The FML-vaccine (Leishmune) against canine visceral leishmaniasis: a transmission blocking vaccine.

Transmission blocking vaccines are one of the control strategies for vector-transmitted protozoan diseases. Antibodies raised in the vaccinated host prevent the development of the parasite in the insect vector, interrupting the epidemiological cycle. The FML antigen of Leishmania donovani in combination with saponin (FML-vaccine and Leishmune) induced 92-97% of protections against zoonotic visceral leishmaniasis. We assayed the ability of FML to inhibit Leishmania donovani and Leishmania chagasi procyclic promastigote-binding to dissected Lutzomyia longipalpis midguts. We found a dose-dependen

Community Perceptions of Blackfly Nuisance and Acceptability of the "Slash and Clear" Vector Control Approach in the Ntui Health District of Cameroon: A Qualitative Study.

Besides being vectors of the onchocerciasis parasite, blackflies are a source of nuisance in onchocerciasis-endemic communities. We investigated the experience of residents in the Ntui Health District (Cameroon) regarding blackfly nuisance and assessed their perceptions of a novel "Slash and Clear" (S&C) intervention for blackfly control. Focus group discussions were conducted before and after S&C implementation (respectively, in February 2022 and December 2023). Blackflies were known to emerge from the river areas and cause disease. To prevent blackfly bites, the population often covered thei

Two classic OBPs modulate the responses of female Holotrichia oblita to three major ester host plant volatiles.

Insects possess a fairly sophisticated olfactory system in their antennae to detect odorants essential for their survival and reproduction. Among them, insect first perceives odour sources by odorant-binding proteins (OBPs) to locate host-plants. Methyl salicylate, (Z)-3-hexenyl acetate and dibutyl phthalate are major volatile components of Ulmus pumila and Ricinus communis and elicit strong responses of the scarab beetle Holotrichia oblita adults. However, olfactory perception of the scarab beetle to these odorant compounds is unclear. In the current study, we cloned the OBP6 and OBP7 of H. o

Fruitless alternative splicing and sex behaviour in insects: an ancient and unforgettable love story?

Courtship behaviours are common features of animal species that reproduce sexually. Typically, males are involved in courting females. Insects display an astonishing variety of courtship strategies primarily based on innate stereotyped responses to various external stimuli. In Drosophila melanogaster, male courtship requires proteins encoded by the fruitless (fru) gene that are produced in different sex-specific isoforms via alternative splicing. Drosophila mutant flies with loss-of-function alleles of the fru gene exhibit blocked male courtship behaviour. However, various individual steps in

Polarized light and oviposition site selection in the yellow fever mosquito: no evidence for positive polarotaxis in Aedes aegypti.

Aquatic insects and insects associated with water use horizontally polarized light (i.e., positive polarotaxis) to detect potential aquatic or moist oviposition sites. Mosquitoes lay their eggs onto wet substrata, in water, water-filled tree/rock holes, or man-made small containers/bottles/old tyres containing water. Until now it has remained unknown whether mosquitoes are polarotactic or not. The knowledge how mosquitoes locate water would be important to develop new control measures against them. Thus, we studied in dual-choice laboratory experiments the role of horizontally polarized light

Physiological selectivity and tolerance of social wasps exposed to Malathion.

Insect pest infestations are a worldwide problem as a result of human actions. Among them, Aedes aegypti stands out as vector of several diseases. Like other pests, A. aegypti has become resistant to the insecticide used for its control, thus harming the local fauna of non-target insects. Therefore, the aim of this study was to study the physiological selectivity of social wasps exposed to Malathion, as well as to investigate possible resistance to this insecticide. Adults of the swarming species Polybia platycephala and Protopolybia sedula and the independent species Polistes versicolor were

Molecular Regulators of Entomopathogenic Nematode-Bacterial Symbiosis.

