Dual Nurr1/RXR agonism of valerenic acid and synthetic mimetics enables dimer-selective Nurr1 modulation.

Nuclear receptor related 1 (Nurr1) is a neuroprotective transcription factor emerging as promising target in Parkinson's disease and multiple sclerosis. It can act in three oligomeric forms as monomer, homodimer and heterodimer on different DNA response elements. We hypothesized that dual Nurr1 and RXR activation might enable selective modulation of Nurr1 in its heterodimeric form. A search for dual ligands revealed valerenic acid and a synthetic mimetic as Nurr1 and RXR activators. Biochemical and cellular characterization demonstrated that dual agonism destabilized the Nurr1 homodimer but le

Targeted Approaches to Treatment of Pleural Mesothelioma: A Review.

Pleural mesothelioma is an aggressive disease that is enriched for inactivating alterations in tumor suppressor genes. Systemic therapeutic strategies for pleural mesothelioma generally involve chemotherapies and immunotherapies that are chosen without consideration of the tumor's molecular profile. As this generalized approach to treatment rarely yields durable responses, alternative therapeutic regimens are urgently indicated. Preclinical studies have identified synthetic lethal protein and metabolic interactions, recurrently overexpressed proteins, and frequent pathway perturbations that ma

Behavioral and ecotoxicology of sulfonamide metabolites in the aquatic environment.

Sulfonamides (SAs) are the first broad-spectrum synthetic antimicrobial agents used in human health and veterinary medicine. The majority of SAs entering human body is discharged into aquatic environment in the form of parent material or metabolites. The residues of SAs and their metabolites in the aquatic environment and the development of drug resistance can be serious threats to ecosystems and human health. We summarized recent advances in the research of SAs. The main metabolite types of SAs and the distribution characteristics of metabolites in different aquatic environments were introduc

Wnt/Ca2+/NFAT signaling maintains survival of Ph+ leukemia cells upon inhibition of Bcr-Abl.

Although Bcr-Abl kinase inhibitors have proven effective in the treatment of chronic myeloid leukemia (CML), they generally fail to eradicate Bcr-Abl(+) leukemia cells. To identify genes whose inhibition sensitizes Bcr-Abl(+) leukemias to killing by Bcr-Abl inhibitors, we performed an RNAi-based synthetic lethal screen with imatinib mesylate in CML cells. This screen identified numerous components of a Wnt/Ca(2+)/NFAT signaling pathway. Antagonism of this pathway led to impaired NFAT activity, decreased cytokine production, and enhanced sensitivity to Bcr-Abl inhibition. Furthermore, NFAT inhi

[Protection of radiosensitive human cells against the action of heavy metals by antimutagens and adapting factors: association with genetic and protein polymorphisms].

Cells of a diploid line obtained from embryos with the Down's syndrome, known to be unable to repair gamma-induced DNA damage, were treated with natural (garlic extract, retinol) and synthetic (crown compound) antimutagens and with adapting factors (heat shock, low CdCl2 concentrations, 10(-8) M). The protective effect was evaluated by registering DNA breaks and cell survival, and the protection coefficients were calculated. The most effective results were obtained with the use of the garlic extract and retinol. No protection of the DNA structure was observed when cells were treated with low c

Current Perspectives on STING Agonists for Anticancer Drug Development.

The cyclic GMP-AMP (cGAS) synthase and stimulator of interferon genes signaling pathway plays a central role in bridging innate and adaptive immunity, particularly within the context of cancer. Activation begins when cytosolic double-stranded DNA is detected, leading to the production of cyclic GMP-AMP (cGAMP) and subsequent activation of STING. This initiates a cascade that induces the expression of type I interferons (IFN) and proinflammatory cytokines, enhancing antitumor immune responses through the stimulation of dendritic cells, cytotoxic T lymphocytes, and natural killer cells. Despite

Decoding next-generation heavy metal bioremediation via species-specific Earthworm and its gut microbiome interactions: insights from molecular responses, multi-omics, synthetic biology, and artificial intelligence.

