We evaluated the performance of the Emit tox benzodiazepine and barbiturate assays (Dade Behring) and fluorescence polarisation immunoassay (FPIA) (Abbott) for use with serum determinations in preliminary therapeutic drug monitoring or acute drug intoxication detection. INTRODUCTION: The toxicity profile of the amphetamine derivative 3,4-methylenedioxymethamphetamine (MDMA, "Ecstasy") is well known. This designer drug is usually taken at "house parties" and may cause severe complications, sometimes leading to death, even when taken in relatively small units (1 or 2 tablets). Up to now, only a few cases of survival after ingestion of an overdose of Ecstasy have been described. In most cases the users developed hyperthermia, disseminated intravascular coagulation, rhabdomyolysis, and renal failure.

Slow voltage-sensitive dyes work by accumulating in brain tissue and report the average membrane potential of neurons and glia. The voltage-sensitive dye diO-C2-5 was used to monitor the polarization state of 27 brain structures in the rat during a systemically induced, behaviorally mild, kainic acid seizure using a 20 s recording period. The effect of two mercury salts, HgCl2 and Hg(NO3)2, on the thermotropic properties of phosphatidylserine (PS) model membranes and sonicated rat red cell membranes was investigated by fluorescence polarization. Both Hg(II) salts abolished the phase transition and decreased the membrane fluidity by interacting with PS.