Entomopathogenic nematodes are parasitic organisms with an exceptional capacity to infect rapidly and efficiently a wide range of insect species. Their distinct pathogenic properties have established entomopathogenic nematodes as supreme biocontrol agents of insects as well as excellent models to simulate and dissect the molecular and physiological bases of conserved strategies employed by parasitic nematodes that cause infectious diseases in humans. The extreme infectivity of entomopathogenic nematodes is due in part to the presence of certain species of Gram-negative bacteria that live in mu

Lethal and sub-lethal effects of select macrocyclic lactones insecticides on forager worker honey bees under laboratory experimental conditions.

Selective insecticide application is one important strategy for more precisely targeting harmful insects while avoiding or mitigating collateral damage to beneficial insects like honey bees. Recently, macrocyclic lactone-class insecticides have been introduced into the market place as selective bio-insecticides for controlling many arthropod pests, but how to target this selectivity only to harmful insects has yet to be achieved. In this study, the authors investigated the acute toxicity of fourmacrocyclic lactone insecticides (commercialized as abamectin, emamectin benzoate, spinetoram, and s

Insecticidal effects of Buthus occitanus tunetanus BotIT6 toxin expressed in Escherichia coli and baculovirus/insect cells.

BotIT6 is a neurotoxin polypeptide derived from the venom of the scorpion Buthus occitanus tunetanus (Bot). Its mature form is composed of 62 amino acids. BotIT6 has been reported to be the most potent toxin from Bot venom that has a strict selectivity for insects. Such toxin may have potential as a potent animal-harmless tool against insects. Using RT-PCR, we isolated and sequenced a cDNA encoding 62 amino acid residues corresponding to the known amino acid sequence of BotIT6. We have expressed a recombinant active form of BotIT6 in significantly high amounts in Escherichia coli. We have also

Impact of IRS: Four-years of entomological surveillance of the Indian Visceral Leishmaniases elimination programme.

BACKGROUND: In 2005, Bangladesh, India and Nepal agreed to eliminate visceral leishmaniasis (VL) as a public health problem. The approach to this was through improved case detection and treatment, and controlling transmission by the sand fly vector Phlebotomus argentipes, with indoor residual spraying (IRS) of insecticide. Initially, India applied DDT with stirrup pumps for IRS, however, this did not reduce transmission. After 2015 onwards, the pyrethroid alpha-cypermethrin was applied with compression pumps, and entomological surveillance was initiated in 2016. METHODS: Eight sentinel sites

Onchocerca volvulus transmission in the Mbam valley of Cameroon following 16 years of annual community-directed treatment with ivermectin, and the description of a new cytotype of Simulium squamosum.

BACKGROUND: The onchocerciasis focus surrounding the lower Mbam and Sanaga rivers, where Onchocerca volvulus is transmitted by Simulium damnosum s.l. (Diptera: Simuliidae), was historically the largest in the southern regions of Cameroon. Annual community-directed treatment with ivermectin (CDTI) has been taking place since 2000, but recent studies have shown that new infections are occurring in children. We aimed to investigate blackfly biting and O. volvulus transmission rates along the lower Mbam river 16 years after the formal onset of annual CDTI. METHODS: Black flies were collected for

Synthetic sex-aggregation pheromone of Lutzomyia longipalpis, the South American sand fly vector of Leishmania infantum, attracts males and females over long-distance.

BACKGROUND: In South America the sand fly Lutzomyia longipalpis is the predominant vector of Leishmania infantum, the parasite that causes canine and human visceral leishmaniasis. Co-location of synthetic male sex-aggregation pheromone with an insecticide provided protection against canine seroconversion, parasite infection, reduced tissue parasite loads, and female sand fly densities at households. Optimising the sex-aggregation pheromone + insecticide intervention requires information on the distance over which female and male Lu. longipalpis would be attracted to the synthetic pheromone in

Optimising targets for tsetse control: Taking a fly's-eye-view to improve the colour of synthetic fabrics.

The savannah tsetse flies, Glossina morsitans morsitans and G. pallidipes, are important vectors of Rhodesian human African trypanosomiasis and animal African trypanosomiasis in East and southern Africa. We tested in Zimbabwe whether robust, synthetic fabrics, and innovative fly's-eye-view approaches to optimise fabric colour, can improve insecticide-treated targets employed for tsetse control. Flies were caught by electrocution at a standard target comprising a 1m x 1m black cotton cloth panel with 1m x 0.5m black polyester net panels on each side. Catches were subdivided by species and sex.

