BACKGROUND: Metronidazole (Mtz) is the frontline drug treatment for multiple anaerobic pathogens, including the gastrointestinal protist, Giardia duodenalis. However, treatment failure is common and linked to in vivo drug resistance. In Giardia, in vitro drug-resistant lines allow controlled experimental interrogation of resistance mechanisms in isogenic cultures. However, resistance-associated changes are inconsistent between lines, phenotypic data are incomplete, and resistance is rarely genetically fixed, highlighted by reversion to sensitivity after drug selection ceases or via passage thr

The discovery of antibiotics was one of the greatest achievements in human history, however, antibiotic resistance evolved no later than the introduction of antibiotics. The rapid evolution of antibiotic-resistant pathogens soon became frightening and remained a global healthcare threat. There is an urgent need to have new alternatives or new strategies to combat the multi-drug resistant superbugs such as methicillin-resistant Staphylococcus aureus (MRSA), vancomycin-resistant enterococci (VRE), carbapenem-resistant Pseudomonas aeruginosa (CR-PA), extended-spectrum β-lactamases (ESBL) bearing

It is more than two decades ago that a European Union conference on "The Microbial Threat" hosted by the Danish Government in Copenhagen in September 1998 issued recommendations to encourage good practice in the use of antimicrobial agents and reduce inappropriate prescribing. Essential components of those recommendations were antimicrobial teams in hospitals and the use of feedback to prescribers as well as educational activities. Two decades later, important surveillance systems on both antimicrobial resistance as well as on antibiotic consumption are functioning at the European level and in

Sequences of 105 Aeromonas species plasmids were probed for acquired anti-microbial resistance (AMR) genes using a bioinformatics approach. The plasmids showed no positive linear correlation between size and GC content and up to 55 acquired AMR genes were found in 39 (37%) plasmids after in silico screening for resistance against 15 antibiotic drug classes. Overall, potential multiple antibiotic resistance (p-MAR) index ranged from 0.07 to 0.53. Up to 18 plasmids were predicted to mediate multiple drug resistance (MDR). Plasmids pS121-1a (A. salmonicida), pWCX23_1 (A. hydrophila) and pASP-a58

Multiple myeloma is not a curable disease, and most patients relapse after plateau phase. Drug resistance is a major problem in effective chemotherapy in this kind of disease. Current approaches are aimed at reversing or preventing drug resistance late in the disease. We studied a drug resistance marker, P-glycoprotein (P-gp), in a total of 43 patients with monoclonal gammopathy. This group included eight with monoclonal gammopathy of undetermined significance (MGUS), five with plasmacytoma (PCM), nineteen with multiple myeloma (MM; six newly diagnosed, seven plateau, five refractory, one rela

Drug resistance is a pivotal factor leading to the failure of cancer therapy, within which heme oxygenase-1 (HMOX1) plays a complex and paradoxical dual role. On one hand, HMOX1 protects cancer cells from oxidative damage induced by chemotherapeutic drugs through its antioxidant properties. Concurrently, its catalytic downstream product, carbon monoxide (CO), inhibits cancer cell apoptosis, thereby mediating acquired drug resistance. On the other hand, HMOX1 is a key source of intracellular free iron. When its activity is excessively induced, it leads to the accumulation of excessive free iron

UNLABELLED: Identifying resistance mechanisms to novel antimicrobials informs treatment strategies during infection and antimicrobial development. Studying resistance that develops during the treatment of an infection can provide the most clinically relevant mutations conferring resistance, but cross-sectional studies frequently identify multiple candidate resistance mutations without resolving the driver mutation. We performed whole-genome sequencing of longitudinal Pseudomonas aeruginosa from a patient whose P. aeruginosa developed imipenem/cilastatin/relebactam and ceftolozane/tazobactam re

Rapid and accurate characterization of antimicrobial resistance is essential for effective patient treatment and outcomes. Infection-causing microorganisms often harbour multi-drug resistance, requiring multiple tests for identification. Here, we present a multiplex functional assay using liquid atmospheric pressure (LAP) matrix-assisted laser desorption/ionization (MALDI) as next-generation MALDI biotyping technology, which can accurately determine antibiotic resistance/susceptibility within three hours from <5 µL of bacterial culture, employing a beta-lactam antibiotic panel. Strains with co

