Neurotransmitters of the olivocochlear lateral efferent system: with an emphasis on dopamine.

The olivocochlear lateral efferent system (OLES) of the adult mammalian cochlea uses variety of neuroactive substances, such as acetyl choline, GABA, dopamine (DA), enkephalins, dynorphins and CGRP. These neuroactive substances have been located within the efferent, small and dense matrix, fibers and terminals of the inner spiral and tunnel bundles. However, some of these neuroactive substances have also been found outside the OLES. Acetyl choline and CGRP, for instance, appear within the olivocochlear medial efferent fibers, and DA and CGRP may also be present in the perivascular innervation.…

Possible role of dopamine in ventilatory acclimatization to high altitude.

Ventilatory acclimatization to high altitude is accompanied by increased hypoxic (HVR) and hypercapnic (HCVR) ventilatory responses which may reflect increased carotid body chemosensitivity. Dopamine is an inhibitory neuromodulator of the carotid body and its activity may be reduced by hypoxic exposure. To determine whether decreased dopaminergic activity could account for the increased chemosensitivity of acclimatization, we examined the response to peripheral dopamine receptor (D2) blockade with domperidone on HVR and HCVR in awake cats before and after exposure to simulated altitude of 14,0…

Association of attention-deficit disorder and the dopamine transporter gene.

Attention-deficit hyperactivity disorder (ADHD) has been shown to be familial and heritable, in previous studies. As with most psychiatric disorders, examination of pedigrees has not revealed a consistent Mendelian mode of transmission. The response of ADHD patients to medications that inhibit the dopamine transporter, including methylphenidate, amphetamine, pemoline, and bupropion, led us to consider the dopamine transporter as a primary candidate gene for ADHD. To avoid effects of population stratification and to avoid the problem of classification of relatives with other psychiatric disorde…

Reinforcing properties of fencamfamine: involvement of dopamine and opioid receptors.

Fencamfamine (FCF) is a psychostimulant classified as an indirect dopamine agonist. The conditioning place preference (CPP) paradigm was used to investigate the reinforcing properties of FCF. After initial preferences had been determined, animals were conditioned with FCF (1.75, 3.5, or 7.0 mg/kg; IP). Only at the dose of 3.5 mg/kg FCF produced a significant place preference. Pretreatment with SCH23390 (0.05 mg/kg; SC) or naloxone (1.0 mg/kg; SC) 10 min before FCF (3.5 mg/kg, IP) blocked both FCF-induced hyperactivity and CPP. Pretreatment with metoclopramide (10.0 mg/kg; IP) or pimozide (1.0 …

Effect of dopamine on carotid chemoreceptor activity in newborn kittens.

In newborn kittens, intravenous bolus injections of dopamine may be inhibitory or excitatory to the carotid chemosensory activity. In order to further identify the role of dopaminergic mechanisms on the chemosensory activity in the kitten, 10 anesthetized, paralyzed and artificially ventilated kittens aged from 1 to 21 days were studied. The activity of a few chemoafferent fibers from one carotid sinus nerve was recorded at three levels of FiO2 in N2, 21, 8 and 100%, before and during a continuous dopamine infusion at a dose of 10 micrograms/kg/min. The immediate effect of dopamine usually was…

Interaction of AZA analogs of chlorpromazine with the dopamine D2 receptor.

1. Permanently charged AZA analogs of chlorpromazine inhibited the binding of [3H]spiperone and antagonized the apomorphine-induced inhibiti … nctional group near the quaternary nitrogen which is capable of forming a hydrogen bond with the D2 dopamine receptor. 4. However, it appears that for optimal binding, as achieved with chlorpromazine, the hydrogen-bonding proton should be on the charged nitrogen.

Cloning and expression of a rat D2 dopamine receptor cDNA.

Dopamine receptors are classified into D1 and D2 subtypes on the basis of their pharmacological and biochemical characteristics. The D2 dopamine receptor has been implicated in the pathophysiology and treatment of movement disorders, schizophrenia and drug addiction. The D2 dopamine receptor interacts with guanine nucleotide-binding proteins to induce second messenger systems. Other members of the family of receptors that are coupled to G proteins share a significant similarity in primary amino-acid sequence and exhibit an archetypical topology predicted to consist of seven putative transmembr…

No evidence of vascular dopamine receptors in the rat portal vein.

