Plant volatile organic compounds (VOCs) represent one of the most dynamic and integrative biochemical signaling systems linking molecular plant stress responses to ecosystem-level processes. This review provides an integrative cross-scale framework for understanding the biochemical pathways, regulatory networks, ecological functions, and technological applications of stress-induced volatile emissions. At the molecular and cellular levels, VOC emissions are regulated through complex enzymatic and hormonal pathways involving jasmonates, salicylates, ethylene, and abscisic acid, enabling plants t

The tuberous sclerosis protein complex (TSC complex) is a key integrator of metabolic signals and cellular stress. In response to nutrient shortage and stresses, the TSC complex inhibits the mechanistic target of rapamycin complex 1 (mTORC1) at the lysosomes. mTORC1 is also inhibited by stress granules (SGs), RNA-protein assemblies that dissociate mTORC1. The mechanisms of lysosome and SG recruitment of mTORC1 are well studied. In contrast, molecular details on lysosomal recruitment of the TSC complex have emerged only recently. The TSC complex subunit 1 (TSC1) binds lysosomes via phosphatidyl

Microglial dysfunction and aberrant lipid metabolism are emerging as key contributors to Parkinson's disease (PD) pathogenesis. However, the … overexpression ameliorated LD formation, improved phagocytic function, and reduced inflammatory and integrated stress responses in microglia. The PD-related VPS35[D620N] mutation abolished these protective effects. We further found that VPS35 interacts with Rab7 to maintain lysosomal function, and the D620N mutation disrupts this interaction, leading to defective LD clearan …

To maintain genomic integrity, cells have evolved several conserved DNA damage response (DDR) pathways in response to DNA damage and stress conditions. Apurinic/apyrimidinic endonuclease 1 (APE1) exhibits AP endonuclease, 3'-5' exonuclease, 3'-phosphodiesterase, and 3'-exoribonuclease activities and plays critical roles in the DNA repair and redox regulation of transcription. However, it remains unclear whether and how APE1 is involved in DDR pathways. In this perspective, we first updated our knowledge of APE1's functional domains and its nuclease activities and their specific associated subs

The widespread emergence of pyrethroid resistance represents a major challenge in pest and vector management. This study examined the function of the xylC-encoded benzaldehyde dehydrogenase in Pseudomonas aeruginosa under deltamethrin stress. The metabolic response of a ΔxylC mutant cultured with 50 mg/L deltamethrin was compared to the wild-type strain, and functional complementation was achieved by adding purified XylC protein. Integrated metabolomic and proteomic analyses were used to characterize the associated metabolic adaptations. Deltamethrin exposure inhibited the production of 3-(2-c

Mitogen-activated protein kinase (MAPK) pathways are fundamental to the regulation of biological processes in eukaryotic organisms. The basidiomycete Cryptococcus neoformans, known for causing fungal meningitis worldwide, possesses five MAPKs. Among these, Cpk1, Hog1, and Mpk1 have established roles in sexual reproduction, stress responses, and cell wall integrity. However, the roles of Cpk2 and Mpk2 are less understood. Our study elucidates the functional interplay between the Cpk1/Cpk2 and Mpk1/Mpk2 MAPK pathways in C. neoformans. We discovered that CPK2 overexpression compensates for cpk1Δ

The traditional linear no-threshold model of toxicity and stress fails to capture the complex, adaptive nature of biological systems. Hormesis-a biphasic dose-response phenomenon where low-dose stress induces beneficial, adaptive responses while high doses cause harm-offers a more comprehensive paradigm. This article asserts that hormesis is a major, unifying explanatory principle of life that efficiently integrates evolutionary theory, genetics, and epigenetics. From an evolutionary perspective, hormesis is an energy-efficient strategy of adaptive homeostasis, permitting organisms to build bi

Pain normally signals a threat to bodily integrity and causes emotional distress. Acute pain serves a protective function, yet, when pain turns chronic, the protective function is lost. A chain of psychophysiological alterations including changes in the stress regulation system, apparent in dysfunctional activity and responsivity of the hypothalamic-pituitary-adrenal (HPA) axis, might be an important factor in this context. Moreover, maladaptive responses may be complicated by affective comorbid symptoms such as anxiety and depression, and alter nociceptive processing. However, the relationshi

BACKGROUND: The brain-derived neurotrophic factor (BDNF) is a key protein in maintaining neuronal integrity. The BDNF gene is thought to play an important role in the pathophysiology of mood and anxiety disorders. The aim of this study was to investigate, for the first time in a single study, the association between BDNF Val(66)Met polymorphism, anxiety, alcohol consumption, and cortisol stress response.
METHOD: 98 healthy university students (54 females and 44 males), genotyped for the Val(66)Met polymorphism, participated in a physical-stress procedure (cold pressure test, CPT) after having

