Population genetic consequences of the fragile-X syndrome, based on the X-inactivation imprinting model.

We have examined the population genetic consequences of the model of Laird (Genetics 117:587-599, 1987) in which the fragile-X syndrome is caused by "imprinting" of a mutant chromosome. The imprinting event in this model results from a block to reactivation of an inactive X chromosome prior to oogenesis. If it is assumed that males carrying the imprinted chromosome never reproduce, the frequencies of females and males carrying the imprinted chromosome are expected to be equal. When a mutation-selection balance is established, there are expected to be somewhat more than twice as many females ca

An etiological approach to Balkan endemic nephropathy based on the investigation of two genetically different populations.

The direct inheritance as a possible etiologic factor of Balkan endemic nephropathy (BEN) has been studied in an endemic area in the district of Slavonski Brod (Croatia-Yugoslavia). The basis for the investigation was given by a natural experiment induced by immigration of people from Ukraina to the endemic and nonendemic areas near Slavonski Brod. 1,536 persons in endemic villages and 733 inhabitants from the villages, where BEN has not been found, were studied with the aim of establishing diagnosis or excluding BEN. The generally adopted criteria enabling to classify the examined persons int

Genetic basis of resistance to diazinon in Victorian populations of the Australian sheep blowfly, Lucilia cuprina.

Populations of L. cuprina from the Bairnsdale, Glenrowan, Hamilton and Torrita areas of Victoria were found to have similar high diazinon resistance status, following near fixation of a single allele at the R1 locus. Pure-breeding resistant strains were derived from each area and used as base populations for a selection program on adults. An approximate doubling of the level of resistance to diazinon was achieved in each strain after eight generations of selection. Relaxation of selection over seven generations showed the response had generated a stable plateau in all but the Torrita selected

Association of Genetic Variants of ABCC8 Gene With Type 2 Diabetes Mellitus in the Mongolian Population.

INTRODUCTION: Type 2 diabetes constitutes 90 percent of all diabetes cases, and some polymorphism in the ABCC8 gene, which encodes the ATP-sensitive potassium channel Sur1 protein involved in pancreatic beta cell insulin release, are associated with Type 2 diabetes. Sulfonylurea is used to treat diabetes caused by this metabolic disorder. Recent studies indicate that the rs1799854 and rs1801261 polymorphisms of the ABCC8 gene may be associated with the development of Type 2 diabetes. MATERIALS AND METHODS: This study was designed as a case-control study. Group 89 included individuals with Typ

Putatively Adaptive and Genome-Wide Genetic Variation Often Rank Populations as Similarly Distinctive.

Identifying which populations within species to prioritize to maintain genetic diversity is challenging. Due to its direct connection to current patterns of adaptation and presumed connection to adaptive capacity, some researchers advocate prioritizing adaptive variation. Other researchers advocate prioritizing populations using genome-wide variation, citing the many limitations in identifying adaptive genetic variation, an unknown future, and verbal theory that suggests patterns of easy-to-gather genome-wide genetic variation and adaptive genetic variation may often be congruent. However, evi

Relationship between genetic risk, social jetlag, and obesity traits in a Turkish adult population.

Social jetlag (SJL), a misalignment between circadian rhythms and social schedules, is associated with obesity susceptibility. This cross-sectional study investigated how a metabolic-genetic risk score (GRS), based on Fat Mass and Obesity Associated (FTO), Melanocortin 4 Receptor (MC4R), and Transcription Factor 7-Like 2 (TCF7L2) genes, influences the relationship of SJL and obesity‑related traits in 223 adults (24-50 y) recruited from an outpatient clinic in Ankara, Turkiye. Anthropometric, clinical, and biochemical data were collected. Blood-derived DNA was genotyped using real-time PCR.SJL

Genetic diversity in RHD and RHCE genes among a selected Kenyan blood donor population.

