Living without pain: A 10-year study of congenital insensitivity to pain with anhidrosis.

BACKGROUND: Congenital Insensitivity to Pain with Anhidrosis (CIPA) is a rare hereditary neuropathy caused by NTRK1 gene mutations, predisposing patients to recurrent infections and chronic wounds. Long-term studies on microbial and clinical outcomes in CIPA are limited. This study presents analysis of infection patterns, antibiotic resistance, and clinical outcomes in CIPA patients. METHODS: A comprehensive ten-year retrospective cohort study was conducted at Soroka University Medical Center, Beer Sheva, Israel, from January 2014 to January 2023. Electronic medical records were reviewed to i

Amoebae as training grounds for microbial pathogens.

Grazing of amoebae on microorganisms represents one of the oldest predator-prey dynamic relationships in nature. It represents a genetic "melting pot" for an ancient and continuous multi-directional inter- and intra-kingdom horizontal gene transfer between amoebae and its preys, intracellular microbial residents, endosymbionts, and giant viruses, which has shaped the evolution, selection, and adaptation of microbes that evade degradation by predatory amoeba. Unicellular phagocytic amoebae are thought to be the ancient ancestors of macrophages with highly conserved eukaryotic processes. Selecti

The role of the microbiome in the neurobiology of social behaviour.

Microbes colonise all multicellular life, and the gut microbiome has been shown to influence a range of host physiological and behavioural phenotypes. One of the most intriguing and least understood of these influences lies in the domain of the microbiome's interactions with host social behaviour, with new evidence revealing that the gut microbiome makes important contributions to animal sociality. However, little is known about the biological processes through which the microbiome might influence host social behaviour. Here, we synthesise evidence of the gut microbiome's interactions with var

Distribution and Catalytic Mechanism of a Novel Carboxylesterase Degrading Multiple Aryloxyphenoxypropionate Herbicides in Bacteria and Archaea.

Aryloxyphenoxypropionate (AOPP), a widely used class of herbicides, pose significant risks to ecosystems and human health. In this study, Bacillus amyloliquefaciens L1 was isolated as a novel degrader, completely degrading 97.42 mg/L cyhalofop-butyl within 12 h via ester bond hydrolysis to cyhalofop acid. A new carboxylesterase, PnbA, was identified from strain L1, showing substrate preference for cyhalofop-butyl > clodinafop-propargyl > haloxyfop-p-methyl > quizalofop-p-ethyl. Molecular docking and mutagenesis identified S190 as a key catalytic residue. Two mutants, K411A and T412A, exhibited

Crohn's Disease: From Immune Dysregulation to Intestinal Damage.

Crohn's disease (CD) is a long-lasting, recurring inflammatory bowel disease (IBD) marked by segmental, transmural inflammation that may affect any part of the gastrointestinal tract. Growing evidence suggests that CD does not originate from a single pathogenic mechanism, but instead results from the convergence of various biological disturbances that collectively disrupt intestinal homeostasis. Genetic alterations that weaken microbial detection, autophagy, and epithelial protection appear to establish initial vulnerabilities, enabling microbes to trigger exaggerated immune responses. This vu

The role of exogenous electron donors in steering microbial electrosynthesis of multi-carbon fatty acids from carbon dioxide.

Microbial electrosynthesis (MES) enables CO2 conversion to multi-carbon fatty acids, but selective upgrading to longer-chain products remains limited by inefficient electron supply. This study developed a sequential MES platform using short-chain alcohols (ethanol, propanol, isopropanol) as exogenous electron donors to direct CO2 conversion into C4-C6 fatty acids. Ethanol preferentially promotes the formation of butyrate (C4, 0.42 g/L) and caproate (C6, 0.13 g/L), whereas propanol shifts selectivity towards valerate (C5, 0.51 g/L), while isopropanol exhibits lower chain-elongation efficiency.

Hologenomic analysis of rectal mucus sampling for detection of adenomatous polyps and colorectal cancer.

