Glutathione reductase activity, riboflavin status, and disease activity in rheumatoid arthritis.

OBJECTIVE: To measure erythrocyte glutathione reductase (EGR) activity and riboflavin status, and their relations to disease activity, in rheumatoid arthritis patients compared to healthy controls. METHODS: Patients with rheumatoid arthritis were classified as active if there were features of articular inflammation which required initiation or change of disease modifying therapy, and as inactive if they had little evidence of articular inflammation. EGR was measured in patients and healthy controls by a functional assay with or without the addition of flavin adenine dinucleotide (FAD). The ra

X-ray structure of kijd3, a key enzyme involved in the biosynthesis of D-kijanose.

D-kijanose is an unusual nitrosugar found attached to the antibiotic kijanimicin. Ten enzymes are required for its production in Actinomadura kijaniata, a soil-dwelling actinomycete. The focus of this investigation is on the protein encoded by the kijd3 gene and hereafter referred to as KijD3. On the basis of amino acid sequence analyses, KijD3 has been proposed to be an FAD-dependent oxidoreductase, which catalyzes the sixth step in d-kijanose biosynthesis by converting dTDP-3-amino-2,3,6-trideoxy-4-keto-3-methyl-d-glucose into its C-3' nitro derivative. This putative activity, however, has n

A gene encoding a putative FAD-dependent L-2-hydroxyglutarate dehydrogenase is mutated in L-2-hydroxyglutaric aciduria.

The purpose of this study was to identify the biochemical and genetic defect in L-2-hydroxyglutaric aciduria, a neurometabolic disorder characterized by the presence of elevated concentrations of L-2-hydroxyglutaric acid in urine, plasma, and cerebrospinal fluid. Evidence is provided for the existence in rat tissues of a FAD-dependent enzyme catalyzing specifically the oxidation of L-2-hydroxyglutarate to alpha-ketoglutarate. This enzyme is mainly expressed in liver and kidney but also at lower levels in heart, brain, and other tissues. Subcellular fractionation indicates that the liver enzyme

Isolation and properties of L-amino acid oxidase from Ophiophagus hannah venom.

This paper reports the isolation and some properties of L-amino acid oxidase from Ophiophagus hannah venom. The differences between L-amino acid oxidase from Ophiophagus hannah and that from other sources in specific activity, properties and the spectrum of isozymes are noticeable. The result of electrophoresis on polyacrylamide gel shows that this purified enzyme is homogenous. The molecular weight determined by gradient polyacrylamide gel slab (4--30%) is around 15 X 10(4) dalton. The molecular weight of the subunit determined by SDS gradient gel electrophoresis (4--30%) is around 7.3 X 10(4

Arginine substitution of conserved Lys609 and Lys733 impairs FMN dynamics, electron transfer, and nitric oxide production in endothelial nitric oxide synthase.

Endothelial nitric oxide synthase (eNOS) produces nitric oxide (NO), a key molecule for maintaining vascular health. While phosphorylation is a well-established regulatory mechanism of eNOS activity, the functional contribution of conserved lysine residues to electron transfer and catalytic coupling remains less clearly defined. In this study, we examined two conserved lysines in eNOS, Lys609 located within the autoinhibitory (AI) region and Lys733 positioned within the FMN-FNR hinge, by substituting them with arginine to preserve positive charge while altering side-chain geometry. Biochemical

Native Fluorescence Characterization of Blood Plasma for Early Diagnosis of Oral Potentially Malignant Disorders.

Oral potentially malignant disorders (PMD) are heterogenous group of oral conditions characterized by increased risk of malignant transformation that are commonly diagnosed clinically and treated. Tissue biopsy is an invasive tool for diagnosis of oral PMDs. Native fluorescence spectroscopy of biofluids has been considered as an alternative and/or complimentary tool in the characterization of tissues of various pathological conditions. In the present study, it is primarily aimed to characterize different fluorophores present in the blood plasma of normal subjects and PMD patients, using excita

Reductive half-reaction of xanthine oxidase: mechanistic role of the species giving rise to the "rapid type 1" molybdenum(V) electron paramagnetic resonance signal.

The reaction of xanthine oxidase with xanthine, 1-methylxanthine, and 2-hydroxy-6-methylpurine has been reinvestigated with the aim of elucidating the mechanistic role of the species giving rise to the "rapid" Mo(V) electron paramagnetic resonance (EPR) signal. It is found that addition of 2.0 mM 1-methylxanthine or 2-hydroxy-6-methylpurine to partially reduced enzyme generates substantial amounts of the Type 1 form of the "rapid" EPR signal, characterized by superhyperfine coupling to one strongly interacting (aav = 13 G) and one weakly interacting (aav = 3 G) proton. The "rapid" signals obse

Nature of the effect of adenosine 5'-monophosphate on the cyanide-insensitive respiration in mitochondria of Moniliella tomentosa.

The alternative oxidase of Moniliella tomentosa mitochondria is stimulated by 5'-AMP. This effect may be masked, depending on the isolation procedure of the mitochondria. The preparation of submitochondrial particles results in the expression of the 5'-AMP effect. Two more methods are now described to reveal the 5'-AMP effect whenever it would be masked: (1) switching on the myokinase activity of the mitochondria to deplete them of endogenous 5'-AMP; (2) using detergents (sodium dodecyl sulphate, sodium deoxycholate) in a controlled detergent:protein ratio, or chloroform. The alternative oxida

Deficient cytochrome b5 reductase activity in nontoxic goiter with iodide organification defect.

A 37-yr-old woman with nontoxic goiter is presented. The thyroid 131I uptake at 3 and 24 hr were, respectively, 77.1% and 81.4% dose. Thiocyanate discharged 65.5% of the accumulated 131I in 30 min. In vitro organification of iodine in the thyroid homogenate from the patient was impaired and it was restored to normal by the addition of H2O2, glucose, and glucose oxidase system, FAD, or reduced cytochrome b5. Riboflavin, FMN, oxidized cytochrome b5, oxidized or reduced cytochrome c, NAD(H), and NADP(H) were ineffective in the reaction. The microsomal NADH-cytochrome b5 reductase activity was def

The Riboflavin Kinase Domain Is Essential for FMNAT Catalysis: A Linker-Dependent Regulatory Mechanism in Staphylococcus aureus FAD Synthetase.

Flavin adenine dinucleotide (FAD) synthetase (FADS) from Staphylococcus aureus is a bifunctional enzyme comprising an N-terminal FMN adenylyltransferase (FMNAT) domain that catalyzes FAD synthesis and a C-terminal riboflavin kinase (RFK) domain responsible for riboflavin phosphorylation. While eukaryotic FMNAT and RFK homologs function as independently, the mechanistic basis for their fusion and interdependence in bacteria remains unclear. Here, we show that S. aureus FADS functions as an obligate structural assembly exhibiting pronounced functional asymmetry between its two domains. The isola