OBJECTIVE: The observed poorer physical function in persons with mental disorders could partly be due to dysregulation in physiological stress systems. However, an integrated picture of the role of physiological stress systems on objective physical function is lacking. This study examined the association of multiple physiological stress systems with objective physical function, and explored whether these stress systems contribute to the relationship between depression/anxiety and poorer physical function.
METHODS: Data of 2860 persons of the Netherlands Study of Depression and Anxiety was use

Stroke is a clinical syndrome characterized by an acute, focal neurological deficit, primarily caused by the occlusion or rupture of cerebral blood vessels. In stroke, neuroinflammation emerges as a pivotal event contributing to neuronal cell death. The occurrence and progression of neuroinflammation entail intricate processes, prominently featuring mitochondrial dysfunction and adaptive responses. Mitochondria, a double membrane-bound organelle are recognized as the "energy workshop" of the body. Brain is particularly vulnerable to mitochondrial disturbances due to its high energy demands fro

Anthropogenic noise is increasing in intensity and scope, resulting in changes to acoustic landscapes and largely negative effects on a range of species. In birds, noise can mask acoustic signals used in a variety of communication systems, including parent-offspring communication. As a result, nestling birds raised in noise may have challenges soliciting food from parents and avoiding detection by predators. Given that passerine nestlings are confined to a nest and therefore cannot escape these challenges, noise may also act as a chronic stressor during their development. Here, we raised Tree

To survive in challenging environments, plants must rapidly activate immune responses while maintaining developmental plasticity and reproductive success. This requires continuous negotiation of limited energy and metabolic resources between growth, development, and defense. Ubiquitin-mediated proteolysis has emerged as a versatile regulatory mechanism that may integrate immune responses with plant developmental programs. In this review, we summarize accumulating evidence that ubiquitination shapes immune responses at multiple regulatory levels. Many of these immune-regulatory mechanisms depen

This study evaluated both the acute and sublethal effects of pyriproxyfen in Cyprinus carpio using an integrated multibiomarker approach. The 96-h LC50 value was calculated as 4.525 mg/L. Following 14 days of sublethal exposure, pyriproxyfen caused clear, dose-dependent changes across hematological, biochemical, genotoxic, and molecular parameters. Hemoglobin and hematocrit levels declined progressively with increasing concentration, pointing to the development of anemia-like conditions. Biochemical analyses revealed a biphasic response pattern: Several parameters initially increased at lower

DYT-PRKRA (dystonia 16 or DYT-PRKRA) is caused by mutations in the PRKRA gene that encodes PACT, the protein activator of interferon (IFN)-induced double-stranded (ds) RNA-activated protein kinase (PKR). PACT participates in several cellular pathways, of which its role as a PKR activator protein during integrated stress response (ISR) is the best characterized. Previously, we have established that the DYT-PRKRA mutations cause enhanced activation of PKR during ISR to sensitize DYT-PRKRA cells to apoptosis. In this study, we evaluate if the most prevalent substitution mutation reported in DYT-P…

Biofouling is a costly burden for the shipping industry. Metal-based antifouling paints are widely used to protect submerged surfaces, but the release of metals from coatings and the recoating of hulls can leach large amounts of copper and zinc into aquatic environments, posing a risk for aquatic ecosystems and biodiversity. With this study, we studied the time-course metal accumulation and oxidative stress in the digestive gland and the gills of Hyriopsis bialatus, an Asian freshwater mussel, exposed to sublethal concentrations of cuprous chloride (50 and 5 µg/L) and zinc sulfate (1000 and 10

OBJECTIVE: The successful preparation of cells for therapy depends on the characterization of causal factors affecting cell quality. Ultra scale-down methods are used to characterize cells in terms of their response to process engineering causal factors of hydrodynamic shear stress and time. This response is in turn characterized in terms of causal factors relating to variations as may naturally occur during cell preparation, i.e., passage number, generation number, time of the final passage stage and hold time in formulation medium.
METHODS: To investigate the influence of all of these causa

The increasing atmospheric CO2 concentration resulting from anthropogenic emissions has been a major contributor to today's climate change. Microalgae have emerged as an integrated biological route for CO2 capture and utilization; however, the altered metabolic fingerprints of indigenous microalgae to varying CO2 concentrations remain underexplored. This study examines the physiological, biochemical, and metabolic fingerprint of the indigenous microalga Scenedesmus sp. under intermittently sparged varying CO2 concentrations (5%, 10%, and 15%) with the aim of integrating microalgae into the cir
