Putting the pieces together: A case report on human remains identification driven by tattoo recognition.

Dismemberment is a relatively uncommon phenomenon in forensic pathology; its aim is to hinder identification and facilitate corpse concealment. We present a case of fragmented remains where facial recognition was impossible and fingerprints yielded no matches in law enforcement databases. Identification initially relied on the meticulous documentation of 11 tattoos, recorded after thorough cleansing and anatomical reconstruction of the cadaver. Notably, the perpetrator had attempted to excise or char specific skin areas to hide these marks, further complicating the process. Due to the lack of

Application of deep learning for detecting implants in computed tomography scout images with multi-institution and multi-vendor for personal identification.

The identification of deceased individuals is essential in forensic investigations, particularly when primary identification methods such as odontology, fingerprint, or DNA analysis are unavailable. In such cases, implanted medical devices may serve as supplementary identifiers for positive identification. This study proposes deep learning-based methods for the automatic detection of metallic implants in scout images acquired from computed tomography (CT). Using a multi-institutional and multi-vendor dataset, two object detection models, RetinaNet and Faster R-CNN, were trained and evaluated t

Antigenic and genetic relatedness of Leptospira strains isolated from the Andaman Islands in 1929 and 2001.

Leptospirosis is a major public health problem in Andaman Islands. Several strains of Leptospira have been isolated from the Andamans over the years. Leptospires isolated recently from human cases were compared with one of the earliest available isolates from these islands, dating back to 1929, to study their serological and genetic relatedness. Randomly amplified polymorphic DNA (RAPD) fingerprints of the isolates, generated with a primer used previously to differentiate between Leptospira species and serovars, revealed that some of the recent isolates were genetically identical to the 1929 i

HIV-related tuberculosis in a transgender network--Baltimore, Maryland, and New York City area, 1998-2000.

During June-August 1998, the Tuberculosis (TB) Control Program of the Baltimore City Health Department (BCHD) identified four cases of TB among young black men. Three of these men also had human immunodeficiency virus (HIV) infection. The four reported belonging to a social network of transgender persons (i.e., persons who identify with or express a gender and/or sex different from their biologic sex) (1). By October 1998, test results on Mycobacterium tuberculosis isolates from the four men demonstrated a matching 11-band DNA fingerprint pattern (2), suggesting that these case-patients were e

Use of PCR to study epidemiology of Serratia marcescens isolates in nosocomial infection.

A method to characterize strains of Serratia marcescens based on the PCR amplification of enterobacterial repetitive intergenic consensus sequences has been developed. The PCR fingerprints were generated from boiled supernatants prepared directly from bacterial colonies without the need for DNA extraction. The technique was applied to isolates obtained during an outbreak of pneumonia from seven mechanically ventilated patients, and its result indicated that the outbreak was due to the spread of two epidemic strains. This technique was validated by comparison with rRNA gene restriction analysis

Performance of the ForenSeqTM Imagen Kit for Forensic DNA Phenotyping Under Partial Genotyping Conditions.

BACKGROUND: Forensic DNA phenotyping (FDP) enables the inference of externally visible characteristics (EVCs) and biogeographic ancestry when conventional STR profiling is inconclusive. The ForenSeq™ Imagen kit (107 SNPs) integrates phenotype-, ancestry-, and Y-SNPs markers; however, its performance under partial genotyping conditions has not been systematically evaluated. METHODS: Ninety-four samples from a Mexican mestizo population were analyzed using the ForenSeq™ Imagen kit on the MiSeq FGx™ platform. Due to incomplete genotype recovery, 41 samples with >60% locus detection were selected

Forensic characterization of the 94 identity-informative SNPs included in the ForenSeq™ DNA Signature Prep Kit in a Mexican population sample.

BACKGROUND: Single Nucleotide Polymorphisms (SNPs) have emerged as valuable markers in human identification when short tandem repeat analysis fails to solve casework. METHODS: The panel of 94 identity-informative SNPs included in the ForenSeq™ DNA Signature Prep Kit was evaluated in Mexican Mestizo populations using massively parallel sequencing. A total of 104 samples from three geographic regions were analyzed and combined with 357 previously published profiles from Nuevo León, Jalisco, and Mexico City to generate a national dataset (n = 461). Forensic parameters, genetic differentiation, a

Fingerprinting of poultry isolates of Enterococcus cecorum using three molecular typing methods.

Enterococcus cecorum is an emerging challenge to the broiler industry. The organism has been implicated in septicemia, spondylitis, arthritis, and osteomyelitis in commercial broilers and broiler breeders, which lead to economic losses attributed to increased mortality and culling rates, decreased average processing weights, and increased feed conversion ratios. The current study evaluated the genetic variability of 30 clinical isolates of E. cecorum from outbreaks in Pennsylvania, using 3 molecular typing methods, namely, pulsed-field gel electrophoresis (PFGE), randomly amplified polymorphic

Validation of a multiplex PCR assay for the forensic identification of Indian crocodiles.

A dependable and efficient wildlife species identification system is essential for swift dispensation of the justice linking wildlife crimes. Development of molecular techniques is befitting the need of the time. The forensic laboratories often receive highly ill-treated samples for identification purposes, and thus, validation of any novel methodology is necessary for forensic usage. We validate a novel multiplex polymerase chain reaction assay, developed at this laboratory for the forensic identification of three Indian crocodiles, Crocodylus palustris, Crocodylus porosus, and Gavialis gange

Sequence polymorphism and PCR-restriction fragment length polymorphism analysis of the flagellin gene of Burkholderia pseudomallei.

