Molecular autopsy and family screening in a young case of sudden cardiac death reveals an unusually severe case of FHL1 related hypertrophic cardiomyopathy.

BACKGROUND: Hypertrophic cardiomyopathy (HCM) is a genetic cardiomyopathy with a prevalence of about 1:200. It is characterized by left ventricular hypertrophy, diastolic dysfunction and interstitial fibrosis; HCM might lead to sudden cardiac death (SCD) especially in the young. Due to low autopsy frequencies of sudden unexplained deaths (SUD) the true prevalence of SCD and especially of HCM among SUD remains unclear. Even in cases of proven SCD genetic testing is not a routine procedure precluding appropriate risk stratification and counseling of relatives. METHODS: Here we report a case of

Individual and family level factors associated with physical and mental health-related quality of life among people living with HIV in rural China.

BACKGROUND: One of the major challenges faced by people living with HIV (PLH) is the compromised quality of life due to the negative impact of HIV on their health. HIV/AIDS control effort should go beyond laboratory and lay more emphasis on improving the health-related quality of life (HRQoL) for PLH. The objective of this study is to evaluate the physical and mental HRQoL of PLH in rural China, and explore the relationship between HRQoL and individual- and family-level factors. METHODS: A cross-sectional study was conducted among 522 PLH in Anhui, China. Participant's sociodemographic charac

An intensive family-based treatment guided intervention for medically hospitalized youth with anorexia nervosa: Parental self-efficacy and weight-related outcomes.

Recent studies suggest the efficacy of family-based treatment (FBT) among youth with anorexia nervosa (AN) in intensive treatment settings. This study aimed to assess weight outcomes in youth who received an FBT intervention while hospitalized for medical complications of AN. Parental self-efficacy among participating caregivers was also measured. Post-discharge weights of 49 participants were compared with weights of 44 youth who were hospitalized prior to the provision of the FBT intervention. Youth who received the FBT intervention gained significantly more weight than youth in the retrospe

Meta-analysis of the correlation between the rs17401966 polymorphism in kinesin family member 1B and susceptibility to hepatitis B virus related hepatocellular carcinoma.

BACKGROUND/AIMS: The association between the kinesin family member 1B (KIF1B) gene polymorphism and the risk of hepatitis B virus-related hepatocellular carcinoma (HCC) has been investigated in many peer-reviewed studies. However, scholars have failed to replicate these results in validation tests. The purpose of the present study was to explore whether the KIF1B rs17401966 polymorphism was associated with susceptibility to HCC. METHODS: The results of case-controlled studies on the correlation between the KIF1B rs17401966 polymorphism and HCC susceptibility were collected using Google Schola

Trends in Alcohol's Harms to Others (AHTO) and Co-occurrence of Family-Related AHTO: The Four US National Alcohol Surveys, 2000-2015.

Various harms from others' drinking have been studied individually and at single points in time. We conducted a US population 15-year trend analysis and extend prior research by studying associations of depression with combinations of four harms - family/marriage difficulties, financial troubles, assault, and vandalism - attributed to partners or family members. Data come from four National Alcohol Surveys conducted by telephone in 2000, 2005, 2010, and 2015 (analytic sample = 21,184). Weighted logistic regression models estimated time trends adjusting for victim characteristics (gender, age,

The role of Misshapen NCK-related kinase (MINK), a novel Ste20 family kinase, in the IRES-mediated protein translation of human enterovirus 71.

Human Enterovirus 71 (EV71) commonly causes Hand, Foot and Mouth Disease in young children, and occasional occurrences of neurological complications can be fatal. In this study, a high-throughput cell-based screening on the serine/threonine kinase siRNA library was performed to identify potential antiviral agents against EV71 replication. Among the hits, Misshapen/NIKs-related kinase (MINK) was selected for detailed analysis due to its strong inhibitory profile and novelty. In the investigation of the stage at which MINK is involved in EV71 replication, virus RNA transfection in MINK siRNA-tre

Molecular genetic analysis of the PLP1 gene in 38 families with PLP1-related disorders: identification and functional characterization of 11 novel PLP1 mutations.

BACKGROUND: The breadth of the clinical spectrum underlying Pelizaeus-Merzbacher disease and spastic paraplegia type 2 is due to the extensive allelic heterogeneity in the X-linked PLP1 gene encoding myelin proteolipid protein (PLP). PLP1 mutations range from gene duplications of variable size found in 60-70% of patients to intragenic lesions present in 15-20% of patients. METHODS: Forty-eight male patients from 38 unrelated families with a PLP1-related disorder were studied. All DNA samples were screened for PLP1 gene duplications using real-time PCR. PLP1 gene sequencing analysis was perfor

Novel homozygous mutation in DSP causing skin fragility-woolly hair syndrome: report of a large family and review of the desmoplakin-related phenotypes.

