Temperature-responsive PNDJ hydrogels provide high and sustained antimicrobial concentrations in surgical sites.

Local antimicrobial delivery is a promising strategy for improving treatment of deep surgical site infections (SSIs) by eradicating bacteria that remain in the wound or around its margins after surgical debridement. Eradication of biofilm bacteria can require sustained exposure to high antimicrobial concentrations (we estimate 100-1000 μg/mL sustained for 24 h) which are far in excess of what can be provided by systemic administration. We have previously reported the development of temperature-responsive hydrogels based on poly(N-isopropylacrylamide-co-dimethylbutyrolactone acrylate-co-Jeffami…

Mechanical properties of the pectin hydrogels and inflammation response to their subcutaneous implantation.

We studied the influence of the mechanical properties of pectin hydrogels on acute inflammation and tissue repair after subcutaneous implantation. We used hard and soft pectin hydrogels. The results of histology and the analysis of serum-level cytokines demonstrated that the intensity of acute inflammation increased with increasing hardness of the pectin hydrogels. We also showed that the pectin hydrogels did not inhibit tissue repair. The results of the morphometric and texture analysis of the pectin hydrogels showed that the in vivo biodegradation kinetics of hard hydrogels were greater than…

Development of hydrogels, oleogels, and bigels as local drug delivery systems for periodontitis.

Periodontal disease is a chronic inflammation of gum and tissues that surround and support the teeth. Nonsteroidal anti-inflammatory drugs (NSAIDs) can be used in the treatment of periodontitis to ease swelling and inflammation. One approach of treating periodontitis is loading the NSAIDs in local drug delivery systems. Therefore, the objective of this study was to investigate the local delivery of the NSAIDs model drug ibuprofen to treat periodontitis using different types of gel formulations (hydrogel, oleogel, and bigel). Gel formulations were characterized in terms of their rheological pro

Biocompatibility and in vivo degradation of chitosan based hydrogels as potential drug carrier.

Carboxymethyl chitosan-graft-polylactide (CMCS-PLA) and carboxymethyl chitosan (CMCS) hydrogels were prepared by using 1-(3-dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride/N-hydroxysuccinimide (EDC/NHS) as crosslinking agent and catalyst at room temperature. The biocompatibility of the hydrogels was evaluated with the aim of assessing their potential as drug carrier. Various aspects of biocompatibility were considered, including MTT assay, agar diffusion test, release of lactate dehydrogenase (LDH), hemolytic test, plasma recalcification time (PRT), and dynamic clotting time. MTT assay …

Rationally Designed 3D Hydrogels Model Invasive Lung Diseases Enabling High-Content Drug Screening.

Cell behavior is highly dependent upon microenvironment. Thus, to identify drugs targeting metastatic cancer, screens need to be performed i … mplex 2 (TSC2) hypomorphic cells, identifying Cdk2 inhibition as a putative LAM therapeutic. The 3D hydrogels mimic the native niche, enable multiple modes of invasion, and delineate phenotypic differences between healthy and diseased cells, all of which are critical to effective drug screens of highly invasive diseases including lung cancer.

Injectable shear-thinning hydrogels for delivering osteogenic and angiogenic cells and growth factors.

Bone nonunion may occur when the fracture is unstable, or blood supply is impeded. To provide an effective treatment for the healing of nonunion defects, we introduce an injectable osteogenic hydrogel that can deliver cells and vasculogenic growth factors. We used a silicate-based shear-thinning hydrogel (STH) to engineer an injectable scaffold and incorporated polycaprolactone (PCL) nanoparticles that entrap and release vasculogenic growth factors in a controlled manner. By adjusting the solid composition of gelatin and silicate nanoplatelets in the STH, we defined optimal conditions that ena

Responsive Protein Hydrogels Assembled from Spider Silk Carboxyl-Terminal Domain and Resilin Copolymers.

Responsive protein hydrogels are known to respond to target external stimuli that cause changes in their properties, attracting considerable attention for diverse applications. Here we report the design and recombinant biosynthesis of protein copolymers via genetic fusion of repeating units of resilin with spider silk carboxyl-terminal (CT) domain. The resulting copolymers were thermoresponsive in aqueous solutions, and formed reversible hydrogels at low temperatures and irreversible hydrogels at high temperatures within minutes, a peculiar dual thermogelation feature endowed by the CT domain.…

Integrin-specific hydrogels as adaptable tumor organoids for malignant B and T cells.