Four flagellin allelic types (I to IV) of Burkholderia pseudomallei were identified based on their sequence variation and restriction fragment length polymorphism (RFLP) analysis of the amplified flagellin gene. Flagellin allelic type I was the most predominantly (75.0%) found among the 100 clinical isolates of B. pseudomallei investigated in this study.

Two cases of keratitis and corneal ulcers caused by Burkholderia gladioli.

We report two cases of protracted keratitis complicated by corneal ulcer. Burkholderia gladioli, primarily known as a plant pathogen, was cultured from corneal swabs. The epithelial defects healed after extended adequate antibiotic therapy. Despite identical patterns of both strains in restriction fragment length polymorphism analysis, a common infection source was not detectable.

Genetic variability of avian Escherichia coli strains evaluated by enterobacterial repetitive intergenic consensus and repetitive extragenic palindromic polymerase chain reaction.

In this study, we tested the capability of enterobacterial repetitive intergenic consensus (ERIC) and repetitive extragenic palindromic (REP) polymerase chain reaction (PCR) to detect genetic diversity among Escherichia coli strains isolated from chickens bearing clinical signs of colibacillosis and compared the genotypes so obtained with the O:H serotypes and virulence of those strains. The DNAs from 50 avian E. coli strains and from E. coli ATCC 25922 were used to amplify ERIC and REP sequences. DNA from avian strains produced from 8 to 17 bands by ERIC-PCR and from 6 to 20 bands by REP-PCR;

Minisatellite instability in severe combined immunodeficiency mouse cells.

We have recently found that okadaic acid, which shows strong inhibitory activity on protein serine/threonine phosphatases and tumor-promoting activity in vivo and in vitro, induces minisatellite mutation (MSM). Human tumors and chemically induced counterparts in experimental animals are also sometimes associated with MSM. In the present study, we demonstrated minisatellite (MS) instability in severe combined immunodeficiency (SCID) cells in which the DNA-dependent protein kinase catalytic subunit (DNA-PKcs) is impaired. Cells from a SCID fibroblast cell line transformed by simian virus 40 larg

Fatal pneumonia following inoculation of healthy bighorn sheep with Pasteurella haemolytica from healthy domestic sheep.

In a series of three experiments, isolates of Pasteurella haemolytica biotype A, serotype 2, ribotype reference WSU-1, from healthy domestic sheep, were inoculated intratracheally into eight bighorn sheep (Ovis canadensis canadensis) and seven domestic sheep with doses of bacteria ranging from 5.3 x 10(8) to 8.6 x 10(11) colony forming units. Seven of eight inoculated bighorn sheep died from acute pneumonia within 48 hr of inoculation, whereas all seven domestic sheep inoculated with comparable or greater doses of bacteria remained healthy. One contact control bighorn sheep also died 6 days af

A guinea-pig hereditary cataract contains a splice-site deletion in a crystallin gene.

A congenital cataract present in guinea pigs provided a unique opportunity to study a hereditary lens disease at the molecular level. zeta-Crystallin, one of the most abundant guinea pig lens proteins, was found to be altered in the lens of cataractous animals. Several zeta-crystallin cDNA clones were isolated from a cataractous lens library and found to contain a 102-bp deletion towards the 3' end of the coding region. This deletion does not interfere with the reading frame but results in a protein 34 amino acids shorter. Sequence analysis of a normal genomic zeta-crystallin clone revealed th

Multiple gunshot suicide: A case study of three self-Inflicted shots using two revolvers.

Firearm suicides typically involve a single weapon and shot. Multiple gunshot suicides are rare, representing only 0-6% of all firearm suicides, with cases involving two weapons being exceptionally uncommon. We report an extraordinary suicide case involving an 86-year-old male with recently diagnosed bone cancer who used two revolvers to inflict three self-inflicted gunshots. The deceased was found in his bathroom, positioned on a mat with a pillow, holding a Taurus 85 0.38 Special revolver in his left hand and having used a Colt Python 0.357 Magnum revolver with his right hand. Two shots were

Emergence and multi-lineages of carbapenemase-producing Acinetobacter baumannii-calcoaceticus complex from canine and feline origins.

The carbapenemase-producing Acinetobacter baumannii is an important opportunistic bacterium and frequently causes hospital-acquired infections in humans. It also has increasingly been reported in veterinary medicine. This study illustrates multiple clones of carbapenemase-producing A. baumannii disseminating and causing diseases in dogs and cats in Thailand. Between 2016 and 2020, 44 A. baumannii and two A. pittii isolates exhibiting imipenem resistance (MIC≥16 μg/mL) from diagnostic samples were characterized by Pasteur multilocus sequence typing (MLST), sequence grouping (SG), repetitive ext

[Identification by enzyme immunoassay of escape mutants S143L and G145R of hepatitis B virus (Hepadnaviridae: Orthohepadnavirus: Hepatitis B virus)].

INTRODUCTION: The achievement of the goal of the World Health Organization to eliminate viral hepatitis B by 2030 seems to be problematic partly due to the presence of escape mutants of its etiological agent, hepatitis B virus (HBV) (<i>Hepadnaviridae: Orthohepadnavirus: Hepatitis B virus</i>), that are spreading mainly in the risk groups. Specific routine diagnostic assays aimed at identification of HBV escape mutants do not exist. The study aimed the evaluation of the serological fingerprinting method adapted for routine detection of escape mutations in 143 and 145 aa positions of