Antibiotic combination therapies are an approach used to counter the evolution of resistance; their purported benefit is they can stop the successive emergence of independent resistance mutations in the same genome. Here, we show that bacterial populations with 'mutators', organisms with defects in DNA repair, readily evolve resistance to combination antibiotic treatment when there is a delay in reaching inhibitory concentrations of antibiotic-under conditions where purely wild-type populations cannot. In populations of Escherichia coli subjected to combination treatment, we detected a diverse

Cholangiocarcinoma (CCA) is an aggressive cancer originating from bile duct epithelium. Surgical resection remains the primary curative treatment for CCA. However, most CCA patients are diagnosed at an advanced stage, which limits the applicability of surgical resection. Gemcitabine is widely used as a first-line chemotherapeutic agent for unresectable CCA. Its efficacy is often compromised by the development of drug resistance, which leads to poor clinical outcomes and low survival rates of CCA patients. At present, the mechanisms underlying gemcitabine resistance in CCA remain unclear. This

Once hailed as the cornerstone of modern medicine, antibiotics are now facing a critical reckoning as resistance outpaces innovation. The relentless rise of multidrug-resistant (MDR) pathogens, particularly the notorious ESKAPE group, has rendered many conventional antibiotics increasingly ineffective, transforming once-manageable infections into formidable clinical challenges. In response, researchers are pursuing diverse and inventive strategies to rejuvenate the antibiotic pipeline. Pharmacognostic exploration continues to uncover rare molecular scaffolds from nature's reservoirs, which are

Antibiotics have been considered miracle drugs since the widespread introduction of penicillin, an antimicrobial substance with the potential to save millions of lives. Unfortunately, their excessive use, especially in agriculture and health care, has resulted in the selection and spread of resistant bacterial strains. The accelerated occurrence of antimicrobial resistance is currently a serious global problem, regardless of the level of economic development of individual countries. For this reason, great pressure is placed on developing new substances with an antimicrobial effect or modifying

INTRODUCTION: Infectious diseases associated with intracellular bacteria such as Staphylococcus aureus, Salmonella typhimurium and Mycobacterium tuberculosis are important public health concern. Emergence of multi and extensively drug-resistant bacterial strains have made it even more obstinate to offset such infections. Bacteria residing within intracellular compartments provide additional barriers to effective treatment.
METHOD: Information provided in this review has been collected by accessing various electronic databases including Google scholar, Web of science, Scopus, and Nature index.

BACKGROUND: Until recently, one of the main reasons for mortality has been infectious diseases, and bacteria that are drug-resistant have emerged as a result of the wide application, as well as the misuse of antibacterial medications. Having multidrug-resistance, bacteria present a great problem for the efficient management of bacterial infections and this challenge has resulted in the creation of other means of dealing with bacterial diseases. Of late, metallic nanoparticles (NPs), employed as antibacterial agents, have the potential for use against resistance to bacterial drugs.

Mycobacterium tuberculosis is the major pathogen of tuberculosis (TB). With the growing problem of M. tuberculosis resistant to conventional antibiotics, especially multi-drug resistant tuberculosis (MDR-TB) and extensively-drug resistant tuberculosis (XDR-TB), the need for new TB drugs is now more prominent than ever. Among the promising candidates for anti-TB drugs, anti-mycobacterial peptides have a few advantages, such as low immunogenicity, selective affinity to prokaryotic negatively charged cell envelopes, and diverse modes of action. In this review, we summarize the recent progress in

The fight against multidrug-resistant (MDR) pathogens continues. This article discusses the gap between the need for new antibiotics and lean research and development (R&D) pipelines. Many large pharmaceutical companies have terminated their antibacterial research programmes as they focus on potentially more lucrative therapeutic areas. At the same time, an increasingly dry funding situation hampers smaller start-up companies. Antibacterial innovation proceeds in waves. Following a wave of broad-spectrum antibiotics in the 1980s and 1990s, many companies focused on the development of small-spe

Malaria, caused by Plasmodium parasites, kills 610,000 people annually. The increasing availability of large-scale whole-genome sequencing (WGS) datasets enables investigation of parasite genetic diversity, population structure, and the spread of drug resistance. To investigate Plasmodium falciparum evolution, we analyse 17,565 isolates (17,489 previously published; 76 newly sequenced) from 39 countries, using identity-by-descent (IBD) networks, population structure inference, and selection scans. Analyses reveal distinct patterns of selection, transmission, and drug resistance. Selection ana