Heavy metal (HM) contamination represents a persistent global threat, demanding bioremediation strategies that are both mechanistically robust and ecologically sustainable. This review provides a next-generation perspective on vermiremediation by integrating species-level physiology, gut microbiome functionality, molecular detoxification pathways, synthetic biology innovations, multi-omics insights, and artificial intelligence (AI)-driven modeling into a unified framework. A central novelty of this work lies in the detailed elucidation of earthworm-microbe consortia and their synergistic contr

De novo designed 3-helix bundle peptides and proteins with controlled topology and stability.

Computational protein design is advancing rapidly. However, approaches and methods are needed to increase success rates and to elaborate designs. Here we describe the combination of rational and computational design to deliver three-helix bundle (3HB) peptide assemblies and single-chain proteins with control over topology and thermal stability. First, we garner sequence-to-structure relationships from antiparallel 3HBs in the Protein Data Bank. This gives core-packing rules, including layers of hydrogen-bonded polar residues, which are combined with surface-charge patterning to design compleme

Inhibition of anti-inflammatory cytokines, IL-10 and TGF-β, in Leishmania major infected macrophage by miRNAs: A new therapeutic modality against leishmaniasis.

Leishmania major (L. major) applies several mechanisms to escape the immune system. Interleukin-10 (IL-10) and Transforming Growth Factor (TGF-β) downregulate nitric oxide synthase (iNOS) leading to the survival of Leishmania within macrophages. The miRNAs are known as the modulators of the immune system. The present study was conducted to assess the effect of synthetic miR-340 mimic on cytokines (IL-10 and TGF-β1) involved in L. major infected macrophages. The miRNAs targeting of IL-10 and TGF-β1 was predicted using bioinformatic tools. Relative expression of predicted miRNA, IL-10, and TGF-β

[Melatonin, synthetic analogs, and the sleep/wake rhythm].

INTRODUCTION: Melatonin, a widespread hormone in the animal kingdom, is produced by several organs and tissues besides the pineal gland. Whilst extrapineal melatonin behaves as a cytoprotective molecule, the pineal produces the hormone in a rhythmic manner. The discovery of melatonin in 1958, and the characterization of its synthesis somewhat later, let to the description of its photoperiodic regulation and its relationship with the biological rhythms such as the sleep/wake rhythm. DEVELOPMENT: The suprachiasmatic nuclei are the anatomical seat of the biological clock, represented by the cloc

Engineering DNA-Targeting CRISPR and CRISPR-Like Effectors: Advances from Rational Design and High-Throughput Screening to AI-Driven Development.

Clustered regularly interspaced short palindromic repeats (CRISPR) and CRISPR-associated (Cas) proteins constitute adaptive immune systems in prokaryotes and have transformed life sciences, precision medicine, and synthetic biology as programmable genome-editing tools. Despite their broad utility, naturally occurring DNA-targeting Cas effectors remain constrained by several intrinsic limitations, including large protein size that complicates delivery, stringent protospacer adjacent motif (PAM) requirements that restrict targetable genomic space, and mismatch tolerance that can lead to off-targ

Engineering responsive polymer building blocks with host-guest molecular recognition for functional applications.

CONSPECTUS: All living organisms and soft matter are intrinsically responsive and adaptive to external stimuli. Inspired by this fact, tremendous effort aiming to emulate subtle responsive features exhibited by nature has spurred the invention of a diverse range of responsive polymeric materials. Conventional stimuli-responsive polymers are constructed via covalent bonds and can undergo reversible or irreversible changes in chemical structures, physicochemical properties, or both in response to a variety of external stimuli. They have been imparted with a variety of emerging applications inclu

Novel gene isolated from Caligus rogercresseyi: a promising target for vaccine development against sea lice.

Sea lice (Copepoda, Caligidae) are the most widely distributed marine pathogens in the salmon industry in the last 30 years. Caligus rogercresseyi is the most important species affecting Chile's salmon industry. Vaccines against caligid copepods have the potential to be a cost-effective means of controlling the infestation and avoid many of the disadvantages of medicine treatments. However, research in the development of such vaccines has begun only recently and approaches used thus far have met with little or no success. In the present study, we characterized a novel gene (denoted as my32) fr

Microbial consortia for pesticide biodegradation: mechanisms, cross-class pathways, and translational challenges.