Potential of essential oils to prevent fly strike and their effects on the longevity of adult Lucilia sericata.

Lucilia sericata is a facultative ectoparasite causing fly strike or myiasis in warm-blooded vertebrates. It is controlled by traps or insecticides, but both have drawbacks and alternative ways of control are urgently needed. Essential oils (EOs) of vetiver (Chrysopogon zizanioides), cinnamon (Cinnamomum zeylanicum), and lavender (Lavandula angustifolia) and their blends (OBs); OB1 (2 ml of each EO plus 4 ml of sunflower oil as a carrier) and OB2 (2 ml of each EO) were tested. Oils were tested at 5% for deterrence assays, and a dose response assay 0.01-0.6%, was conducted to determine forced-c

Field trials of fatty acids and geraniol applied to cattle for suppression of horn flies, Haematobia irritans (Diptera: Muscidae), with observations on fly defensive behaviors.

Adult horn fly populations were tracked on cattle for 2-week periods before, during and after multiple treatments (every 3-4days) with two repellents in a mineral oil carrier. Cattle were sprayed four times in a two-week period either with 2% geraniol (125ml/cow) or a 15% mixture of short chain fatty acids (C8-C9-C10)(250ml/cow), and there were untreated control cattle. Trials were conducted in California and North Carolina for 3 summers. Short-term fly counts (same day) on treated cattle were reduced by 61-99%, depending on material and trial, and the fatty acid mixture provided better contro

A Molecular Method to Discriminate between Mass-Reared Sterile and Wild Tsetse Flies during Eradication Programmes That Have a Sterile Insect Technique Component.

BACKGROUND: The Government of Senegal has embarked several years ago on a project that aims to eradicate Glossina palpalis gambiensis from the Niayes area. The removal of the animal trypanosomosis would allow the development more efficient livestock production systems. The project was implemented using an area-wide integrated pest management strategy including a sterile insect technique (SIT) component. The released sterile male flies originated from a colony from Burkina Faso. METHODOLOGY/PRINCIPAL FINDINGS: Monitoring the efficacy of the sterile male releases requires the discrimination bet

Long-lasting insecticidal nets fail at household level to reduce abundance of sandfly vector Phlebotomus argentipes in treated houses in Bihar (India).

OBJECTIVE: To determine whether the use of long-lasting insecticidal nets (LLINS) at household level are effective in reducing the abundance of Phlebotomus argentipes, vector of anthroponotic visceral leishmaniasis in India, Nepal and Bangladesh. METHODS: The impact of two long-lasting nets (Olyset and PermaNet) on indoor sandfly abundance was evaluated in selected houses of three endemic hamlets in Bihar (India). It was assumed that most sandflies breed inside the houses and that LLINs would progressively reduce the indoor density during the reproduction season. A campaign of indoor spraying

[Integral insecticide spraying followed by immediate triatomine research, as an additional strategy for the epidemiological surveillance of Chagas' disease, in areas of residual persisting foci].

The locality of Altolândia (Tapiraí Municipality, Minas Gerais, Brazil), frequently presents Panstrongylus megistus infestation in the domiciliary environment. In order to improve epidemiological surveillance in this area, a strategy of domiciliary pyrethroid spraying with high flushing-out effect followed by immediate triatomine research, was implemented in all the existing dwellings. The main objectives were the detection and elimination of cryptic foci, thus reducing the opportunities for new infestations.

[A severe anaphylactic shock caused by spraying the oak processionary caterpillar (Thaumetopoea processionea) in North Brabant].

The processionary caterpillar, Thaumetopoea processionea, caused much inconvenience in the Netherlands in 1996-1997; from the medical point of view, mostly itching and skin rash. After contact with stinging bristles of the caterpillar and with the pesticide Dimilin SC-48, of which diflubenzurone is the active agent, a 72-year-old man had to be resuscitated because of ventricular fibrillation caused by hypotension related to relative underfilling and low systemic vascular resistance. He made a good recovery. Such life-threatening situations can be prevented by publicly announcing plague control
