The flathead mutation causes CNS-specific developmental abnormalities and apoptosis.

We describe a new mutation, flathead (fh), that arose spontaneously in an inbred colony of Wistar rats. The mutation is autosomal recessive, and the behavioral phenotype of fh/fh rats includes spontaneous seizures, tremor, impaired coordination, and premature death. A striking feature of the fh mutation is a dramatic reduction in brain size (40% of normal at birth). In contrast, no abnormalities are evident in the peripheral nervous system or in other tissues outside of the CNS. Although bromodeoxyuridine incorporation assays indicate that the rate of cell proliferation in the fh/fh cortex is

Trigeminal ganglion axons are repelled by their presumptive targets.

Previous work suggested that in mouse, presumptive targets of the trigeminal ganglion, rather than intermediate structures, attract pioneer axons from the time their growth cones exit the ganglion (Lumsden and Davies, 1986). In rat we find that some presumptive targets repel trigeminal axons. The repellant activity is concentrated in the anterior and ventral epithelium of the mandibular arch at embryonic day 12 (E12) and was also present in the maxillary arch. The activity is blocked by anti-neuropilin-1. E13 mandible explants repel trigeminal axons during the first day of outgrowth in vitro,

Contributions of heterospecific tissue recombinations to odontogenesis.

Determining the functions of cell surface and substrate adhesion molecules during heterospecific tissue recombinations constitutes a significant problem in biology. The developing tooth organ provides a fine model to pursue this problem, especially in the context of instructive epithelial-mesenchymal interactions. The interpretation of many experimental heterospecific tissue combinations is difficult because of the complexity of the embryonic systems. According to some authors, the expressed phenotype corresponds to the genotype of the epithelium; on the contrary, other studies have demonstrat

Development of neurotransmitter systems in the mouse embryo following acute ethanol exposure: a histological and immunocytochemical study.

Acute maternal ethanol administration to C57B1/6J mice on gestational day 7 (GD7) results in facial and brain abnormalities similar to those reported in human fetal alcohol syndrome (FAS). Using this model, we assessed the damage to brain structures using histological methods and changes in developing neurotransmitter systems with immunocytochemistry. Cholinergic neurons in the forebrain were stained with a monoclonal antibody to choline acetyltransferase (ChAT). Catecholaminergic neurons in the midbrain and serotoninergic neurons in the hindbrain were stained with polyclonal antisera to tyros

Urachal Carcinoma with Divergent Glandular Enteric-Type and Squamous Differentiation Associated with Bladder Exstrophy: Case Report of an Extremely Rare Entity.

Background and Clinical Significance: Urachal carcinoma (UrC) is an uncommon neoplasm derived from residual embryonic structures connecting the bladder to the umbilicus. Owing to its rarity, deep anatomic location, and histologic overlap with other glandular malignancies, accurate diagnosis remains challenging. Congenital anomalies of the lower urinary tract, including bladder exstrophy, are recognized as conditions that may predispose to malignant transformation of urachal remnants, although documented cases remain scarce. Case presentation: We describe the case of a 52-year-old male with bla…

A concise overview of mammalian spermatogenesis.

The male gamete, the spermatozoon, is the carrier of paternal inheritance. Besides paternal genome, the spermatozoon contributes essential factors for successful fertilization, embryonic development, and continuation of species. A dynamic and extraordinarily complex process called spermatogenesis produces spermatozoa capable of maturing and acquiring full fertilizing capacity. Spermatogenesis proceeds through a series of events that involve cell division (proliferative phase assuring continuous restocking of spermatogenic stem cells), meiosis (recombination and haploidization of paternal genom

SNED1 fibrillar assembly in the extracellular matrix requires fibronectin and collagen I.

The extracellular matrix (ECM) is a meshwork of proteins that orchestrates a broad range of cellular phenotypes, including proliferation, adhesion, migration and differentiation. SNED1 is a newly characterized ECM glycoprotein that promotes cell adhesion and is essential for embryonic development. Its upregulation is also associated with breast cancer metastasis and poor prognosis for breast cancer patients. We have recently shown that SNED1 assembles into fibrillar structures, but the mechanisms guiding its incorporation into the ECM scaffold remained unknown. Here, combining biochemical assa

Genetic variation near ROBO1 is associated with craniofacial microsomia and related phenotypes in the Finnish population.

Craniofacial microsomia (CFM) encompasses a phenotypic continuum of congenital anomalies ranging from isolated microtia to more complex manifestations within the oculo-auriculo-vertebral spectrum, including Goldenhar syndrome, reflecting abnormal development of first and second pharyngeal arch-derived structures. While several genetic susceptibility loci have been reported, population-based evidence in individuals of European ancestry remains limited. Using nationwide data from FinnGen in the Finnish founder population, we identified a genome-wide significant association at a conserved interge

Migrasomes: new communicators between cells.