Colorectal cancer (CRC) is the fourth most common cancer and the third leading cause of cancer-related mortality worldwide, with incidence rising among younger populations. The significant clinical and economic burden highlights the need for minimally invasive technologies capable of detecting pre-malignant and early-stage disease. Although liquid biopsy approaches have advanced, they have not achieved sufficient performance for clinical adoption when compared with colonoscopy, the current diagnostic gold standard. CRC is a mucosal pathology, yet current diagnostic methods have not leveraged m

Using bacterial and mitochondrial DNA markers to assess fecal pollution sources in stream water and sediments of a mixed land-use watershed.

AIMS: Although stream bed sediment can be an important reservoir of fecal bacteria and pathogens, it is rarely analyzed when assessing microbial water quality. This study aims to identify fecal contamination sources in stream water and sediment and evaluates the impact of rainfall events on microbial water quality in a mixed land-use watershed. METHODS AND RESULTS: Quantitative polymerase chain reaction was employed to quantify human-, cattle-, and chicken-associated genetic markers during dry and wet periods in the Middle Tallapoosa watershed in Alabama, USA. Human- and cattle-associated mar

Integrating microbial siderophores into concepts of plant iron nutrition.

Iron is a crucial micronutrient for plants, but its availability in soil is often limited. Iron deficiency compromises plant growth, and low iron content in crops contributes substantially to the 'hidden hunger' that affects human health globally. The elucidation of Strategy I (reduction-based) and Strategy II (phytosiderophore-based) for iron acquisition was a milestone in plant biology and enabled the development of biofortification concepts. However, recent genetic evidence reveals that the boundary between the two strategies is blurred, with many plants possessing elements of both. Here we

Engineered Lactobacillus rhamnosus Producing 3-Hydroxybutyrate: A Dual-Action Therapeutic Strategy for Colon Cancer Cachexia.

3-hydroxybutyrate (3-HB), an essential endogenous metabolite, shows significant therapeutic potential in several disease contexts. However, its clinical application has been hampered by limitations, such as adverse effects on the gut microbiota. This study introduces a genetically engineered strain of Lactobacillus rhamnosus GG (LGGK) that integrates the benefits of 3-HB production with the probiotic properties of LGG. Using a murine colon cancer cachexia (CAC) model, LGGK supplementation significantly improved survival, reduced tumor progression, and alleviated muscle wasting. LGGK restored g

Infectious Triggers and Immune Dynamics in Guillain-Barré Syndrome: Revisiting Campylobacter jejuni and the Silent Role of Haemophilus influenzae.

Guillain-Barré Syndrome (GBS) is a rapidly progressing immune-mediated neuropathy that remains the leading cause of acute flaccid paralysis worldwide. A substantial proportion of GBS cases are precipitated by infectious agents, most notably Campylobacter jejuni and Haemophilus influenzae, which initiate pathogenic autoimmunity via molecular mimicry. This narrative review aimed to synthesize current evidence on the microbial triggers of GBS and elucidate the immune mechanisms linking infection to neuropathic damage. We discuss the evolving landscape of GBS pathogenesis, emphasizing the roles of

Type I interferon remodels lysosome function and modifies intestinal epithelial defense.

Organelle remodeling is critical for cellular homeostasis, but host factors that control organelle function during microbial infection remain largely uncharacterized. Here, a genome-scale CRISPR/Cas9 screen in intestinal epithelial cells with the prototypical intracellular bacterial pathogen Salmonella led us to discover that type I IFN (IFN-I) remodels lysosomes. Even in the absence of infection, IFN-I signaling modified the localization, acidification, protease activity, and proteomic profile of lysosomes. Proteomic and genetic analyses revealed that multiple IFN-I-stimulated genes including

Taxonomical and Functional Assessment of the Endometrial Microbiota in A Context of Recurrent Reproductive Failure: A Case Report.