Dopamine (5.3 X 10(-6) to 5.3 X 10(-5) M) decreased the force of spontaneous myogenic contractions of the rat portal vein in a concentration-dependent manner in the presence of the alpha-adrenoreceptor antagonist BE-2254. The DA1-selective agonist fenoldopam and the DA2-preferential agonist (-)-EOE were both inactive. Dopamine-induced relaxation was not inhibited by (+)-butaclamol (10(-8) M), but was reduced by a potent beta 2-selective adrenoreceptor antagonist ICI 118,551 (2 X 10(-8) M). Our results demonstrate that the rat portal vein is devoid of postsynaptic DA1 and DA2 dopamine receptors…

The influence of dopamine agonists and antagonists on gastric lesions in mice.

The dopamine agonist bromocriptine and L-dopa significantly inhibited whereas dopamine antagonist haloperidol aggravated the gastric lesions induced by pylorus ligation in mice as found earlier for rats. Furthermore, the successful use of a dopamine antagonist alone for the induction of gastric lesions also in mice was demonstrated, since the gastric lesions were induced by a single dose of haloperidol without any additional noxious treatment. Bromocriptine successfully inhibited both the gastric lesion-potentiating as well as the gastric lesion-inducing effect of haloperidol.

Simultaneous voltammetric and chemical monitoring of dopamine release in situ.

The validity of voltammetric monitoring of stimulated dopamine (DA) release was tested by simultaneous chemical assay of the extracellular fluid region next to the tip of a selective Nafion-coated graphite voltammetric electrode. Micro alumina probes were positioned next to the recording electrode tip to adsorb released catecholamines. The catecholamines were desorbed into microvolumes of acid and analyzed by very sensitive HPLC assays to provide unequivocal chemical verification of the electrochemical signals. The results fully confirm that selective voltammetric electrodes can detect release…

Effects of dopamine, isoproterenol, and lobeline on cranial and phrenic motoneurons.

Recent studies have suggested that the upper airway muscles receive a substantial portion of their excitatory input from the peripheral chemoreceptors. We examined the responses of the phrenic, hypoglossal, and recurrent laryngeal nerves to agents that are known to modify carotid body activity. Dopamine, an inhibitor, and isoproterenol and lobeline, two stimulators of carotid body activity, were administered into the lingual arteries of 14 anesthetized, paralyzed cats artificially ventilated with 100% O2. Dopamine decreased the activity of all three nerves (P less than 0.001) but inhibited the…

Interaction of dopamine receptor ligands with subtypes of the opiate receptor.

There are currently several studies in which individual dopamine receptor ligands have been reported to bind with relatively low affinity to opiate receptors. To extend these studies, and to examine the opiate receptor subclass selectivity of such agents, we have examined the ability of six dopamine receptor ligands (prochlorperazine, chlorpromazine, haloperidol, bromocriptine, pimozide and metoclopramide) to compete with four tritiated tracers - [3H]naloxone, [3H][D-Ala2,D-Leu5]enkephalin, [3H]morphine and [3H]ethylketocyclazocine - for binding to rat brain membrane opiate receptors. The dopa…

Effect of naloxone on dopamine uptake and release in vitro in the striatum.

The opiate antagonist naloxone (NX) can inhibit the stereotyped behavior elicited by the directly acting dopamine (DA) agonist apomorphine, while potentiating the same behavior induced by amphetamine, an indirect DA agonist. Since NX does not appear to bind to striatal 3H-DA receptors, these effects may be mediated by an indirect action exerted by naloxone on the nigrostriatal pathway. To confirm this, the effect of NX on DA uptake in striatal homogenates was assessed in vitro, and also the effect of NX on DA release in both striatal synaptosomes and slices. While NX had no effect on its own i…

A comprehensive method for the quantitative determination of dopamine receptor subtypes.

We have demonstrated that three subtypes of dopamine receptors can be characterized using several radioligand binding techniques. Indirect binding assays in which several competing ligands were used to inhibit the binding of the nonselective radioligand spiroperidol resulted in shallow displacement curves with Hill coefficients less than 1. Nonlinear regression analysis of these curves also indicated that there were two subtypes of the D-2 receptor present in a ratio of approximately 3 to 1. Direct binding assays with [3H]alpha-flupenthixol showed that this radioligand nonselectively labeled D…

Cortical tubular and glomerular dopamine receptors in the rat kidney.

Dopamine receptors in glomeruli and renal cortical tubules were characterized using radioligand binding and adenylate cyclase studies. The binding of [3H]haloperidol to glomeruli and tubules was rapid, saturable with time and ligand concentration, reversible, of high affinity, and demonstrated stereoselectivity and antagonist and agonist rank potency for binding to dopamine receptors. Analysis of kinetic data and Rosenthal plots in glomeruli revealed a single class of [3H]haloperidol binding sites with an apparent dissociation constant (Kd) of 6 nM and maximum receptor density (Bmax) of 0.42 p…