Migrasomes are a recently discovered class of organelles, typically generated at the intersections and tips of retraction fibers (RFs) in migrating cells. These monolayer vesicular structures encapsulate a variety of bioactive molecules and have been shown to participate in essential physiological processes, including intercellular communication, embryonic development, immune microenvironment modulation, and mitochondrial homeostasis. Beyond their physiological roles, accumulating evidence has revealed that migrasomes are also closely associated with the pathogenesis of various diseases. These

Adamantane-appended koneramine metal complexes: anticancer activity and therapeutic potential.

Adamantane-based compounds have gained significant interest in drug discovery due to their versatile therapeutic potential and hydrophobic character, which allows for structural modifications across a wide range of drug classes. Their distinct structural and physicochemical properties continue to make them valuable assets in the design and development of new pharmaceuticals. Herein, we report the synthesis of three new metal complexes derived from an adamantane-appended tridentate koneramine ligand (LkAd), coordinated with Cu(II), Zn(II), and Fe(III). These complexes exhibit good stability and

Delayed gonadal development of estrogen-induced ZZ females in Chinese soft-shelled turtles†.

Exogenous estrogen induces sex reversal during embryonic development in various animal species including reptiles, yet the mechanisms underlying post-reversal gonadal development remain poorly understood. In this study, we generated ZZ female Chinese soft-shelled turtle Pelodiscus sinensis through 17β-estradiol treatment and investigated their reproductive traits and sex-specific gene expression at 1, 3, 6, 8, and 12 months post-hatching. Our results showed that by 12 months, ZZ females maintained ovarian structures and exhibited low expression levels of the male-specific genes SOX9 and DMRT1.

Cell fusion dynamics: mechanisms of multinucleation in osteoclasts and macrophages.

Cell-cell fusion is a vital biological process where the membranes of two or more cells merge to form a syncytium. This phenomenon is critical in various physiological and pathological contexts, including embryonic development, tissue repair, immune responses, and the progression of several diseases. Osteoclasts, which are cells from the monocyte/macrophage lineage responsible for bone resorption, have enhanced functionality due to cell fusion. Additionally, other multinucleated giant cells (MGCs) also arise from the fusion of monocytes and macrophages, typically during chronic inflammation an

Human Retinal Organoids in Therapeutic Discovery: A Review of Applications.

Human embryonic stem cells (hESCs)- and induced pluripotent stem cells (hiPSCs)-derived retinal organoids (ROs) are three-dimensional laminar structures that recapitulate the developmental trajectory of the human retina. The ROs provide a fascinating tool for basic science research, eye disease modeling, treatment development, and biobanking for tissue/cell replacement. Here we review the previous studies that paved the way for RO technology, the two most widely accepted, standardized protocols to generate ROs, and the utilization of ROs in medical discovery. This review is conducted from the

Bioengineering Human Pluripotent Stem Cell-Derived Retinal Organoids and Optic Vesicle-Containing Brain Organoids for Ocular Diseases.

Retinal organoids are three-dimensional (3D) structures derived from human pluripotent stem cells (hPSCs) that mimic the retina's spatial and temporal differentiation, making them useful as in vitro retinal development models. Retinal organoids can be assembled with brain organoids, the 3D self-assembled aggregates derived from hPSCs containing different cell types and cytoarchitectures that resemble the human embryonic brain. Recent studies have shown the development of optic cups in brain organoids. The cellular components of a developing optic vesicle-containing organoids include primitive

Identification of two AMH gene variants in two unrelated patients with persistent Müllerian duct syndrome: one novel variant.

INTRODUCTION: Persistent müllerian duct syndrome (PMDS) is a rare form of 46, XY disorder of sex development characterized by the persistence of the müllerian structures (uterus, fallopian tubes, the upper part of the vagina) in phenotypically and genotypically normal males. This disease occurs as a result of impairment in the synthesis, release or effect of anti-Müllerian hormone (AMH) during the embryonic period. Approximately 85-88% of PMDS cases have been reported to have AMH or AMHRII mutation. CASE: Herein, we report two PMDS cases from unrelated two families who presented with bilatera

Excitatory neuronal CHD8 in the regulation of neocortical development and sensory-motor behaviors.

CHD8 (chromodomain helicase DNA-binding protein 8) is a chromatin remodeler associated with autism spectrum disorders. Homozygous Chd8 deletion in mice leads to embryonic lethality, making it difficult to assess whether CHD8 regulates brain development and whether CHD8 haploinsufficiency-related macrocephaly reflects normal CHD8 functions. Here, we report that homozygous conditional knockout of Chd8 restricted to neocortical glutamatergic neurons causes apoptosis-dependent near-complete elimination of neocortical structures. These mice, however, display normal survival and hyperactivity, anxio

Generation of mature compact ventricular cardiomyocytes from human pluripotent stem cells.

Compact cardiomyocytes that make up the ventricular wall of the adult heart represent an important therapeutic target population for modeling and treating cardiovascular diseases. Here, we established a differentiation strategy that promotes the specification, proliferation and maturation of compact ventricular cardiomyocytes from human pluripotent stem cells (hPSCs). The cardiomyocytes generated under these conditions display the ability to use fatty acids as an energy source, a high mitochondrial mass, well-defined sarcomere structures and enhanced contraction force. These ventricular cells
