Polygenic risk scores and Parkinson's disease in South Africa advancing ancestry informed disease prediction.

Parkinson's disease (PD) is a complex neurodegenerative disorder with environmental and genetic influences. Using genotyping array data of 661 South African PD cases and 737 controls, we conduct a polygenic risk score (PRS) analysis using PRSice-2. Summary statistics from two PD association studies have been used as base datasets. We split the target dataset into training (70%; n = 979) and validation (30%; n = 419) cohorts. We test various clumping window sizes, linkage disequilibrium thresholds, and p-value thresholds to determine the optimal combination for risk prediction. Additionally, we

The effect of prenatal maternal stress on adolescent internalizing symptoms and the genetic moderation of executive functioning and prefrontal cortex.

BACKGROUND: Prenatal maternal stress is a known risk factor for later emotional problems, yet not all exposed offspring develop psychopathology. Executive functioning and prefrontal cortex structure and activity may buffer these effects, but it remains unclear whether genetic predispositions in these domains moderate associations with adolescent internalizing symptoms. METHODS: We examined 3,111 adolescents (1,881 females, 1,230 males). Prenatal stress was measured via a cumulative index across four domains. Internalizing symptoms were self-reported at ages 14 and 16. Genetic predisposition f

Evaluating the causal effect of mitochondrial dysfunction on Alzheimer's disease and Parkinson's disease using polygenic risk scores and Mendelian randomization.

INTRODUCTION: Mitochondrial DNA copy number (mtDNAcn), a measure of mitochondrial genomes per nucleated cell, has an unclear causal relationship with Alzheimer's disease (AD) and Parkinson's disease (PD). We integrated genetic correlation, polygenic risk scores (PRSs), and Mendelian randomization (MR) to assess whether mtDNAcn influences the risk of AD and PD, and evaluate how study-specific factors in mtDNAcn genome-wide association studies (GWASs) distort these causal estimates. METHODS: Using GWASs of four mtDNAcn measures, AD, AD/dementia, and PD, we evaluated genetic correlations, genera

Shared genetic architecture between Alzheimer's disease and brain morphology.

BACKGROUND: A hallmark of Late-onset Alzheimer's disease (LOAD) is brain tissue atrophy across multiple cortical and subcortical regions. However, studies report no significant genetic correlation between LOAD and brain morphology, indicating that the genetic drivers of neurodegeneration are complex. Resolving this shared genetic architecture is key to uncovering the biological mechanisms linking brain morphology to LOAD. METHODS: We analyzed UK Biobank genotype data (n = 272,513; 57.1 ± 8.0 years; 54.6% female) alongside genome-wide association study summary statistics for LOAD and 89 brain

Genetic predisposition to elevated total immunoglobulin E levels defines a distinct adult-onset-predominant asthma phenotype.

Asthma heterogeneity remains a major barrier in precision medicine. Although elevated total serum immunoglobulin E (IgE) is a hallmark of asthma, even in nonatopic patients, its causal role in asthma pathogenesis is debated. We hypothesized that genetic predisposition to increased IgE defines a distinct asthma endotype. A genome-wide association study of total serum IgE in 1,287 non-asthmatic Japanese adults was used to construct IgE polygenic risk scores (IgE_PRS). Applying IgE_PRS to 745 patients with asthma, we performed cluster analysis incorporating age at onset, total IgE levels, IgE_PRS

Association Between the Genetic Risk for Attention-Deficit/Hyperactivity Disorder and Cognitive Function in Older Age: The MYHAT Population-Based Study.

OBJECTIVES: Attention-deficit/hyperactivity disorder (ADHD) has been associated with increased risk for dementia, including Alzheimer's disease (AD). Polygenic risk scores for ADHD (ADHD-PRS) reflect the genetic liability for the disorder and have been linked to cognitive decline in selected clinical samples. This study examined associations between ADHD-PRS and cognitive function in older adults from a population-based cohort and tested whether these associations were moderated by plasma biomarkers of AD and related disorders (ADRD). METHODS: We analyzed data from 1,468 dementia-free partici

Spontaneous Coronary Artery Dissection and a Family History of Aortic Dissection: A Genetic Association Study.

BACKGROUND: Spontaneous coronary artery dissection (SCAD) is an increasingly recognized cause of acute coronary syndrome or sudden cardiac death, primarily affecting relatively young women (median age, 51 years) without typical cardiovascular risk factors. SCAD has a genetic component, with genome-wide association studies identifying multiple risk loci. Thoracic aortic dissection (type A) shares some genetic overlap with SCAD, suggesting potential common predispositions. METHODS: We performed genetic screening or whole-genome sequencing of 17 patients with SCAD (94% women) with a first- or se

Using Large Genomic Biobanks to Generate Insights into Genetic Kidney Disease.

Chronic kidney disease (CKD) affects approximately 9% of the global population, leading to increased risks of end-stage kidney disease (ESKD), cardiovascular disease (CVD), and mortality. Patients with CKD are a huge burden on health care resources globally. CKD is a complex condition influenced by a combination of genetic, environmental, and traditional risk factors. Family studies have suggested heritability rates for CKD ranging from 30% to 75%, and large genomic biobank studies have proven essential in identifying genes with substantial effects on CKD risk and in capturing cumulative genet

Genome-Wide Dissection of the Neutrophil-to-Lymphocyte Ratio Uncovers Polygenic Determinants Linked to Inflammatory Gastrointestinal Disorder Susceptibility.