Non-Hodgkin lymphomas are a heterogeneous group of lymphoproliferative disorders of B and T cell origin that are treated with chemotherapy drugs with variable success rate that has virtually not changed over decades. Although new classes of chemotherapy-free epigenetic and metabolic drugs have emerged, durable responses to these conventional and new therapies are achieved in a fraction of cancer patients, with many individuals experiencing resistance to the drugs. The paucity in our understanding of what regulates the drug resistance phenotype and establishing a predictive indicator is, in gre

Biodegradable DNA-enabled poly(ethylene glycol) hydrogels prepared by copper-free click chemistry.

Significant research has focused on investigating the potential of hydrogels in various applications and, in particular, in medicine. Specifically, hydrogels that are biodegradable lend promise to many therapeutic and biosensing applications. Endonucleases are critical for mechanisms of DNA repair. However, they are also known to be overexpressed in cancer and to be present in wounds with bacterial contamination. In this work, we set out to demonstrate the preparation of DNA-enabled hydrogels that could be degraded by nucleases. Specifically, hydrogels were prepared through the reaction of dib…

Chitosan-based hydrogels to induce neuronal differentiation of rat muscle-derived stem cells.

In this study, we used a chitosan hydrogel as a 3-dimensional substrate for the attachment, proliferation, and differentiation of rat muscle-derived stem cells (rMDSCs) in the presence of valproic acid (VA). Chitosan solutions containing glycerol phosphate disodium salt form a hydrogel at body temperature. The chitosan hydrogel exhibited a porous 3-dimensional network that allowed the culture medium to penetrate. The chitosan hydrogel acted as a suitable biocompatible substrate for the attachment and proliferation of rMDSCs. On chitosan hydrogel in the presence of VA, rMDSCs exhibited higher e

Fish Scale-derived Chitooligosaccharide and Chitosan Hydrogels as Functional Biomaterials for Wound Healing.

Chronic and poorly healing wounds require dressings that do more than provide coverage, but actively regulate infection, oxidative stress, and tissue repair. In this study, hydrogels formulated from fish-scale-derived low-molecular-weight chitosan and chitooligosaccharide (COS) were evaluated using integrated antimicrobial, antioxidant, and in vivo wound-healing analyses to assess their comparative therapeutic performance. Antimicrobial testing against Gram-positive bacteria (Bacillus subtilis and Micrococcus luteus) and Gram-negative bacteria (Aeromonas hydrophila and Klebsiella pneumoniae) s…

Injectable Thermoresponsive Hydrogels for Antibacterial Applications: Design Strategies, Antibacterial Mechanisms, and Biomedical Applications.

Injectable thermoresponsive hydrogels provide flowable precursors that form local depots on warming, offering a route to sustained antibacterial delivery in irregular tissue defects. This narrative review examines how thermal gelation, carrier architecture, and anatomical conditions jointly determine antibacterial exposure. It distinguishes polymer phase separation from macroscopic gelation, injectable precursors from topical and instilled formulations, and bacterial killing from biofilm and host-repair outcomes. Representative studies are compared by composition, delivery, thermal behavior, r…

Oxime hydrogels with ultrafast, pH-independent gelation for robust and adaptive vascular embolization.

Vascular embolization is a key treatment for hemorrhages and tumors, yet conventional agents including coils and microspheres often suffer from incomplete filling, off-target embolism, and heavy dependence on systemic coagulation. While in situ-gelling liquid agents offer conformable filling, achieving robust gelation under high-shear hemodynamics without compromising distal penetration remains a translational hurdle. Here, we report a bioorthogonal oxime-ligation hydrogel engineered for robust and adaptive vascular embolization. This system is composed of aminooxy-functionalized gelatin (Gel-

Immune‑Friendly Skin Interface Remodeling: Chiral Hydrogels for Diabetic Non-Healing Wound Therapy.

Diabetic non‑healing wounds pose a clinical challenge due to persistent inflammation, dysregulated immunity, and poor integration of dressings with the skin. Conventional hydrogel dressings have proven effective in providing physical barriers, moisture retention, and local drug delivery. Further optimization is still feasible for precise immune regulation and dynamic skin interfacial matching. In non-healing wound therapy, enantiomerically defined chiral hydrogels have emerged as a promising platform for targeted immune remodeling. By selectively steering macrophage polarization from pro-infla…