The use of synthetic pesticides estimated at 4.1 million metric tons annually worldwide, has led to widespread contamination of soils and aquatic environments, with documented risks to ecosystem integrity and human health. Physicochemical remediation methods are costly and often generate secondary pollutants and toxic intermediates, making microbial bioremediation a lower-residue alternative to physicochemical treatment. Because pesticide degradation in natural environments is rarely achieved by single microbial species, this review critically synthesizes evidence on multi-species microbial co

Bioinspired and Biomimetic Nanocarriers: Advances in Exosomes and Cell-Membrane-Coated Systems for Therapeutic Applications.

The translation of nanomedicine from bench to bedside has been thwarted by the limitations of conventional synthetic nanoparticles including rapid mononuclear phagocyte system (MPS) clearance, lack of target specificity, immunogenicity and poor intracellular delivery. To overcome these limitations, bioinspired and biomimetic nanocarriers are emerging as the next generation of therapeutic platforms. Bioinspired systems replicate the core biological design principles with engineered synthetic materials and biomimetic platforms use natural biological components such as cell membranes and extracel

Bacteria-algae synergy in carbon sequestration: Molecular mechanisms, ecological dynamics, and biotechnological innovations.

Rising atmospheric CO₂ levels require innovative strategies to increase carbon sequestration. Bacteria-algae interactions, as pivotal yet underexplored drivers of marine and freshwater carbon sinks, involve multiple mechanisms that amplify CO₂ fixation and long-term storage. This review systematically describes the synergistic effects of bacteria-algae consortia spanning both microalgae (e.g., Chlorella vulgaris and Phaeodactylum tricornutum) and macroalgae (e.g., Macrocystis and Laminaria) on carbon sequestration. These effects include (1) molecular-level regulation (e.g., signal transduction

Taxonomic and functional fingerprints in the gut microbiota of Iberian hare (Lepus granatensis) inhabiting organic and conventional farming fields.

Intensive farming leading to both landscape homogenization and massive use of pesticides is threatening biodiversity associated with agricultural landscapes. Pesticides may alter the composition of the gut microbiota, which contributes critically to a variety of host metabolic and immune functions, and the consequences on wildlife health are still unknown. We studied potential effects of farming practices on the gut microbiota of wild Iberian hare populations (Lepus granatensis) inhabiting conventional (synthetic pesticide-treated, mostly glyphosate) and organic (synthetic pesticide-free) farm

Cellular behavior and extracellular matrix turnover in bovine annulus fibrosus cells under hydrostatic pressure and deviatoric strain.

Intervertebral disc herniation is a common spinal disorder that is often treated with discectomy when conservative measures fail. To devise therapeutic strategies for tears in the annulus fibrosus (AF), the regenerative capability of AF cells under spinal loading needs to be addressed. We hypothesized that the compressive loading associated with deformation in AF cells reduces synthetic and degradative activities in extracellular matrix and cell proliferation. We evaluated expression of key matrix molecules and cell proliferation by RT-PCR and immunohistochemistry by inner and outer bovine AF

A synthetic probiotic engineered for colorectal cancer therapy modulates gut microbiota.

BACKGROUND: Successful chemoprevention or chemotherapy is achieved through targeted delivery of prophylactic agents during initial phases of carcinogenesis or therapeutic agents to malignant tumors. Bacteria can be used as anticancer agents, but efforts to utilize attenuated pathogenic bacteria suffer from the risk of toxicity or infection. Lactic acid bacteria are safe to eat and often confer health benefits, making them ideal candidates for live vehicles engineered to deliver anticancer drugs. RESULTS: In this study, we developed an effective bacterial drug delivery system for colorectal ca

[Development of targeted chemoradiotherapy for malignant melanoma by exploitation of metabolic pathway].

Malignant melanoma cell possesses a unique metabolic pathway which consists of conversion of tyrosine, an essential amino acid, to dopa and subsequently to dopaquinone in the presence of tyrosinase to form melanin, the end product of the metabolic pathway. This tyrosinase-mediated melanin biosynthesis occurs within normal melanocyte and its transformed cell, malignant melanoma cell. It is in general highly elevated in malignant melanoma cells compared to normal melanocytes. The objective of our research is to develop targeted therapeutic approach for malignant melanoma by utilizing this unique