Desmoplakin is an important cytoskeletal linker for the function of the desmosomes. Linking desmoplakin to certain types of cardiocutaneous syndromes has been a hot topic recently. Skin fragility-woolly hair syndrome is a rare autosomal recessive disorder involving the desmosomes and is caused by mutation in the desmoplakin gene (DSP). We report five members from a large family with skin fragility-woolly hair syndrome. The index is a 14-year-old girl with palmoplantar keratoderma, woolly hair, variable alopecia, dystrophic nails, and excessive blistering to trivial mechanical trauma. No cardia

Glucocorticoid-induced TNF receptor family related gene activation overcomes tolerance/ignorance to melanoma differentiation antigens and enhances antitumor immunity.

Glucocorticoid-induced TNF receptor family related protein (GITR) is present on many different cell types. Previous studies have shown that in vivo administration of an anti-GITR agonist mAb (DTA-1) inhibits regulatory T cells (Treg)-dependent suppression and enhances T cell responses. In this study, we show that administration of DTA-1 induces >85% tumor rejection in mice challenged with B16 melanoma. Rejection requires CD4+, CD8+, and NK1.1+ cells and is dependent on IFN-gamma and Fas ligand and independent of perforin. Depletion of Treg via anti-CD25 treatment does not induce B16 rejection,

Analysis of ileal sodium/bile acid cotransporter and related nuclear receptor genes in a family with multiple cases of idiopathic bile acid malabsorption.

The etiology of most cases of idiopathic bile acid malabsorption (IBAM) is unknown. In this study, a Swedish family with bile acid malabsorption in three consecutive generations was screened for mutations in the ileal apical sodium-bile acid cotransporter gene (ASBT; gene symbol, SLC10A2) and in the genes for several of the nuclear receptors known to be important for ASBT expression: the farnesoid X receptor (FXR) and peroxisome proliferator activated receptor alpha (PPARalpha). The patients presented with a clinical history of idiopathic chronic watery diarrhea, which was responsive to choles

Health-Related Quality of Life and Family Impact in Young Children with Infantile Epileptic Spasms Syndrome: A Cross-Sectional Caregiver-Reported Study.

Background/Objectives: Infantile epileptic spasms syndrome (IESS) is a developmental and epileptic encephalopathy characterized by epileptic spasms and epileptiform EEG abnormalities. Classical West syndrome represents a phenotype within the IESS spectrum characterized by epileptic spasms, hypsarrhythmia, and developmental plateau or regression. Beyond seizure control, assessment of health-related quality of life (HRQoL) and family impact may provide important information on the long-term burden of the disease. This study aimed to characterize caregiver-reported HRQoL in young children with IE

Normal electrocardiography at presentation masks SCN5A-related long QT syndrome: the importance of Holter monitoring in children with a family history of Brugada syndrome.

A 10-year-old girl who showed no abnormalities on resting or exercising stress electrocardiograms underwent Holter monitoring because of a family history of Brugada syndrome, which revealed marked QT prolongation during sleep. SCN5A-related inherited arrhythmia was suspected, and genetic testing confirmed the p.Glu1783Lys variant in SCN5A. Children with a family history of Brugada syndrome should undergo Holter monitoring proactively, suspecting the possibility of SCN5A-related inherited arrhythmia, even if 12-lead electrocardiograms show no abnormalities.

Transactional associations of health-related worries among Asian families: Evidence from six-wave longitudinal data during COVID-19.

During public health crises, family members often experience heightened health-related worry. However, little is known about how the transactional processes of such worry operate within family systems involving mother, father, and their child. To address this gap, we conducted a six-wave longitudinal survey of 303 families in China. Each month, mothers (Mage = 33.35 years) and fathers (Mage = 34.97 years) of children (Mage = 3.98 years, age range 2-7 years, 55.6% male) reported their own and their perceptions of their child's COVID-19-related health worries (e.g., infection concerns). Self-rated functioning in family, social, and social-life domains in relation to reasons for living among young adults: a cross-sectional study.

INTRODUCTION: Suicide prevention research has increasingly emphasized protective factors in addition to risk factors. Reasons for living represent a protective construct in suicidology, reflecting beliefs and commitments that may discourage suicidal behavior. However, less is known about how self-rated functioning in family, social, and social-life domains is associated with reasons for living among young adults. METHODS: An anonymous cross-sectional online survey using non-probability convenience sampling was conducted between March and June 2025 among 546 young adults aged 18-35 years in Po