Investigation of the microbial community in the female reproductive tract has revealed that the replacement of a community dominated by Lactobacillus with pathogenic bacteria may be associated with implantation failure or early spontaneous abortion in patients undergoing assisted reproductive technology (ART) treatment. Herein we describe taxonomically and functionally the endometrial microbiome of an infertile patient with repeated reproductive failures (involving an ectopic pregnancy and two clinical miscarriages). The microbiological follow-up is presented over 18-month in which the microbi

Using meta-omics of contaminated sediments to monitor changes in pathways relevant to climate regulation.

Microbially mediated biogeochemical processes are crucial for climate regulation and may be disrupted by anthropogenic contaminants. To better manage contaminants, we need tools that make real-time causal links between stressors and altered microbial functions, and the potential consequences for ecosystem services such as climate regulation. In a manipulative field experiment, we used metatranscriptomics to investigate the impact of excess organic enrichment and metal contamination on the gene expression of nitrogen and sulfur metabolisms in coastal sediments. Our gene expression data suggest

Physiological and gene expression responses to nitrogen regimes and temperatures in Mastigocladus sp. strain CHP1, a predominant thermotolerant cyanobacterium of hot springs.

Cyanobacteria are widely distributed primary producers with significant implications for the global biogeochemical cycles of carbon and nitrogen. Diazotrophic cyanobacteria of subsection V (Order Stigonematales) are particularly ubiquitous in photoautotrophic microbial mats of hot springs. The Stigonematal cyanobacterium strain CHP1 isolated from the Porcelana hot spring (Chile) was one of the major contributors of the new nitrogen through nitrogen fixation. Further morphological and genetic characterization verified that the strain CHP1 belongs to Stigonematales, and it formed a separate clad

Human nutrition: safety and risk.

Man has always been existing in an "oecotoxic" environment and he will be existing in the future, though the noxious factors are changing. Today the output of modern chemical industry and its effects on health attract abundant interest. The risks in nutrition derive from imbalanced nutrition, food borne diseases of microbial origin, environmental contaminants, toxicants naturally present in food, food additives, and food allergens, the most important risks seem to connected with false nutrition and with microbial hazards. The toxic burden of modern man by chemicals can not be judged as the ove

Genetics of sulfate transport by Salmonella typhimurium.

Sixty-four mutants were isolated from the LT-2 wild-type strain of Salmonella typhimurium by selecting for chromate resistance. The majority of lesions were shown to lie in the cysA gene. (i) The mutants cannot take up sulfate, a finding which verifies the role of cysA in sulfate transport. In addition, 52 sulfate-transport mutants isolated without chromate selection were defective in the cysA gene. (ii) Most had less than 25% of the binding activity of the wild-type strain. (iii) Most had normal sulfite reductase (H(2)S-nicotinamide adenine dinucleotide phosphate oxidoreductase, EC 1.8.1.2) a

Genomic Medicine in Periodontal Disease: Old Issue, New Insights.

Genetic variability is the main cause of phenotypic variation. Some variants may be associated with several diseases and can be used as risk biomarkers, identifying animals with higher susceptibility to develop the pathology. Genomic medicine uses this genetic information for risk calculation, clinical diagnosis and prognosis, allowing the implementation of more effective preventive strategies and/or personalized therapies. Periodontal disease (PD) is the inflammation of the periodontium induced mainly by bacterial plaque and is the leading cause of tooth loss. Microbial factors are responsibl

Research progress of Rumen microbes utilizing lignocellulose for high-value chemical production.

Lignocellulose is the most abundant renewable carbon resource in nature. However, its structural recalcitrance, arising from the complex association of cellulose, hemicellulose, and lignin, results in high pretreatment costs and inefficient enzymatic hydrolysis, thereby limiting its high-value utilization. The rumen harbors a diverse anaerobic microbial community with efficient lignocellulose-degrading capacity, interconnected metabolic networks, and considerable functional adaptability, making the rumen microbiome a promising biological platform for lignocellulose conversion.