BACKGROUND: Serologic typing for ABO and RhD is standard in transfusion services, with extended serology and genotyping performed to reduce red cell alloimmunization risk. In Kenya, RH typing is limited to RhD, and genotyping is unavailable. This study used RHD/RHCE genotyping to predict phenotypes and their distribution in a Kenyan blood donor population. STUDY DESIGN AND METHODS: A total of 191 donors (114 D-, 74 D+, and 3 weak D) from the Kenya National Blood Transfusion Service were selected. Next-generation sequencing was performed on DNA extracts using a targeted blood group sequencing

Genetic Dissection of Wheat Heterosis Identifies Candidate Genes in Biparental and Diverse Hybrid Populations.

Understanding how numerous heterotic quantitative trait loci (hQTL) shape heterosis and shifting from hQTL to candidate genes are central challenges. To address these challenges, we performed a systematic, genome-wide mapping in a biparental population and a population of diverse hybrids. Leveraging both a triple testcross and immortalized F2 designs, we examined the genetic basis of the heterosis of the wheat hybrid Piko × Hermann. We detected a major hQTL on chromosome 4B, primarily driven by epistasis. This region contains the Green Revolution gene Rht-B1 as the primary candidate gene. Then

Genetic Dissection of Image-Derived Pod-Related Traits in an Interspecific Soybean RIL Population.

Soybean pod-related traits are important agronomic characteristics associated with seed development, domestication, cultivar identification, and breeding improvement. However, conventional phenotyping methods mainly rely on manual measurements, which are time-consuming and labor-intensive and capture only limited dimensions of pod variation, while the genetic basis of skeleton- and curvature-based pod descriptors remains insufficiently characterized in biparental populations. In this study, eight quantitative traits representing pod size, shape, and color components were extracted from an exis

Reconstruction of the genetic history of the Austro-Asiatic- and Dravidian-speaking tribal populations of South Asia.

The ancestors of Austro-Asiatic- and Dravidian-speaking tribal populations are among the earliest settlers of South and Southeast Asia (S&SEA). While Austro-Asiatic speakers are distributed across S&SEA, Dravidian speakers are primarily confined to South India. Previous studies have identified the Indian Austro-Asiatic and Dravidian tribal populations to maximally represent two distinct genetic ancestral components, ancestral Austro-Asiatic (AAA) and ancestral South Indian (ASI), respectively. Leveraging the whole genome sequence (WGS) dataset from GenomeAsia 100K project, we investigated the

Whole-genome resequencing reveals population genetic structure and selective sweeps in Bombus waltoni.

BACKGROUND: Bombus waltoni is a typical alpine bumblebee species and an important indicator species of the Qinghai-Tibetan Plateau. However, whole-genome studies of bumblebees from this region remain limited. In this study, we investigated the genetic diversity, population structure, and adaptive genomic variation of B. waltoni, providing essential genomic resources for understanding its population genetic structure and signatures of local adaptation. RESULTS: Based on whole-genome SNP data, genetic diversity was relatively similar across B. waltoni populations, with expected heterozygosity (

Genetic polymorphisms of SPP1 and MGP are associated with thumb osteoarthritis: a replication study in the Chinese population.

BACKGROUND: In a recent meta-analysis of genome-wide association studies, three novel common variants were reported to be associated with erosive hand osteoarthritis. In this study, we aimed to investigate whether rs17013495, rs11243284 and rs1800801 are associated with the development of thumb OA in the Chinese population. METHODS: All three were genotyped in 529 patients and 822 healthy controls. The gene expression was evaluated in synovium tissue samples collected from 32 patients undergoing trapeziectomy. The Chi-square analysis was performed to compare the frequency of allele frequency b

Germline genetic risk and prostate cancer-specific mortality in a population-based incident cohort.

BACKGROUND: Multiple germline variants are associated with prostate cancer (PCa) susceptibility and aggressive features; however, their value for predicting prostate cancer-specific mortality (PCSM) at the time of diagnosis remains uncertain, particularly among men with clinically localized disease. METHODS: We evaluated associations between germline risk factors and PCSM among 14,644 men with incident PCa in the UK Biobank. Associations between PCSM and reported germline risk factors, including 11 genes recommended by the National Comprehensive Cancer Network (NCCN), nine other candidate gen
