PNH is a disorder of the pluripotent stem cells resulting in a deficient expression of membrane-bound GPI-anchored proteins in different cell types. Several flow cytometric approaches are designed to detect this antigen deficiency. But they all require drawing and testing of normal samples as control. Therefore, in the present study two flow cytometric assays for the detection of CD55 and CD59 deficiency in erythrocytes (REDQUANT CD55/CD59) and granulocytes (CELLQUANT CD55/CD59) are proposed. Precalibrated beads are used to define the cut off between normal and deficient cell populations. The

Paroxysmal nocturnal haemoglobinuria (PNH) terminating in acute leukaemia (AL) is an infrequent condition. In several cases, flow cytometric analysis of glycosylphosphatidylinositol anchored membrane proteins such as DAF and CD59/MACIF has suggested the leukaemic cells to be derived from the PNH clone, thereby implicating PNH as a potential preleukaemic disease. In the present paper, we review the data for one patient treated in our hospital and 20 cases reported in the literature from 1969 to 1993. The sex ratio is 1 female/2 males, mean age at diagnosis of PNH was 46 years and the mean inter

Renal pathology associated with haemoglobinaemia resulting from Babesia canis infection is ascribed to haemoglobinuria, with or without a contribution from anaemic hypoxia. This study was undertaken to investigate the relative roles of haemoglobinaemia and hypoxia in renal function and pathology in the dog. Three groups of 6 dogs each were used over a 4-day period. The dogs in the 1st group were infused with homologous canine haemoglobin, anaemic hypoxia was induced in the 2nd group, and both treatments were applied in the 3rd group. Full urinalyses, serum urea and creatinine concentrations, f

Paroxysmal nocturnal haemoglobinuria (PNH) is an acquired clonal disorder thought to arise in a multipotent haemopoietic stem cell. A distinct clinical feature is a tendency to thrombosis, with a particular predilection for the hepatic veins (Budd-Chiari syndrome). We report here on two patients with PNH who developed hepatic vein thrombosis (HVT) and who were treated with tissue plasminogen activator (t-PA). Both patients had a marked clinical and radiological improvement following the t-PA treatment and remain well over 2 years and 6 years after the treatment. This method of thrombolysis for

Examination of 4379 routine urinalysis specimens with dipsticks sensitive to ascorbic acid showed that 22.8% were positive specimens. The mean urinary vitamin C concentration in this population was 2120 mumol/L. There was a high rate of false-negative dipstick results for hemoglobin in patients with vitamin C in the urine. The highest false-negative rates were observed in urine samples containing less than 50 erythrocytes per high-power field. In further experiments when volunteers consumed supplemental oral USP vitamin C at doses of 100, 250, 500, and 1000 mg or vitamin C-containing fruit jui

A peroxidase-antiperoxidase (PAP) technique was used to diagnose bacillary haemoglobinuria in formalin-fixed, paraffin-embedded liver tissues of cattle. The PAP method revealed Clostridium haemolyticum in the zone of liver necrosis characteristic of the disease and also in culture smears of this microorganism, but C. novyi type B, C. chauvoei, C. septicum and C. perfringens types B and C remained unstained by the PAP reaction. The PAP technique performed provides a specific, simple and rapid method to diagnose bacillary haemoglobinuria.

A total of 17 cases of anomalous hemoglobin M (Hb M) were detected among subjects of varying nationalities in different regions of the USSR. The methods used for identification of Hb M Saskatoon, Hb M Boston, Hb M Iwate, Hb M Hyde Park have been described, among them--electron paramagnetic resonance. Spectral characteristics, electrophoretic mobility of these Hb in pH gradient, reaction with cyanides, thermal stability, in vitro reduction with methemoglobin reductase, isolated from donor's red blood cells, have been investigated. The functional parameters (log P50 and n) have been determined f

A 2-year-old Suffolk gimmer presented with clinical signs of haemoglobinuria, jaundice, anaemia and acute mastitis. Clinical investigation and bacteriological examination of milk samples revealed Clostridium perfringens type A to be the causal agent.

Before a method of investigation is introduced in the daily clinical routine, its reproducibility and correctness must be proved. In the present investigation, the reproducibility of urine stix reactions for glucose, haemoglobin and protein were assessed. The urine stix tests were carried out on freshly voided urine from 30 patients selected at random by three junior nurses with seniorities of one, five and nine years. Complete inter- and intraobserver agreement was found for glukose and haemoglobin whereas the reproducibility for protein was unsatisfactory.

Along with the hereditary forms of pathological hemoglobin, there are a certain number of acquired hemoglobin abnormalities, in particular hemoglobin H and reappearance of foetal hemoglobin. Reactivation of foetal hemoglobin synthesis is seen in a wide variety of physiopathological states and may be accompanied by other changes in erythrocytic properties. On the other hand, hemoglobin H is far more more rare and is always associated with leukemia. Study of family history and the hematologic content generally allow differentiation between an acquired abnormality and hereditary forms of patholog

A case of severe but transient haemolytic disease occurring after a febrile episode is described. The thermal amplitude of the haemolysin was high during the acute phase, since the autoantibody fixed complement at 31 degrees C. After 4 months complement fixation could exclusively be demonstrated at 4 degrees C. The patient was treated in a room heated to 30-32 degrees C. The treatment consisted of prednisone and azathioprine and during the acute phase plasmapheresis was attempted in order to reduce the antibody concentration. However, the haemolysis decreased when the thermal amplitude of the

Two calves were infected with a British leptospire of the Pomona serogroup. Both showed symptoms of anorexia and fever and one had marked haemoglobinurea and leptospiruria. The serum antibody titres rose rapidly in both calves.