Background: The neutrophil-to-lymphocyte ratio (NLR) is a simple biomarker that reflects the balance between innate immune response and adaptive immunity. Currently, the genetic basis and clinical implications of NLR in relation to inflammatory gastrointestinal diseases have not been extensively explored. Methods: We carried out a genome-wide association study (GWAS) on European individuals from the UK Biobank to detect genetic variants related to NLR, followed by post-GWAS analyses including colocalization analysis, transcriptome-wide association studies (TWAS), and LD score regression. Logis

Intrinsic Capacity, Polygenic Risk Score, and Risk of Glaucoma.

PURPOSE: The purpose of this study was to investigate the prospective association between intrinsic capacity (IC) and the risk of incident glaucoma. METHODS: We performed prospective cohort analyses among 397,881 UK Biobank participants. An IC deficit composite score was constructed based on six IC components. Cox proportional hazards models were utilized to estimate the associations of IC deficit scores and individual IC components with the risk of glaucoma. We tested gene-IC interactions using polygenic risk score (PRS) data for glaucoma. RESULTS: During a median follow-up time of 13.8 yea

Low Stability and Specificity of Polygenic Risk Scores for Major Psychiatric Disorders Limit Their Clinical Utility.

BACKGROUND: There has been little examination of the stability and validity of polygenic risk scores (PRSs), i.e., whether individuals identified as high risk for a disorder with one PRS are identified as high risk with another PRS and whether high-risk individuals have the disorder. METHODS: The UK Biobank recruited 502,534 individuals ages 37 to 73 years in the United Kingdom between 2006 and 2010. PRSs were calculated for 408,853 White British individuals. PRS continuous shrinkage (CS), which uses single nucleotide polymorphism effect sizes under CS, was used to calculate 3 different PRSs

Polygenic risk scores of rheumatoid arthritis associated with seropositivity and bone erosions in a Taiwanese population.

Polygenic risk scores (PRS) are widely used to estimate disease risks and predict clinical outcomes. However, ethnic differences exist worldwide. We aimed to use PRS for predicting the development of rheumatoid arthritis (RA) in a Taiwanese population and investigate whether PRS of RA may be associated with structural damage. PRS was calculated via GWAS data from 2,042 RA patients and 7,950 controls from the Taiwan Precision Medicine Initiative. LDpred2, PLINK, and PRSice-2 models were used to evaluate RA susceptibility via AUC. Clinical factors, including rheumatoid factor (RF), anti-citrulli

Polygenic risk scores as a marker for epilepsy risk across lifetime and after unspecified seizure events.

A diagnosis of epilepsy has significant consequences for an individual but is often challenging in clinical practice. Novel biomarkers are thus greatly needed. Here, we investigated how common genetic factors (epilepsy polygenic risk scores, [PRSs]) influence epilepsy risk in detailed longitudinal electronic health records (EHRs) of > 700k Finns and Estonians. We found that a high genetic generalized epilepsy PRS (PRSGGE) increased risk for genetic generalized epilepsy (GGE) (hazard ratio [HR] 1.73 per PRSGGE standard deviation [SD]) across lifetime and within 10 years after an unspecified sei

No evidence for genetic role of the tumor necrosis factor pathway in Parkinson's disease.

Tumor necrosis factor (TNF) inhibition is under investigation as a therapeutic strategy for Parkinson's disease (PD) and REM sleep behavior disorder (RBD), yet supporting genetic evidence is limited. We used Summary-data-based Mendelian Randomization (SMR) to test whether expression level of ten TNF-related genes were causally linked to PD risk, PD progression, or RBD risk. We also analyzed associations between common and rare variants in these genes, and performed pathway specific polygenic risk score analysis, with PD. Overall, our findings do not support a genetic link between the TNF signa

HPA-axis multilocus genetic variation moderates longitudinal link between friendship quality and adolescent depressive symptoms after childhood abuse.

BACKGROUND: Studies indicate that genetic variations associated with the functioning of the hypothalamic-pituitary-adrenal (HPA) axis can influence the relationship between environmental stressors and depressive symptoms, but examining gene-environment interactions with single polymorphisms limits power. OBJECTIVE: This study examined a multilocus genetic profile score (MGPS) for HPA-axis function and investigated its interaction with childhood abuse, friendship quality, and the longitudinal trajectory of depressive symptoms among adolescents. METHOD: A total of 931 adolescents (Mage = 14.21

A systematic review of genome-wide research on psychotic experiences and negative symptom traits: new revelations and implications for psychiatry.

We present a systematic review of genome-wide research on psychotic experience and negative symptom (PENS) traits in the community. We integrate these new findings, most of which have emerged over the last four years, with more established behaviour genetic and epidemiological research. The review includes the first genome-wide association studies of PENS, including a recent meta-analysis, and the first SNP heritability estimates. Sample sizes of <10 000 participants mean that no genome-wide significant variants have yet been replicated. Importantly, however, in the most recent and well-powere
