-aware EEG framework based on Chaotic Pattern of Prime Numbers (CPPN) features for depression treatment-response modelling across one SSRI cohort and two rTMS cohorts. CPPN features were evaluated through a seven-protocol validation hierarchy spanning random segment splitting, segment-level cross-validation, nested segment-level cross-validation, leave-N-subjects-out, fixed-feature leave-one-subject-out (LOSO), nested leave-N-subjects-out, and nested LOSO, with normalisation, NCA ranking, feature-count selection where applicable, and model fitting confined to th

HIV-2 derived lentiviral vectors: gene transfer in Parkinson's and Fabry disease models in vitro.

Lentiviral vectors are prime candidate vectors for gene transfer into dividing and non-dividing cells, including neuronal cells and stem cells. For safety, HIV-2 lentiviral vectors may be better suited for gene transfer in humans than HIV-1 lentiviral vectors. HIV-2 vectors cross-packaged in HIV-1 cores may be even safer. Demonstration of the efficacy of these vectors in disease models will validate their usefulness. Parkinson's disease and Fabry disease provide excellent models for validation. Parkinson's disease is a focal degeneration of dopaminergic neurons in the brain with progressive lo

The "Survivor Peptide" Hypothesis: Structural Resilience and Immunological Persistence of Food Allergens in the Gut-Mammary Axis.

Background: The translocation of diet-derived antigens from the maternal intestine to breast milk represents a primary gateway for neonatal immune priming, yet the structural basis for why certain proteins survive this transit while others do not remains poorly understood. This review introduces the "Survivor Peptide" hypothesis, proposing that specific food allergens possess intrinsic "stability architectures" that enable them to resist maternal digestion and navigate the gut-mammary axis to reach the infant in an immunologically active form. Methods: We analyzed the current literature regard

Beyond the tumor microenvironment: a hierarchical immune-circuit model of immune checkpoint blockade resistance.

Immune checkpoint blockade (ICB) produces durable tumor control in a subset of patients, yet resistance is usually interpreted through tumor-intrinsic lesions or suppression within the tumor microenvironment (TME). We propose that ICB resistance can also be organized as failure of a multiscale antitumor immune circuit comprising local immune execution, regional immune education in tumor-draining lymph nodes (TDLNs), and systemic immune calibration by the host. In this model, "hierarchical" denotes nested functional dependence rather than one-way anatomical control: local killing depends probab

The nociceptor-CGRP-lactate axis in cancer immune escape: Linking neural signaling, immunometabolism, and immunotherapy resistance.

Cancer neuroscience has expanded the conceptual landscape of tumor biology by revealing that nerves are not passive bystanders within malignant tissues, but active regulators of tumor progression, immune surveillance, and therapeutic response. Among neural components of the tumor microenvironment, nociceptive sensory neurons have emerged as critical interfaces between tissue injury, inflammation, cancer-associated pain, and immune regulation. This review proposes that nociceptive neuron-driven immune escape represents a tissue-level pathological process positioned at the intersection of cancer

Research of childhood tuberculosis in suspected populations by molecular methods: A multicenter study in China.

The research of childhood tuberculosis is inadequate in china. The cross-priming amplification (CPA) of specific DNA in clinical samples is increasingly adopted for the diagnosis of childhood tuberculosis. In this study, a multicenter research was performed to investigate the incidence and characteristics of childhood tuberculosis in suspected populations mainly by CPA method. 851 children suspected of tuberculosis were enrolled in seven centers across China. All samples were tested by a CPA method and 159 subjects were tested by Xpert MTB/RIF and liquid culture method in parallel to assess th…

Lexical Access in Preschool Mandarin-Speaking Children With Cochlear Implants.

PURPOSE: Children with cochlear implants (CIs) have less experience accessing spoken language. Mandarin Chinese uses pitch information to contrast word meaning, and the signal that the CI devices provide is degraded. Thus, Mandarin-speaking children with CIs may face more challenges in the development of language skills. This study examines preschool Mandarin-speaking children's performance in lexical access. We hypothesized that children with CIs and their peers with normal hearing (NH) have comparable naming ability, but they process phonological or semantic information differently. METHOD:

Establishment of Systems to Enable Isolation of Porcine Monoclonal Antibodies Broadly Neutralizing the Porcine Reproductive and Respiratory Syndrome Virus.

The rapid evolution of porcine reproductive and respiratory syndrome viruses (PRRSV) poses a major challenge to effective disease control since available vaccines show variable efficacy against divergent strains. Knowledge of the antigenic targets of virus-neutralizing antibodies that confer protection against heterologous PRRSV strains would be a catalyst for the development of next-generation vaccines. Key to discovering these epitopes is the isolation of neutralizing monoclonal antibodies (mAbs) from immune pigs. To address this need, we sought to establish systems to enable the isolation o

Age-related changes in the cognitive function of sleep.

Healthy aging is characterized by a diminished quality of sleep with decreased sleep duration and increased time awake after sleep onset. Older adults awaken more frequently and tend to awaken less from rapid eye movement (REM) sleep and more from non-REM (nREM) sleep than young adults. Sleep architecture also begins changing in middle age leading to a dramatic decrease in the deepest stage of nREM-slow wave sleep (SWS)-as aging progresses. Other less marked nREM changes include reduced numbers of sleep spindles and K-complexes. In contrast, the amount of REM diminishes only slightly. Both cir

Inhibition of ADP ribosylation prevents and cures helicobacter-induced gastric preneoplasia.

Gastric adenocarcinoma develops as a consequence of chronic inflammation of the stomach lining that is caused by persistent infection with the bacterium Helicobacter pylori. Gastric carcinogenesis progresses through a sequence of preneoplastic lesions that manifest histologically as atrophic gastritis, intestinal metaplasia, and dysplasia. We show here in several preclinical models of Helicobacter-induced atrophic gastritis, epithelial hyperplasia, and metaplasia that the inhibition of ADP ribosylation by the small-molecule inhibitor PJ34 not only prevents the formation of gastric cancer precu

Development of peptide mimics of a protective epitope of Vibrio cholerae Ogawa O-antigen and investigation of the structural basis of peptide mimicry.

As an alternative approach toward the development of a cholera vaccine, the potential of peptide mimics of Vibrio cholerae lipopolysaccharide (LPS) to elicit cross-reactive immune responses against LPS was investigated. Two closely related protective monoclonal antibodies, S-20-4 and A-20-6, which are specific for Ogawa O-antigen (O-specific polysaccharide; O-SP) of V. cholerae O1, were used as the target antibodies (Abs) to pan phage display libraries under different elution conditions. Six phage clones identified from S-20-4 panning showed significant binding to both S-20-4 and A-20-6. Thus,
